Aim

To compare the internal balances of nutrients and the rates of nutrient cycling across nine cocoa agroforestry systems consisting of various combination of soil types (Latosols and Cambisols), production systems (cabruca and Erythrina glauca-shade) and fertilization regimes in southern Bahia, Brazil.

Methods

We measured nutrient stocks in litter fall production, in the accumulated litter and fruits. The internal nutrient balance for various simulations was obtained by the following expressions: (1) Balance 1?=?litter – fruit (seeds and husks) and (2) Balance 2?=?(litter?+?husks) – seeds. Annual litter decomposition coefficients (k) and subsequent potential of nutrient release were also investigated. The data were analyzed by principal components analysis and by Pearson correlations.

Results

There was a high degree of dissimilarity among the cocoa agrosystems in relation to the nutrient cycling and the internal nutrient balance. The mean annual litterfall production ranged from 4.6 to 8.5 Mg/ha, and the amount of accumulated litter ranged from 7.7 to 16.8 Mg/ha. The results showed significant differences in quality among litter from cocoa agroforests; the decomposition coefficient of litter and the subsequent nutrient release were regulated by the litter quality. In general, the cocoa-erythrina system presented a higher capacity to recycle nutrients compared to the cocoa-cabruca system, with the cocoa-erythrina system having the largest transfer rate of nutrients through litterfall, high values for the decomposition coefficient of litter and the lowest values for the Mean Residence Time of nutrients. Cocoa tree leaves functioned as a sink of nutrients, while shade tree leaves functioned predominantly as a source. The nutritional reserves of litter?+?cocoa fruit husks, with respect only to the nutrients exported in the seeds, the balance was positive for all nutrients (N, P, K, Ca and Mg) in all agroforests, which emphasizes the potential productive capacity of these agroforests to sustain the estimated production in different harvest cycles.

Conclusions

The internal balance of nutrients reflects an agroforests’s productive capacity, which accumulated litter and cocoa fruit husks may be important nutrient sources that could enable the development of fertilizer recommendation systems aimed at increasing the efficiency of fertilizer use and at maintaining soil fertility in cocoa agroforests. Therefore, further research is needed to develop nutritional balance systems integrating litter?+?fruits stock and other nutrient pathways (e.g., soil quality, biological N fixation, leaching), which were not measured, for making recommendations regarding liming and fertilizers that are suitable for highly complex biological agrosystems, such as cocoa agroforests that have low levels of elements exported during seed production.  相似文献

SUMMARY 1. In a comparative study, we examined the potential for fish to structure planktonic food webs in shallow mesotrophic to hypereutrophic Northeast Brazilian reservoirs. The food webs were dominated by three guilds of fish (facultative piscivores, generalist planktivores and omnivores), small herbivorous zooplankton and bloom‐forming cyanobacteria, with few littoral macrophytes. 2. A principal component's analysis on data from 13 reservoirs (27 sampling dates in 1995–99) revealed that euphotic depth, the relative density of phytoplankton (i.e. the percentage of overall phytoplankton density) represented by cyanobacteria, and the relative biomass of fish (i.e. percentage of overall biomass) represented by omnivores and facultative piscivores, explained most of the variance in the data. Physico‐chemical conditions, lake morphometry and rainfall were secondary factors. 3. Phytoplankton was related to fish guild structure. Chlorophyll concentration increased with total phosphorus and the relative biomass of omnivorous fish, decreased with the relative biomass of facultative piscivores, but was unrelated to the biomass and mean body size of herbivorous zooplankton. Chlorophyll concentration and the densities of filamentous and colonial cyanobacteria decreased with the ratio of the biomass of facultative piscivores to that of omnivores (FP : OM). 4. We propose two complementary mechanisms for the observed relationships between fish and phytoplankton. At a low biomass of facultative piscivores, juvenile zooplanktivorous fishes may induce a trophic cascade on zooplankton in the littoral zone. Regardless of piscivore biomass, piscivores and omnivores may regulate phytoplankton via multichannel omnivory because of the predominance of omnivorous or detritivorous foraging behaviour. 5. Manipulative experiments are needed to explore further whether, depending on priorities in the use of the reservoir, fisheries management could alter the FP : OM ratio either to enhance fish yields or to reduce phytoplankton densities and cyanobacterial blooms.  相似文献

Although proteins populate large structural ensembles, X-ray diffraction data are traditionally interpreted using a single model. To search for evidence of alternate conformers, we developed a program, Ringer, which systematically samples electron density around the dihedral angles of protein side chains. In a diverse set of 402 structures, Ringer identified weak, nonrandom electron-density features that suggest of the presence of hidden, lowly populated conformations for >18% of uniquely modeled residues. Although these peaks occur at electron-density levels traditionally regarded as noise, statistically significant (P < 10−5) enrichment of peaks at successive rotameric χ angles validates the assignment of these features as unmodeled conformations. Weak electron density corresponding to alternate rotamers also was detected in an accurate electron density map free of model bias. Ringer analysis of the high-resolution structures of free and peptide-bound calmodulin identified shifts in ensembles and connected the alternate conformations to ligand recognition. These results show that the signal in high-resolution electron density maps extends below the traditional 1 σ cutoff, and crystalline proteins are more polymorphic than current crystallographic models. Ringer provides an objective, systematic method to identify previously undiscovered alternate conformations that can mediate protein folding and function.  相似文献

