1. Deoxyribonucleoprotein was prepared from rat thymus and was studied chiefly by the method of electric birefringence. The birefringence depends on the electrical and optical properties of the molecules and on their volume; the decay time of birefringence (after the removal of the electric field) depends on molecular length. 2. A comparison was made of the properties of deoxyribonucleoprotein redissolved after precipitation in 0.15m-sodium chloride with those of the original material, the main object being to find if structural changes have occurred in the former. As a preliminary, the dependence of the solubility of the deoxyribonucleoprotein on the concentration of added salt was investigated, and the birefringence properties were also studied after the addition of sodium chloride at low concentrations, after the alteration of pH and after dialysis. 3. It was found that precipitation of deoxyribonucleoprotein occurs in two fractions, beginning at about 0.4mm-sodium chloride. The solubility is minimal at about 0.15m-sodium chloride. 4. The electric birefringence and decay time of the deoxyribonucleoprotein decrease with increasing concentration of added sodium chloride, and the birefringence also decreases after dialysis. Prolonged dialysis leads to precipitation. 5. The properties of deoxyribonucleoprotein redissolved after precipitation with 0.15m-sodium chloride differ considerably from those of the original deoxyribonucleoprotein. This is attributed to some type of disorganization process occurring during precipitation. It is concluded that the organization of the original structure is very specific. 相似文献

Abstract Three well-defined E. coli operator regions were examined for recurring conformational deviation from a regular B-DNA helix. All three, the lac, trp, and gal, show repeats of the same set of neighboring helical twist angles. These angles recur with a periodicity equal to the helix periodicity on one side of the operator's axes of symmetry. The probability that their occurrence is random was found to be extremely small. Therefore, we propose that in addition to specific bases, repeating twist angle patterns are likely to be among the local parameters involved in repressor-operator recognition. 相似文献

In neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, multiple sclerosis and amyotrophic lateral sclerosis, neuroinflammation can lead to blood‐brain barrier (BBB) breakdown. After intravenous or intra‐arterial injection into mice, endothelial progenitor cells (EPCs) home to the damaged BBB to promote neurovascular repair. Autologous EPCs transfected to express specific therapeutic proteins offer an innovative therapeutic option. Here, we demonstrate that EPC transfection by electroporation with plasmids encoding the reporter protein GFP or an anti‐β‐amyloid antibody fragment (Fab) leads to secretion of each protein. We also demonstrate the secreted anti‐β‐amyloid Fab protein functions in β‐amyloid aggregate solubilization. 相似文献

Extreme heat wave events are now causing ecosystem degradation across marine ecosystems. The consequences of this heat‐induced damage range from the rapid loss of habitat‐forming organisms, through to a reduction in the services that ecosystems support, and ultimately to impacts on human health and society. How we tackle the sudden emergence of ecosystem‐wide degradation has not yet been addressed in the context of marine heat waves. An examination of recent marine heat waves from around Australia points to the potential important role that respite or refuge from environmental extremes can play in enabling organismal survival. However, most ecological interventions are being devised with a target of mid to late‐century implementation, at which time many of the ecosystems, that the interventions are targeted towards, will have already undergone repeated and widespread heat wave induced degradation. Here, our assessment of the merits of proposed ecological interventions, across a spectrum of approaches, to counter marine environmental extremes, reveals a lack preparedness to counter the effects of extreme conditions on marine ecosystems. The ecological influence of these extremes are projected to continue to impact marine ecosystems in the coming years, long before these interventions can be developed. Our assessment reveals that approaches which are technologically ready and likely to be socially acceptable are locally deployable only, whereas those which are scalable—for example to features as large as major reef systems—are not close to being testable, and are unlikely to obtain social licence for deployment. Knowledge of the environmental timescales for survival of extremes, via respite or refuge, inferred from field observations will help test such intervention tools. The growing frequency of extreme events such as marine heat waves increases the urgency to consider mitigation and intervention tools that support organismal and ecosystem survival in the immediate future, while global climate mitigation and/or intervention are formulated. 相似文献

Myriophyllum, among the most species‐rich genera of aquatic angiosperms with ca. 68 species, is an extensively distributed hydrophyte lineage in the cosmopolitan family Haloragaceae. The chloroplast (cp) genome is useful in the study of genetic evolution, phylogenetic analysis, and molecular dating of controversial taxa. Here, we sequenced and assembled the whole chloroplast genome of Myriophyllum spicatum L. and compared it to other species in the order Saxifragales. The complete chloroplast genome sequence of M. spicatum is 158,858 bp long and displays a quadripartite structure with two inverted repeats (IR) separating the large single copy (LSC) region from the small single copy (SSC) region. Based on sequence identification and the phylogenetic analysis, a 4‐kb phylogenetically informative inversion between trnE‐trnC in Myriophyllum was determined, and we have placed this inversion on a lineage specific to Myriophyllum and its close relatives. The divergence time estimation suggested that the trnE‐trnC inversion possibly occurred between the upper Cretaceous (72.54 MYA) and middle Eocene (47.28 MYA) before the divergence of Myriophyllum from its most recent common ancestor. The unique 4‐kb inversion might be caused by an occurrence of nonrandom recombination associated with climate changes around the K‐Pg boundary, making it interesting for future evolutionary investigations. 相似文献
