A bacterial strain, designated Pseudomonas sp. strain DCA1, was isolated from a 1,2-dichloroethane (DCA)-degrading biofilm. Strain DCA1 utilizes DCA as the sole carbon and energy source and does not require additional organic nutrients, such as vitamins, for optimal growth. The affinity of strain DCA1 for DCA is very high, with a Km value below the detection limit of 0.5 microM. Instead of a hydrolytic dehalogenation, as in other DCA utilizers, the first step in DCA degradation in strain DCA1 is an oxidation reaction. Oxygen and NAD(P)H are required for this initial step. Propene was converted to 1,2-epoxypropane by DCA-grown cells and competitively inhibited DCA degradation. We concluded that a monooxygenase is responsible for the first step in DCA degradation in strain DCA1. Oxidation of DCA probably results in the formation of the unstable intermediate 1,2-dichloroethanol, which spontaneously releases chloride, yielding chloroacetaldehyde. The DCA degradation pathway in strain DCA1 proceeds from chloroacetaldehyde via chloroacetic acid and presumably glycolic acid, which is similar to degradation routes observed in other DCA-utilizing bacteria. 相似文献

Experimental infections of the hamster liver were carried out with five strains from patients with clinical amoebiasis and ten strains from asymptomatic carriers. Inocula of comparatively small size (12000-36000 amoebae) were injected under the liver capsule. 1. The virulence of the patient strains varied from 21-96% (see article) and declined sharply within 7-15 weeks after elimination of the associated bacterial flora. The virulence of the carrier strains varied from 0-100%, probably fluctuating with changes in the concomitant bacterial flora (Table 1). 2. The interrelation between size of inoculum, period of bacteria-free growth, and virulence was demonstrated with a Crithidia-associated patient strains (Table 2). 3. A patient strain showed a faster decrease of virulence during axenic than in Crithidia-associated cultivation (Table 3). 4. Two successive passages through hamster liver resulted in a marked increase of virulence of two bacteria-free strains, lasting for several months (Table 4). 5. A significant enhancement of virulence of Crithidia-associated and axenic amoebae by reassociation with a mixed bacterial flora during two weeks, followed by elimination of the bacteria, was demonstrated with two strains. The restored virulence was lost again within a few weeks (Table 5). 6. The virulence of an attenuated patient strain did not become manifest by adding large numbers of dead amoebae to the inoculum (Table 6). 7. The pathology of the different lesions caused in the hamster liver by the amoebae is described, including one of a granulomatous type, frequently found after inoculation with bacteria-free amoebae. 8. In an attempt to explain the occurrence of strains differing in pathogenicity an hypothesis is put forward based on the idea of selection of virulence and avirulent amoebae. 相似文献

Using freeze-fracture electron microscopy, compositional changes were analysed in the surface membrane of Xenopus oocytes during maturation after in vitro progesterone treatment, as well as in eggs before and after fertilization. Investigated stages were as follows: (1) defolliculated full-grown oocytes; (2) defolliculated oocytes after 5 min exposure to 5 micrograms/ml progesterone; (3) ditto at germinal vesicle breakdown (GVBD) after 5 h progesterone treatment; (4) unfertilized eggs at oviposition and (5) zygotes 30 min post-fertilization. Comparing the patterns of intramembranous particle (IMP) density and IMP size during these stages the following changes were found: a transient decrease in IMP density was found after 5 min progesterone treatment; a 48% increase during maturation; a further 17% increase after fertilization. In defolliculated oocytes tight-junction-like structures were found, but no gap junctions. These results are discussed with reference to progesterone action, membrane remodelling, protein synthesis and membrane lipid organization. 相似文献

The cellular contact sites between the full-grown oocyte of Xenopus laevis and the surface extensions of surrounding follicles cells were analysed by electron microscopy of ultrathin sections, freeze-fracture replicas and critical point-dried specimens. Evidence is given for the presence of clusters of intramembranous particles (IMPs) at the P-face which represent gap junctions in diverse forms. Most common are maculae (phi 0.2-0.5 micron) of densely packed IMPs (phi 12 +/- 2 nm) which represent focal gap junctions generally found at the tips of follicle cell surface extensions. Inside many maculae an IMP-free area occurs which appears as a smooth disk (phi 70-80 nm) at both fracture faces. Occasionally a few IMPs are trapped within the smooth disks. Beside the maculae, networks of arrayed IMPs occur that enclose several smooth disks. These latter gap junctions probably are more frequent in side-to-side contacts between surface extensions of the oocyte and the follicle cells. The possible function of these IMP networks is discussed as being related to similar membrane specializations in excitable cells. Summary The morphological and histochemical characteristics of endocrinelike cells of the pulmonary epithelium of the right lower lobe of 12 human adult lungs were studied.Few cells were reactive to the argyrophil silver method of Grimelius and of Sevier and Munger and cells with a similar morphology and distribution emitted a green or yellow fluorescence after treatment of the lung epithelium with the amine precursors L-DOPA or L-HTP, respectively. A greater number of cells seems to be demonstrated by electron microscopy. The cells were characterized by small, round secretory granules showing a central dense core and a very thin clear halo between the core and the surrounding membrane.The cells are thought to be related to the endocrine-like cells of the pulmonary epithelium of the human foetal lung and to cells of carcinoids of larger bronchi. 相似文献

Several strategies have evolved in the vertebrate lineage to facilitate signal transmission in vocal communication. Here, I present a mechanism to facilitate signal transmission in a group of communicating common squirrel monkeys (Saimiri sciureus sciureus). Vocal onsets of a conspecific affect call initiation in all other members of the group in less than 100 ms. The probability of vocal onsets in a range of 100 ms after the beginning of a vocalization of another monkey was significantly decreased compared to the mean probability of call onsets. Additionally, the probability for vocal onsets of conspecifics was significantly increased just a few hundreds of milliseconds after call onset of others. These behavioral data suggest neural mechanisms that suppress vocal output just after the onset of environmental noise, such as vocalizations of conspecifics, and increase the probability of call initiation of group mates shortly after. These findings add new audio–vocal behaviors to the known strategies that modulate signal transmission in vocal communication. The present study will guide future neurobiological studies that explore how the observed audio–vocal behaviors are implemented in the monkey brain. 相似文献
