Six cases of glomus tumor in superficial soft tissues were investigated immunohistochemically for the presence of different types of intermediate filaments, myosin, laminin, a basal lamina glycoprotein, and the endothelial cell markers, factor VIII-related antigen (FVIIIR:Ag) and Ulex europaeus I lectin (UEA I) binding sites. The tumor cells appeared to contain only vimentin, the fibroblast-type of intermediate filament protein. They were also positive for myosin, and were invested by laminin-positive basal lamina-like material, but did not express endothelial cell markers. Ultrastructural studies revealed prominent arrays of both intermediate filaments and microfilaments, the latter resembling the myofilament bundles seen in smooth muscle cells. The results show that glomus tumor cells resemble smooth muscle cells in their content of myosin and in some ultrastructural features. In their lack of desmin, however, they differ from most types of smooth muscle cell, although they are similar in this respect to some vascular smooth muscle cells. 相似文献

To devise effective conservation actions, it is important to know which factors are associated with the population parameters of a declining population. Using mark–recapture methods, we estimated the annual population size, growth rate and survival probability of an ear-tagged flying squirrel population over a 15-year period in a 4,500 ha study area in western Finland. The species is considered vulnerable, but detailed knowledge concerning population sizes or trends is lacking. The population parameters and changes therein were regressed against habitat availability, an indicator of predation pressure, and mean winter temperature (an indicator of climate change), to reveal potential reasons for trends in the population. The best-fit models suggested the annual growth rate to be below one, and on average it was 0.93 (±0.06; SE) across the 15-year period. The survival probability was about 0.22 (±0.03) for juveniles and 0.50 (±0.03) for adults. The population size of adult flying squirrels decreased from 65 (±11) individuals in 1995 to 29 (±6) individuals in 2009. The number of flying squirrels was associated with the amount of available habitat, but the decline in population size was more rapid than the loss of habitat area. If the current decreasing trend in habitat availability continues, the population might become extinct by the year 2020. To halt the population decline, it is necessary to refrain from clear-cutting mature spruce stands until new suitable habitats develop from the maturation of younger forests. 相似文献

Eight cases of primary and metastatic nasopharyngeal lymphoepithelioma and four cases of malignant lymphoma of the pharyngeal region were studied for the presence of keratin by indirect immunofluorescence microscopy. The results showed that all the cases of primary and metastatic lymphoepithelioma contained keratin-positive cells, whereas all the lymphomas were negative for keratin. Anti-keratin antibody thus seems to be a valuable aid in the differential diagnosis between lymphoepithelioma and lymphoma. 相似文献

Protein hydration plays an integral role in determining protein function and stability. We develop a simple method with atomic level precision for predicting the solvent density near the surface of a protein. A set of proximal radial distribution functions are defined and calculated for a series of different atom types in proteins using all-atom, explicit solvent molecular dynamic simulations for three globular proteins. A major improvement in predicting the hydration layer is found when the protein is held immobile during the simulations. The distribution functions are used to develop a model for predicting the hydration layer with sub-1-Ångstrom resolution without the need for additional simulations. The model and the distribution functions for a given protein are tested in their ability to reproduce the hydration layer from the simulations for that protein, as well as those for other proteins and for simulations in which the protein atoms are mobile. Predictions for the density of water in the hydration shells are then compared with high occupancy sites observed in crystal structures. The accuracy of both tests demonstrates that the solvation model provides a basis for quantitatively understanding protein solvation and thereby predicting the hydration layer without additional simulations. 相似文献

