We report the cloning and determination of the nucleotide sequence of the gene encoding nucleoside diphosphate kinase (Ndk) from Pseudomonas aeruginosa. The amino acid sequence of Ndk was highly homologous with other known bacterial and eukaryotic Ndks (39.9 to 58.3% amino acid identity). We have previously reported that P. aeruginosa strains with mutations in the genes algR2 and algR2 algH produce extremely low levels of Ndk and, as a consequence, are defective in their ability to grow in the presence of Tween 20, a detergent that inhibits a kinase which can substitute for Ndk. Hyperexpression of ndk from the clone pGWS95 in trans in the P. aeruginosa algR2an6 algR2 algH double mutant restored Ndk production to levels which equalled or exceeded wild-type levels and enabled these strains to grow in the presence of Tween 20. Hyperexpression of ndk from pGWS95 in the P. aeruginosa algR2 mutant also restored alginate production to levels that were approximately 60% of wild type. Nucleoside diphosphate kinase activity was present in both the cytosolic and membrane-associated fractions of P. aeruginosa. The cytosolic Ndk was non-specific in its transfer activity of the terminal phosphate from ATP to other nucleoside diphosphates. However, the membrane form of Ndk was more active in the transfer of the terminal phosphate from ATP to GDP resulting in the predominant formation of GTP. We have investigated structural transitions in Poly(dG-dC) and Poly(dG-Me5dC) in order to understand the exact role of cations in stabilizing left-handed helical structures in specific sequences andthe
biological role, if any, of these structures. From a novel temperature dependent Z ⇌ B transition it has been shown that a
minor fluctuation in Na+ concentration at ambient temperature can bring about B to Z transition. Forthe first time, wehave observed a novel Z⇌B⇌Zuble
transition in poly(dG-Me5dC) as the Na+ concentration is gradually increased. This suggests that a minor fluctuation in Na+ concentration in conjunction with methylation may transform small stretches of CG sequences from one conformational state
to another. These stretches could probably serve as sites for regulation. Supercoiled formV DNA reconstituted from pBR322
and pβG plasmids have been studied as model systems, in order to understand the nature and role of left-handed helical conformation
in natural sequences. A large portion of DNA in form V, obtained by reannealing the two complementary singlestranded circles
is forced to adopt left-handed double helical structure due to topological constraints (L
k = 0). Binding studies with Z-DNA specific antibody and spectroscopic studies confirm the presence of left-handed Z-structure
in the pβG and pβR322 form V DNA. Cobalt hexamine chloride, which induces Z-form in Poly(dG-dC) stabilizes the Z-conformation
in form V DNA even in the non-alternating purine-pyrimidine sequences. A reverse effect is observed with ethidium bromide.
Interestingly, both topoisomerase I and II (from wheat germ) act effectively on form V DNA to give rise to a species having
an electrophoretic mobility on agarose gel similar to that of open circular (form II) DNA. Whether this molecule is formed
as a result of the left-handed helical segments of form V DNA undergoing a transition to the right-handed B-form during the

A total synthesis of yashabushidiol (1a), a linear diarylheptanoid having 1,3- diol system and its analogues has been achieved by alkynylation of 3-hydroxy-5-phenyl pentanal with substituted phenyl acetylenes. All the compounds have shown significant anti-proliferative activity on human leukemia (THP-1, U-937) and melanoma (A-375) cell lines. Compounds 2a and 2b were found to be most potent with an IC50 of 12.82 μg/mL and 12.62 μg/mL, respectively, on THP-1 leukemia cell line. 相似文献

ABSTRACT: BACKGROUND: Cost-effective production of lignocellulosic biofuels remains a major financial and technical challenge at the industrial scale. A critical tool in biofuels process development is the techno-economic (TE) model, which calculates biofuel production costs using a process model and an economic model. The process model solves mass and energy balances for each unit, and the economic model estimates capital and operating costs from the process model based on economic assumptions. The process model inputs include experimental data on the feedstock composition and intermediate product yields for each unit. These experimental yield data are calculated from primary measurements. Uncertainty in these primary measurements is propagated to the calculated yields, to the process model, and ultimately to the economic model. Thus, outputs of the TE model have a minimum uncertainty associated with the uncertainty in the primary measurements. RESULTS: We calculate the uncertainty in the Minimum Ethanol Selling Price (MESP) estimate for lignocellulosic ethanol production via a biochemical conversion process: dilute sulfuric acid pretreatment of corn stover followed by enzymatic hydrolysis and co-fermentation of the resulting sugars to ethanol. We perform a sensitivity analysis on the TE model and identify the feedstock composition and conversion yields from three unit operations (xylose from pretreatment, glucose from enzymatic hydrolysis, and ethanol from fermentation) as the most important variables. The uncertainty in the pretreatment xylose yield arises from multiple measurements, whereas the glucose and ethanol yields from enzymatic hydrolysis and fermentation, respectively, are dominated by a single measurement: the fraction of insoluble solids (fIS) in the biomass slurries. CONCLUSIONS: We calculate a $0.15/gal uncertainty in MESP from the TE model due to uncertainties in primary measurements. This result sets a lower bound on the error bars of the TE model predictions. This analysis highlights the primary measurements that merit further development to reduce the uncertainty associated with their use in TE models. While we develop and apply this mathematical framework to a specific biorefinery scenario here, this analysis can be readily adapted to other types of biorefining processes and provides a general framework for propagating uncertainty due to analytical measurements through a TE model. 相似文献

A pot experiment was carried out to investigate the effect of iron supplementation on physiological and biochemical status of the low-chill peach cultivars (Saharanpur Prabhat, Shan-e-Punjab and Pratap) suffered from iron chlorosis in artificially created calcareous soil. Three most commonly used iron sources viz. Fe-sulphate (1.0 % and 0.5 %), Fe-citrate (1.0 % and 0.5 %) and FeEDTA (0.1 % and 0.2 %) were sprayed on the 4th and 5th leaves from the apex of the twig. And after 1 week of spraying, observation on various physiological and biochemical parameters in leaves were recorded. Improvement in plant physiological parameters like chlorophyll content index (CCI), photosynthetic rate (Pn), stomatal conductance (gs) and intercellular CO2 conc. (Ci) were recorded best with the application of 1.0 % Fe-sulphate both in treated and untreated upper leaves. The maximum recovery in biochemical parameters such as total leaf chlorophyll content, superoxide dismutase (SOD) and peroxidase (POD) activity was also noted with the application of 1.0 % Fe-sulphate. However, application of 1.0 % Fe-sulphate and 0.5 % Fe-sulphate had similar effect for most of the parameters under study. The ability of iron sources to induce physiological and biochemical responses in iron deficient low-chill peach plants were in the following order Fe-sulphate>Fe-citrate>FeEDTA. Differential responses in plant physiological and biochemical parameters were also exhibited by the low-chill peach cultivars with regard to supplementation of various iron sources. Among the low-chill peach cultivars, Saharanpur Prabhat responded best with the application of iron sources followed by Shan-e-Punjab and Pratap. 相似文献
