Data from 338 blood samples of 31 patas monkeys is presented to show the normal hematolgoic development from birth to one year of age, and is compared to 120 samples from normal adults. Marked changes in hematocrit, hemoglobin, red cell numbers, and leukocyte distribution which occurred during the first month were followed by a slow progression to adult values. Body weight data is also presented which shows a linear growth rate during the first year of life.  相似文献

Kinesin superfamily motor proteins contain a structurally conserved loop near the ATP binding site, termed L5. The function of L5 is unknown, although several drug inhibitors of the mitotic kinesin Eg5 bind to L5. We used electron paramagnetic resonance spectroscopy (EPR) to investigate the function of L5 in Eg5. We site-specifically attached EPR probes to ADP, L5, and the neck linker element that docks along the enzymatic head to drive forward motility on microtubules (MTs). Nucleotide-dependent spectral mobility shifts occurred in all of these structural elements, suggesting that they undergo coupled conformational changes. These spectral shifts were altered by deletion of L5 or addition of S-trityl-l-cysteine (STLC), an allosteric inhibitor that binds to L5. In particular, EPR probes attached to the neck linker of MT-bound Eg5 shifted to a more immobilized component in the nucleotide-free state relative to the ADP-bound state, consistent with the neck linker docking upon ADP release. In contrast, after L5 deletion or STLC addition, EPR spectra were highly immobilized in all nucleotide states. We conclude that L5 undergoes a conformational change that enables Eg5 to bind to MTs in a pre-powerstroke state. Deletion or inhibition of L5 with the small-molecule inhibitor STLC blocks this pre-powerstroke state, forcing the Eg5 neck linker to dock regardless of the nucleotide state.  相似文献

High numbers of mononuclear leukocytes (MNL) in one compartment of a diffusion chamber profoundly suppressed lymphocyte responses in the other compartment. Suppression was expressed by decreased incorporation of labeled thymidine, uridine, and leucine and lowered blastogenesis in T-lymphoproliferative cultures as well as reduced Ig synthesis. The inhibitory effect, which was reversible, was experienced predominantly during the later stages of the culture period. Suppression of both autologous and allogeneic effector cells was associated with a radioresistant, moderately adherent non-T-cell subpopulation that did not require activation. Cell contact was not required suggesting that suppression was related to release of a mediator. This mediator was of low molecular weight and was remarkably labile. Only a portion of this modulation could be ascribed to prostaglandins and none to products of the lipoxygenase pathway, hydrogen peroxide, or the accumulation of thymidine.  相似文献

Analyses of mitochondrial DNA and morphological variation were performed on specimens of all five currently recognised Syngnathus pipefish species from the eastern Pacific Ocean with type localities currently considered to lie within the Californian marine biogeographic province: kelp pipefish Syngnathus californiensis, bay pipefish S. leptorhynchus, barred pipefish S. auliscus, barcheek pipefish S. exilis and chocolate pipefish S. euchrous. Results consistently differentiate S. auliscus from the other species and fail to distinguish all other specimens as distinct species, as indicated by extensive morphological overlap as well as incomplete lineage sorting and considerably low genetic divergence for 16s and coI genes(<1%). This study presents a taxonomic revision of eastern Pacific Syngnathus spp. and proposes the synonymy of S. leptorhynchus, S. euchrous and S. exilis, under the senior synonym, S. californiensis. There is still a need to study populations of Syngnathus spp. from north and south of the Californian province to assess whether these too are synonyms of the two-species recognised here.  相似文献

Decreased bone mineral density (BMD) in astronauts returning from long-duration spaceflight missions has been well documented, but the altered mechanical loading environment experienced by the musculoskeletal system, which may contribute to these changes, has not been well characterized. The current study describes the loading environment of the lower extremity (LE) during typical days on the International Space Station (ISS) compared to similar data for the same individuals living on Earth. Data from in-shoe force measurements are also used as input to the enhanced daily load stimulus (EDLS) model to determine the mechanical “dose” experienced by the musculoskeletal system and to associate this dose with changes in BMD.Four male astronauts on approximately 6-month missions to the ISS participated in this study. In-shoe forces were recorded using capacitance-based insoles during entire typical working days both on Earth and on-orbit. BMD estimates from the hip and spine regions were obtained from dual energy X-ray absorptiometry (DXA) pre- and post-flight.Measurable loading was recorded for only 30% of the time assigned for exercise. In-shoe forces during treadmill walking and running on the ISS were reduced by 25% and 46%, respectively, compared to similar activities on Earth. Mean on-orbit LE loads varied from 0.20 to 1.3 body weight (BW) during resistance exercise and were ~0.10 BW during bicycle ergometry. Application of the EDLS model showed a mean decrease of 25% in the daily load experienced by the LE. BMD decreased by 0.71% and 0.83% per month during their missions in the femoral neck and lumbar spine, respectively.Our findings support the conclusion that the measured ISS exercise durations and/or loading were insufficient to provide the loading stimulus required to prevent bone loss. Future trials with EDLS values closer to 100% of Earth values will offer a true test of exercise as a countermeasure to on-orbit bone loss.  相似文献

Members of the serine family of site-specific DNA recombinases use an unusual constellation of amino acids to catalyze the formation and resolution of a covalent protein-DNA intermediate. A recent high resolution structure of the catalytic domain of Sin, a particularly well characterized family member, provided a detailed view of the catalytic site. To determine how the enzyme might protonate and stabilize the 3′O leaving group in the strand cleavage reaction, we examined how replacing this oxygen with a sulfur affected the cleavage rate by WT and mutant enzymes. To facilitate direct comparison of the cleavage rates, key experiments used suicide substrates that prevented religation after cleavage. The catalytic defect associated with mutation of one of six highly conserved arginine residues, Arg-69 in Sin, was partially rescued by a 3′ phosphorothiolate substrate. We conclude that Arg-69 has an important role in stabilizing the 3′O leaving group and is the prime candidate for the general acid that protonates the 3′O, in good agreement with the position it occupies in the high resolution structure of the active site of Sin.  相似文献
