Eleven spectral vegetation indices that emphasize foliar plant pigments were calculated using airborne hyperspectral imagery and evaluated in 2004 and 2005 for their ability to detect experimental plots of corn manually inoculated with Ostrinia nubilalis (Hübner) neonate larvae. Manual inoculations were timed to simulate infestation of corn, Zea mays L., by first and second flights of adult O. nubilalis. The ability of spectral vegetation indices to detect O. nubilalis-inoculated plots improved as the growing season progressed, with multiple spectral vegetation indices able to identify infested plots in late August and early September. Our findings also indicate that for detecting O. nubilalis-related plant stress in corn, spectral vegetation indices targeting carotenoid and anthocyanin pigments are not as effective as those targeting chlorophyll. Analysis of image data suggests that feeding and stem boring by O. nubilalis larvae may increase the rate of plant senescence causing detectable differences in plant biomass and vigor when compared with control plots. Further, we identified an approximate time frame of 5-6 wk postinoculation, when spectral differences of manually inoculated "second" generation O. nubilalis plots seem to peak. 相似文献

Population genetics strategies offer an alternative and powerful approach for obtaining information about long-distance movement, and have been widely used for examining patterns and magnitude of insect dispersal over geographic and temporal scales. Such strategies are based on the principle that genetic divergence between local populations reflects the interplay between genetic drift and gene flow, and thus can function as an indicator of dispersal capacity. Relatively new approaches for inferring population history are widely applicable for documenting introduction routes of invasive or quarantine species. These approaches are based on genetic variability calculated from changes in gene frequency of subpopulations, measured using molecular genetic markers. Inferences from population genetics can supplement and corroborate conventional observational approaches for characterizing insect dispersal and have provided important clues to many questions raised in the field of behavior and ecology of insects. Here, we summarize our work on the boll weevil as a case study to illustrate the kinds of information on dispersal capacity and dispersal patterns that can be obtained from population genetics techniques that would be difficult or impossible to acquire in other ways. Then we provide examples of how the molecular markers and population genetics tools have been applied to answer immediate questions of relevance to eradication program managers. Though the latter are idiosyncratic to this particular pest, they demonstrate the kinds and range of problems that can be addressed in other systems through application of population genetics strategies. 相似文献

The navel orangeworm, Amyelois transitella (Walker) (Lepidoptera: Pyralidae, Phycitini), is an economically important pest of nut crops in California, USA. Improved management will require better understanding of insect dispersal, particularly relative to when mating occurs. A previous study demonstrated a more robust laboratory flight capacity compared to other orchard moth pests, but it was unclear how mating affects dispersal, and how dispersal affects fecundity. In this study, 1‐ and 2‐day‐old females were allowed to fly overnight on a flight mill either before or after mating, respectively, and were then allowed to oviposit. Data on fecundity were compared between treatments to minimally handled or tethered‐only control females. Females that mated before flight flew longer and covered a greater distance than those flying prior to mating. However, timing of flight relative to mating did not affect fecundity, nor did any measure of flight performance. There was no effect on fecundity when females were forced to fly for designated durations from 3 min to 2 h. Together, our data revealed no obvious trade‐off between flight activity and reproductive output. Distances measured on the flight mills (mean ca. 15 km for mated females) may overestimate net displacement in the field where flight tracks are often meandering. The results suggest that most females mate and oviposit in or near their natal habitat, but that some may disperse potentially long distances to oviposit elsewhere. 相似文献

Abstract: Crop rotation has been a valuable technique for control of Diabrotica virgifera virgifera for almost a century. However, during the last two decades, crop rotation has ceased to be effective in an expanding area of the US corn belt. This failure appears to be due to a change in the insect's oviposition behaviour, which, in all probability, has an underlying genetic basis. A preliminary genome scan using 253 amplified fragment-length polymorphism (AFLP) markers sought to identify genetic variation associated with the circumvention of crop rotation. Samples of D. v. virgifera from east-central Illinois, where crop rotation is ineffective, were compared with samples from Iowa at locations that the behavioural variant has yet to reach. A single AFLP marker showed signs of having been influenced by selection for the circumvention of crop rotation. However, this marker was not diagnostic. The lack of markers strongly associated with the trait may be due to an insufficient density of marker coverage throughout the genome. A weak but significant general heterogeneity was observed between the Illinois and Iowa samples at microsatellite loci and AFLP markers. This has not been detected in previous population genetic studies of D. v. virgifera and may indicate a reduction in gene flow between variant and wild-type beetles. 相似文献

