Certain PKC isoforms are stimulated by insulin and interact with IR as well as with IRS, but it is still not clear if specific PKC isoforms regulate IR signaling directly or through IRS-1. PKCalpha may regulate IRS activity in response to insulin. We investigated the possibility that PKCalpha may be important in insulin signaling. Studies were conducted on skeletal muscle in adult mice and on L6 skeletal cells. PKCalpha is constitutively associated with IRS-1, and insulin stimulation of PKCalpha causes disassociation of the two proteins within 5 min. Blockade of PKCalpha inhibited insulin-induced disassociation of PKCalpha from IRS1. Selective inhibition of PKCalpha increased the ability of insulin to reduce blood glucose levels. Insulin stimulation activates PKB and increases the association of PKCalpha with PKB. Blockade of PKCalpha increased threonine phosphorylation of PKB. We suggest that PKCalpha regulates insulin signaling in skeletal muscle through its disassociation from IRS-1 and association with PKB.  相似文献

Decision making is one of the principal cognitive processes underlying goal-directed behaviour and thus there is justifiably strong interest in modeling it. However, many of these models have yet to be tested outside of the laboratory. At the same time, field work would benefit from the use of experimental methods developed in the laboratory to determine the causal relationships between environmental variables and behaviour. We therefore adapted a laboratory-derived experimental paradigm to test decision making in the wild. The experiment used an indifference-point procedure to determine the influence of both the amount and distance of food on choice behaviour. Free-ranging rhesus monkeys were given the choice between a smaller amount of food at a closer distance and a larger amount farther away. In four conditions, we held the closer amount constant across trials and varied the farther amount to determine the point at which the monkeys were indifferent to the choice alternatives. For example, in condition one, we used one piece of food at the closer location, and determined how many pieces would be equivalent in the farther location. Four different closer amounts were tested to obtain an indifference point curve, with the indifference amounts at the farther location plotted against the closer amounts. The slope of the obtained linear indifference curve was surprisingly high, suggesting that rhesus monkeys significantly discount food that is farther away. Possible reasons for this steep spatial discounting are discussed.  相似文献

Mass spectrometry (MS) coupled with 1-D and 2-D electrophoresis can be utilized to detect and identify immunogenic proteins, but these methods are laborious and time-consuming. We describe an alternative, simple, rapid gel-free strategy to identify multiple immunogenic proteins from Bordetella pertussis (Bp). It couples immunoprecipitation to nano liquid chromatography- tandem mass spectrometry (IP-nLC-MS/MS) and is significantly both time- and labor-saving. We developed a gel-free magnetic bead-based immunoprecipitation (IP) method using different NP-40/PBS concentrations in which solubilized proteins of Bp Tohama I membrane fractions were precipitated with polyclonal rabbit anti-Bp whole cell immune sera. Immune complexes were analyzed by MS and Scaffold analysis (> 95% protein identification probability). Total immunoproteins identified were 50, 63 and 49 for 0.90%, 0.45% and 0.22% NP-40/PBS buffer concentrations respectively. Known Bp proteins identified included pertactin, serotype 2 fimbrial subunit and filamentous hemagglutinin. As proof of concept that this gel-free protein immunoprecipitation method enabled the capture of multiple immunogenic proteins, IP samples were also analyzed by SDS-PAGE and immunoblotting. Bypassing gels and subjecting immunoprecipitated proteins directly to MS is a simple and rapid antigen identification method with relatively high throughput. IP-nLC-MS/MS provides a novel alternative approach for current methods used for the identification of immunogenic proteins.  相似文献

Nucleic acid-based vaccination is a topic of growing interest, especially plasmid DNA (pDNA) encoding immunologically important antigens. After the engineered pDNA is administered to the vaccines, it is transcribed and translated into immunogen proteins that can elicit responses from the immune system. Many ways of delivering DNA vaccines have been investigated; however each delivery route has its own advantages and pitfalls. Skin tattooing is a novel technique that is safe, cost-effective, and convenient. In addition, the punctures inflicted by the needle could also serve as a potent adjuvant. Here, we a) demonstrate the intradermal delivery of plasmid DNA encoding enhanced green fluorescent protein (pCX-EGFP) in a mouse model using a tattooing device and b) confirm the effective expression of EGFP in the skin cells using confocal microscopy.  相似文献

