To study the population genetics as well as the mating system of captive zebra finch (Taeniopygia guttata) populations, we developed primers for 12 microsatellite loci and screened them in 529 individuals from two successive generations of a single captive population. All markers were polymorphic with five to 14 alleles per locus. We checked all markers for Mendelian inheritance in 307 offspring whose parents were known for sure. Four markers showed evidence for the presence of null alleles. Once allowing for null alleles, we found no mismatches between offspring and parents, suggesting a very low rate of mutation. Average observed and expected heterozygosities across the eight loci showing no evidence for null‐alleles was 0.819 and 0.812, respectively.  相似文献

Anogeissus dhofarica (Combretaceae) is an endemic tree of the monsoon affected coastal mountains of the southern Arabian Peninsula, being the character species of the Hybantho durae–Anogeissetum dhofaricae association, a drought deciduous, monsoon forest community found only in the Dhofar region of southern Oman and the eastern Al‐Mahra region of south‐east Yemen. Due to the steep precipitation gradient from the centre to the edges in this monsoon affected area, A. dhofarica is found in two different habitat types: in continuous woodland belts of the Hawf and Dhofar mountains, and in isolated, scattered woodland patches, as found especially in the Fartak Mts (south‐east Yemen). Fifteen populations (212 individuals) from across the whole distribution area of the species were analysed using amplified fragment length polymorphism fingerprinting to: (1) evaluate the consequences of population fragmentation on the genetic diversity harboured in isolated patches versus cohering stands of the species and (2) to reconstruct the phylogeographical pattern of A. dhofarica as a consequence of oscillations in the monsoon activity during the Pleistocene and Holocene. The analysis of among‐population genetic differentiation and within‐population genetic diversity in A. dhofarica populations resulted in a lack of genetic pauperization and genetic differentiation of populations of the distinctly isolated patches of the Fartak Mts compared to the more luxurious forests of the Hawf and Dhofar regions. This is considered to be due to the high buffer capacity against the loss of genetic diversity caused by the long‐lived life‐form of the species combined with the capability to propagate clonally and the relatively recent fragmentation of Anogeissus forests into the described patches rather than due to high values of gene flow among remnant populations caused by bee pollination and anemochorical and hydrochorical diaspore dispersal. The phylogeographical pattern of the species argues for a quite recent fragmentation of a once continuous forest belt of A. dhofarica that is rather connected with climate changes in the Holocene than triggered by aridity–humidity oscillations reported for the Pleistocene. © 2009 The Linnean Society of London, Biological Journal of the Linnean Society, 2009, 97 , 40–51.  相似文献

Asplenium fontanum subsp. fontanum and A. petrarchae subsp. bivalens are diploid rock ferns of limestone outcrops of the western Mediterranean region. Asplenium fontanum subsp. fontanum occurs from Valencia through northeastern Spain to the Alpes‐Maritimes and Swiss Jura. Asplenium petrarchae subsp. bivalens occurs only on Majorca, in Valencia and possibly in southern Spain. We analysed allozyme and chloroplast genetic marker diversity in 75 populations of A. fontanum subsp. fontanum and 12 populations of A. petrarchae subsp. bivalens sampled from across their respective ranges. The two species show similar levels of species and population genetic diversity to one another and to other diploid European Asplenium taxa. Both are predominantly outbreeding, as indicated by FIS = 0.108 and 0.167 respectively. Substantial between‐population differentiation results largely from differentiation between regions. Isolation by distance operates over limited geographic ranges, up to 50 km. In A. fontanum subsp. fontanum, the major geographical differentiation between Valencia and the rest of the taxon range probably represents an ancient range fragmentation. A less pronounced differentiation divides populations in the SW from those in the NE of the range, with evidence for a biogeographic link between the eastern Pyrenees and southeastern France. High diversity in the Pyrenees may either represent ancient population differentiation, or a suture zone. In A. petrarchae subsp. bivalens, populations on Majorca exhibit a subset of the genetic diversity present in Valencia, although the two regions are strongly differentiated by differing allele frequencies. Dispersal from the mainland may have founded Majorcan populations, although a role for in situ island survival cannot be excluded.  相似文献

1. The causes of distribution patterns of stygobionts (obligate subterranean-dwelling aquatic species) were examined with special emphasis on vicariance and dispersal.
2. Dispersal was investigated on the premise that if migration is important, then migration at small scales should predict patterns at larger scales. Data on the copepod fauna of epikarst in Slovenia were especially useful for the study of migration, because data on habitat occupancy could be collected at scales of individual drips located metres apart to the scale of individual caves to entire karst regions. Occupancy of drips in one cave was a remarkably good predictor of occupancy of caves in a region, although not of the overall range of a given species. These results were also supported by occupancy patterns of the general stygobiotic fauna of West Virginia caves, compared at different scales.
3. Vicariance was investigated by noting that proximity to marine embayments increases the likelihood of vicariant speciation. In the U.S.A., only the fauna of the Edwards Aquifer of Texas has a significant component of marine-derived species. Differences in shape of the relationship between species number and number of caves in a county indicated that the marine-derived component represented an addition to rather than a replacement of the other stygobiotic species.
4. Thus, we found evidence for the importance of both vicariance and dispersal. The techniques employed could be used to study these patterns more generally, as more data become available.  相似文献

The phylogenetic relationships and biogeographical history of the tropical genus Nephrolepis, a popular ornamental fern, are investigated using plastid DNA data from three regions: rbcL, rps4 plus the rps4‐trnS intergenic spacer (IGS) and the trnG intron plus the trnGtrnR IGS. Our taxon sampling includes all but one of the 19 species of the genus. We confirm the monophyly of Nephrolepis, resolve infrageneric relationships and propose monophyly of the widespread species Nephrolepis biserrata and Nephrolepis abrupta, awaiting a denser sampling of Nephrolepis cordifolia and Nephrolepis undulata. The controversial inclusion of Nephrolepis in Lomariopsidaceae is not clearly supported. With a view to identifying synapomorphies for the clades retrieved, we reconstruct the evolution in Nephrolepis of sorus position and indusium shape. Finally, based on an estimation of divergence times and reconstruction of ancestral distributions for the genus, we propose an origin of the crown group in the Eocene in the forests of the Laurasian tropical belt, from where two main lineages would have dispersed and become isolated, one in the Neotropics and the other in Asia–Australasia.
