Chromatophores of the purple photosynthetic bacteria Rhodospirillum rubrum and Rhodobacter (Rhodopseudomonas) sphaeroides were excited by means of 35-ps flashes at 532 nm of varying intensities, both at room temperature and at 4 K. With increasing exciting energy densities the integrated yield of fluorescence produced by these flashes was found to decrease considerably due to singlet-singlet annihilation. An analysis of the results showed that in R. rubrum the number of connected antenna molecules between which energy transfer is possible decreases from about 1000 to about 150 when the temperature is lowered from 298 to 4 K. In Rb. sphaeroides the B875 light-harvesting complex appears to contain about 100 connected bacteriochlorophyll (BChl) 875 molecules at 4 K, while the B800–850 complex contains about 45 BChl 850 molecules. The data are explained by a model for the antenna of Rb. sphaeroides in which units of B875, containing about four reaction centres, are separated by an array of B800–850 units that surrounds B875. By applying a random walk model we found that in both species the rate of energy transfer between neighbouring antenna molecules decreased about 10-fold upon lowering the temperature. The rate of energy transfer from antenna molecules to either open or closed reaction centres decreased only 3- to 4-fold in R. rubrum and remained approximately constant in Rb. sphaeroides upon cooling. A blue shift of the emission spectra at 4 K of both species was observed when the excitation energy density was increased to a level where singlet-singlet annihilation plays a significant role. This observation appears to support the notion that an additional long-wave pigment exists in the antenna of these bacteria. 相似文献

While it is universally recognised that environmental factors can cause phenotypic trait variation via phenotypic plasticity, the extent to which causal processes operate in the reverse direction has received less consideration. In fact individuals are often active agents in determining the environments, and hence the selective regimes, they experience. There are several important mechanisms by which this can occur, including habitat selection and niche construction, that are expected to result in phenotype–environment correlations (i.e. non-random assortment of phenotypes across heterogeneous environments). Here we highlight an additional mechanism – intraspecific competition for preferred environments – that may be widespread, and has implications for phenotypic evolution that are currently underappreciated. Under this mechanism, variation among individuals in traits determining their competitive ability leads to phenotype–environment correlation; more competitive phenotypes are able to acquire better patches. Based on a concise review of the empirical evidence we argue that competition-induced phenotype–environment correlations are likely to be common in natural populations before highlighting the major implications of this for studies of natural selection and microevolution. We focus particularly on two central issues. First, competition-induced phenotype–environment correlation leads to the expectation that positive feedback loops will amplify phenotypic and fitness variation among competing individuals. As a result of being able to acquire a better environment, winners gain more resources and even better phenotypes – at the expense of losers. The distinction between individual quality and environmental quality that is commonly made by researchers in evolutionary ecology thus becomes untenable. Second, if differences among individuals in competitive ability are underpinned by heritable traits, competition results in both genotype–environment correlations and an expectation of indirect genetic effects (IGEs) on resource-dependent life-history traits. Theory tells us that these IGEs will act as (partial) constraints, reducing the amount of genetic variance available to facilitate evolutionary adaptation. Failure to recognise this will lead to systematic overestimation of the adaptive potential of populations. To understand the importance of these issues for ecological and evolutionary processes in natural populations we therefore need to identify and quantify competition-induced phenotype–environment correlations in our study systems. We conclude that both fundamental and applied research will benefit from an improved understanding of when and how social competition causes non-random distribution of phenotypes, and genotypes, across heterogeneous environments. 相似文献

We have measured the singlet-singlet quenching of the bacteriochlorophyll (BChl) fluorescence yield as a function of excitation intensity in a number of antenna complexes isolated from photosynthetic bacteria. Our results show that the lithium dodecyl sulfate (LDS)-B875, LDS-B800 – 850 and lauryldimethylamine N-oxide complexes of Rhodopseudomonas sphaeroides contain 8, greater than 25 and greater than 600 BChl a molecules, respectively. The size of the Rhodopspirillum rubrum B880 complex is greater than 70 BChl a and that of the water-soluble BChl a complex from Prosthecochloris aestuarii about 20–25 BChl a. These results are discussed in relation to current models of the arrangement of antenna complexes within the photosynthetic membranes. 相似文献

Cyanobacterial thylakoid membranes are known to host photosynthetic and respiratory complexes. This hampers a straight forward interpretation of the highly dynamic fluorescence originating from photosynthetic units. The present study focuses on dark-to-light transitions in whole cells of a PSI-deficient mutant of the cyanobacterium Synechocystis sp. PCC 6803. The time-dependent cellular fluorescence spectrum has been measured, while having previously exposed the cells to different conditions that affect respiratory activity. The analysis method used allows the detected signal to be decomposed in a few components that are then assigned to functional emitting species. Additionally, we have worked out a minimal mathematical model consisting of sensible postulated species to interpret the recorded data. We conclude that the following two functional complexes play a major role: a phycobilisome antenna complex coupled to a PSII dimer with either two or no closed reaction centers. Crucially, we present evidence for an additional species capable of strongly quenching fluorescence, whose formation requires the presence of oxygen. 相似文献

This Special Issue of Photosynthesis Research honors Louis M. N. Duysens, Roderick K. Clayton, and George Feher, three pioneering researchers whose work on bacterial photosynthesis laid much of the groundwork for our understanding of the role of the reaction center in photosynthetic light energy conversion. Their key discoveries are briefly summarized and an overview of the special issue is presented. 相似文献