Although chemotherapy plays significant role in the treatment of colorectal cancer, morbidity and mortality due to drug resistance and cancer metastasis are yet to be eliminated. Recently, doxycycline has been reported to have cytotoxic and anti-proliferating properties in various cancer cells. The effects of an ABA 8′-hydroxylase inhibitor, (±)abscinazole-E2B (Abz-E2B), were examined in “Fuji” apple seedlings exposed to sodium chloride (NaCl). Water potential, stomata aperture, and endogenous ABA, proline, and sodium (Na+) levels were analyzed, along with expression of the 9-cis-epoxycarotenoid dioxigenase (MdNCED) and ABA 8′-hydroxylase (MdCYP707A) genes. Water potential and stomatal aperture in the leaves treated with Abz-E2B and NaCl (Abz-E2B+NaCl+ group) were lower than those in the untreated controls, but remained higher than those in the leaves treated with NaCl alone (Abz-E2B?NaCl+ group). Endogenous ABA levels were higher in the Abz-E2B+NaCl+ leaves at 4 days after treatment, whereas those in the Abz-E2B?NaCl+ leaves increased with time and peaked at 12 days after treatment. Expressions of the MdNCED1 and MdNCED2 genes remained lower in Abz-E2B+NaCl+ compared with Abz-E2B?NaCl+ leaves, especially at 12 days after treatment. Proline levels increased sharply at 100-mM NaCl, and Na+ levels increased with time. The Na+ concentrations in the Abz-E2B?NaCl+ leaves increased fourfold compared to those in the Abz-E2B+NaCl+ leaves. The expressions of MdCYP707A1 and MdCYP707A2 were higher in Abz-E2B+NaCl+ at 4 days, but declined at 12 days. The present data suggest that the increased endogenous ABA during the early stages of Fuji apple cultivation under salinity-stress conditions might be a good approach to prevent long-lasting damage and allow plants to recover.  相似文献

Escherichia coli is one of the most used host microorganism for the production of recombinant products, such as heterologous proteins and plasmids. However, genetic, physiological and environmental factors influence the plasmid replication and cloned gene expression in a highly complex way. To control and optimize the recombinant expression system performance, it is very important to understand this complexity. Therefore, the development of rapid, highly sensitive and economic analytical methodologies, which enable the simultaneous characterization of the heterologous product synthesis and physiologic cell behavior under a variety of culture conditions, is highly desirable. For that, the metabolic profile of recombinant E. coli cultures producing the pVAX‐lacZ plasmid model was analyzed by rapid, economic and high‐throughput Fourier Transform Mid‐Infrared (FT‐MIR) spectroscopy. The main goal of the present work is to show as the simultaneous multivariate data analysis by principal component analysis (PCA) and direct spectral analysis could represent a very interesting tool to monitor E. coli culture processes and acquire relevant information according to current quality regulatory guidelines. While PCA allowed capturing the energetic metabolic state of the cell, e.g. by identifying different C‐sources consumption phases, direct FT‐MIR spectral analysis allowed obtaining valuable biochemical and metabolic information along the cell culture, e.g. lipids, RNA, protein synthesis and turnover metabolism. The information achieved by spectral multivariate data and direct spectral analyses complement each other and may contribute to understand the complex interrelationships between the recombinant cell metabolism and the bioprocess environment towards more economic and robust processes design according to Quality by Design framework. © 2016 American Institute of Chemical Engineers Biotechnol. A study was conducted to determine the effects of increasing the levels of dietary carotenoid‐rich microalgae biomass on the skin colouration of angelfish (Pterophyllum scalare). A natural microalgae product characterised by a high content (approximately 5% w/w) of the carotenoid astaxanthin was included in a basal diet at 0.25, 0.50, 1.00 and 2.00 g per 100 g as a substitute for wheat flour. Each diet was fed at 3% of live weight to three replicates of 10 fish each for a period of 28 days. Final weights of replicates were determined as indications of growth. Further, non‐invasive techniques were applied to evaluate skin colour. Whole body images of individual fish were processed using specialised computer software to measure the different components of the RGB, HSV and CIE L*a*b* colour evaluation systems, which were also assessed by three independent judges familiar with the rearing and trade of ornamental fishes. No significant influence of the microalgae addition was detected (P > 0.05) on the final live weight of the fish. The colour components Red, Hue, Value and L* were not linearly related (P < 0.05) to dietary microalgae inclusion levels. Where related (Green, Blue, Saturation, a*, b*), correlation coefficients were 0.66 at most. Second‐order polynomial regression models showed that maximum or minimum values for colour measurements were frequently outside the range (0.00–2.00 g per 100 g) of the microalgae inclusion levels applied. When visual evaluation (the most common assessment approach in practice) was used, the judges clearly confirmed that the colour changes were directly related to increasing levels of microalgae inclusion.  相似文献