Deregulation of the cell cycle is a hallmark of cancer that enables limitless cell division. To support this malignant phenotype, cells acquire molecular alterations that abrogate or bypass control mechanisms in signaling pathways and cellular checkpoints that normally function to prevent genomic instability and uncontrolled cell proliferation. Consequently, therapeutic targeting of the cell cycle has long been viewed as a promising anti-cancer strategy. Until recently, attempts to target the cell cycle for cancer therapy using selective inhibitors have proven unsuccessful due to intolerable toxicities and a lack of target specificity. However, improvements in our understanding of malignant cell-specific vulnerabilities has revealed a therapeutic window for preferential targeting of the cell cycle in cancer cells, and has led to the development of agents now in the clinic. In this review, we discuss the latest generation of cell cycle targeting anti-cancer agents for breast cancer, including approved CDK4/6 inhibitors, and investigational TTK and PLK4 inhibitors that are currently in clinical trials. In recognition of the emerging population of ER+ breast cancers with acquired resistance to CDK4/6 inhibitors we suggest new therapeutic avenues to treat these patients. We also offer our perspective on the direction of future research to address the problem of drug resistance, and discuss the mechanistic insights required for the successful implementation of these strategies. 相似文献

Genetically modified, insecticidal Bacillus thuringiensis (Bt) corn, Zea mays L., hybrids are used throughout the Corn Belt for European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), control. To slow development of Bt corn resistance, the Environmental Protection Agency requires growers to plant a refuge. Determining the appropriate distance between a refuge and Bt corn, and development of mitigation-remediation strategies such as mass releases of susceptible moths, requires an understanding of adult dispersal and mating behavior. However, much remains unknown about these behaviors. Because mating often occurs in grass near cornfields where adult O. nubilalis aggregate, we planted small-grain plots as aggregation sites in an attempt to retain mass-released adults. The objectives of this study were to examine influences of pheromone lure, plant density, and plant species on distributions of feral and newly emerged, laboratory-reared O. nubilalis among small-grain aggregation plots. Feral adults were collected in aggregation plots in relative abundance, indicating that small-grain plots were acceptable aggregation sites. In contrast, newly emerged adults that were released weekly as dye-marked pupae were rarely found in aggregation plots, with approximately 150-1,500-fold fewer adults captured than expected if all released adults had occupied the plots for > or = 1 d. The majority of newly emerged adults did not colonize the aggregation plots, suggesting that recently eclosed adults leave their natal field and do not colonize the first aggregation sites encountered. Plant species significantly influenced adult distributions among aggregation plots. Mass releases of laboratory-reared pupae in the field may not be a viable remediation tactic because almost all of the newly emerged adults dispersed beyond 300 m of the release point. 相似文献

BAK is a pro-apoptotic BCL-2 family protein that localizes to mitochondria. Here we evaluate the function of BAK in several mouse models of neuronal injury including neuronotropic Sindbis virus infection, Parkinson's disease, ischemia/stroke, and seizure. BAK promotes or inhibits neuronal death depending on the specific death stimulus, neuron subtype, and stage of postnatal development. BAK protects neurons from excitotoxicity and virus infection in the hippocampus. As mice mature, BAK is converted from anti- to pro-death function in virus-infected spinal cord neurons. In addition to regulating cell death, BAK also protects mice from kainate-induced seizures, suggesting a possible role in regulating synaptic activity. BAK can alter neurotransmitter release in a direction consistent with its protective effects on neurons and mice. These findings suggest that BAK inhibits cell death by modifying neuronal excitability. 相似文献

Accumulating data support the view that sepsis is associated with an acquired intrinsic derangement in the ability of cells to consume O(2), a phenomenon that has been termed "cytopathic hypoxia." We sought to use an in vitro "reductionist" model system using cultured cells stimulated with proinflammatory cytokines to test the hypothesis that cytopathic hypoxia is mediated, at least in part, by depletion of intracellular levels of NAD(+)/NADH secondary to activation of the nuclear enzyme poly(ADP-ribose) polymerase (PARP). We measured O(2) consumption by Caco-2 enterocytes growing on microcarrier beads after cells were incubated for 24 h under control conditions or with cytomix, a mixture of tumor necrosis factor-alpha, interleukin-1beta, and interferon-gamma. Immunostimulated cells consumed O(2) at about one-half the rate of control cells, but this effect was largely prevented if any one of the following pharmacological agents was present during the period of incubation with cytomix: 4,5-dihydroxy-1,3-benzene disulfonic acid, a superoxide radical anion scavenger; 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide, a nitric oxide scavenger; 5,10,15,20- tetrakis-[4-sulfonatophenyl]-porphyrinato-iron[III], a peroxynitrite (ONOO(-)) decomposition catalyst; urate, an ONOO(-) scavenger; 3-aminobenzamide, a PARP inhibitor; or N-(6-oxo-5,6-dihydrophenanthridin-2-yl)-N,N-dimethylacetamide HCl, a chemically dissimilar and more potent PARP inhibitor. The decrease in O(2) uptake induced by cytomix was associated with decreased cellular levels of NAD(+)/NADH. The decrease in cellular NAD(+)/NADH content and the decrease in O(2) uptake induced by cytomix were completely abrogated if liposome-encapsulated NAD(+) was added to the cultures during immunostimulation. Empty liposomes also increased O(2) uptake by immunostimulated Caco-2 cells, but much less effectively than liposomes containing NAD(+). These data are consistent with the view that enterocytes exposed to proinflammatory cytokines consume less O(2) due to NAD(+)/NADH depletion secondary to activation of PARP by ONOO(-) or other oxidants. 相似文献