Diminished Na,K-ATPase expression has been reported in several carcinomas and has been linked to tumor progression. However, few studies have determined whether Na,K-ATPase function and expression are altered in lung malignancies. Because cigarette smoke (CS) is a major factor underlying lung carcinogenesis and progression, we investigated whether CS affects Na,K-ATPase activity and expression in lung cell lines. Cells exposed to CS in vitro showed a reduction of Na,K-ATPase activity. We detected the presence of reactive oxygen species (ROS) in cells exposed to CS before Na,K-ATPase inhibition, and neutralization of ROS restored Na,K-ATPase activity. We further determined whether Na,K-ATPase expression correlated with increasing grades of lung adenocarcinoma and survival of patients with smoking history. Immunohistochemical analysis of lung adenocarcinoma tissues revealed reduced Na,K-ATPase expression with increasing tumor grade. Using tissue microarray containing lung adenocarcinomas of patients with known smoking status, we found that high expression of Na,K-ATPase correlated with better survival. For the first time, these data demonstrate that CS is associated with loss of Na,K-ATPase function and expression in lung carcinogenesis, which might contribute to disease progression.  相似文献

Western honey bee (Apis mellifera) colonies in Nova Scotia, Canada were sampled in spring and late summer 2007 to evaluate efficacy of fumagillin dicyclohexylammonium (hereafter, fumagillin) against Nosema ceranae. Colonies treated with fumagillin in September 2006 (n = 94) had significantly lower Nosema intensity in spring 2007 than did colonies that received no treatment (n = 51), but by late summer 2007 no difference existed between groups. Molecular sequencing of 15 infected colonies identified N. ceranae in 93.3% of cases, suggesting that fumagillin is successful at temporarily reducing this recent invasive parasite in western honey bees.  相似文献

Atriplex nummularia is a polyploid Australian saltbush which has been identified as a suitable species for use in the rehabilitation of agricultural land affected by salinity. We isolated 12 polymorphic loci for a preliminary assessment of genetic variability and structure within the species as a basis for a breeding programme. Preliminary screening of loci in 40 individuals from two populations revealed multibanded genotypes consisting of up to seven alleles in a single individual, with up to 29 alleles observed at a single locus. The multibanded patterns are consistent with the polyploid status of this species.  相似文献

Flowers of Pseudowintera traversii (Buchan.) Dandy possess a terminal unicarpellate gynoecium. The present study of carpel morphogenesis was initiated for the purposes of (1) providing additional developmental documentation of the occurrence of terminal carpels in the Winteraceae and (2) comparing the mode of initiation and development of the ascidiate terminal carpel of P. traversii with the essentially conduplicate terminal carpel of Drimys lanceolata. Following its axillary origin, the floral apex of P. traversii initiates 2–3 connate sepals, 5–6 petals, 4–15 stamens, and usually a single terminal carpel, in acropetal succession. Bicarpellate gynoecia may occur with a frequency of up to 15 % on a given plant. The floral apex is zonate and shows increased expression of its zonation during later stages of floral development. The terminal carpel is ascidiate from inception and originates as a cylindrical growth around the entire circumference of the floral apex; transformation of the floral meristem into a carpel primordium terminates apical growth of the floral axis. Carpel growth continues to be cylindrical and is mediated by a ring of marginal and submarginal initials at its summit. Earlier and more extensive division of initials and their derivatives on the dorsal rim causes the primordium to become canted adaxially, shifting the apical cleft to a subterminal adaxial position. Continued marginal meristematic activity results in closure of the cleft as well as elevation and elaboration of the stigmatic crests. Five to seven bitegmic ovules are initiated at the same time as crest elaboration and arise in two rows from the adaxial (laminar) position. Carpel maturation is signified by tannin deposition and oil cell differentiation, beginning at the base and proceeding acropetally; carpel margins bordering the cleft are the last to differentiate. Carpel procambialization is continuous and acropetal from inception, with the dorsal median bundle differentiating before the ventral strands. The significance of occasional bicarpellate flowers is discussed.  相似文献