Vulnerability of 100 European butterfly species to climate change was assessed using 13 different criteria and data on species
distributions, climate, land cover and topography from 1,608 grid squares 30′ × 60′ in size, and species characteristics increasing
the susceptibility to climate change. Four bioclimatic model-based criteria were developed for each species by comparing the
present-day distribution and climatic suitability of the occupied grid cells with projected distribution and suitability in
the future using the HadCM3-A2 climate scenario for 2051–2080. The proportions of disadvantageous land cover types (bare areas,
water, snow and ice, artificial surfaces) and cultivated and managed land in the occupied grid squares and their surroundings
were measured to indicate the amount of unfavourable land cover and dispersal barriers for butterflies, and topographical
heterogeneity to indicate the availability of potential climatic refugia. Vulnerability was also assessed based on species
dispersal ability, geographical localization and habitat specialization. Northern European species appeared to be amongst
the most vulnerable European butterflies. However, there is much species-to-species variation, and species appear to be threatened
due to different combinations of critical characteristics. Inclusion of additional criteria, such as life-history species
characteristics, topography and land cover to complement the bioclimatic model-based species vulnerability measures can significantly

The presence and localization of lectin receptor sites on rat liver cell nuclear and other endomembranes was studied by light and electron microscopy using fluorescein and ferritin-coupled lectin conjugates. Isolated nuclei labelled with fluorescein-conjugated Concanavalin A (Con A) or wheat germ agglutinin (WGA) often showed membrane staining, which sometimes was especially bright on small stretches of the nuclear surface. Unlabelled nuclei and nuclei with a complete ring fluorescence were also seen. The nuclear fluorescence corresponded in intensity to that seen on the surface of isolated rat liver cells. Con A-ferritin particles were seldom detected on the cytoplasmic surface of the intact nuclear envelope. However, at places where the 2 leaflets of the envelope were widely separated or where the outer nuclear membrane was partly torn away, heavy labelling was seen on the cisternal surface of both the inner and outer nuclear membranes. Labelling with Con A-ferritin was also found on the cisternal side of rough endoplasmic reticulum present in the specimens. No labelling was seen on the cytoplasmic surface of mitochondrial outer membrane. The results demonstrate the presence of binding sites for Con A and WGA in nuclei and an asymmetric localization of these sites on the cisternal side of ribosome-carrying endomembranes in rat liver cells. 相似文献

In the present study, the chain specificity of 16 commonly used monoclonal antibodies to human laminin(s) was analysed by using recombinant laminin beta1 and gamma1 chains. By ELISA, all antibodies reacted with purified placenta laminin, and most antibodies recognised either recombinant beta1 or gamma1 chains. Reactivity and chain specificity was confirmed against the recombinant chains in Western blotting under non-reducing conditions, and only a few antibodies were reactive under reducing conditions. Most antibodies were able to immunoprecipitate associated laminin beta1/gamma1 chains from platelet lysates. Based on these results and data from the literature, a tentative epitope map is presented. 相似文献

Biotic interactions may strongly affect the distribution of individual species and the resulting patterns of species richness. However, the impacts can vary depending on the species or taxa examined, suggesting that the influences of interactions on species distributions and diversity are not always straightforward and can be taxon-contingent. The aim of this study was therefore to examine how the importance of biotic interactions varies within a community. We incorporated three biotic predictors (cover of the dominant vascular species) into two correlative species richness modelling frameworks to predict spatial variation in the number of vascular plants, bryophytes and lichens in arctic–alpine Fennoscandia, in N Europe. In addition, predictions based on single-species distribution models were used to determine the nature of the impact (negative vs. positive outcome) of the three dominant species on individual vascular plant, bryophyte and lichen species. Our results suggest that biotic variables can be as important as abiotic variables, but their relative contributions in explaining the richness of sub-dominant species vary among dominant species, species group and the modelling framework implemented. Similarly, the impacts of biotic interactions on individual species varied among the three species groups and dominant species, with the observed patterns partly reflecting species’ biogeographic range. Our study provides additional support for the importance of biotic interactions in modifying arctic–alpine biodiversity patterns and highlights that the impacts of interactions are not constant across taxa or biotic drivers. The influence of biotic interactions, including the taxon contingency and range-based impacts, should therefore be accounted for when developing biodiversity forecasts. 相似文献
