Investigations on the effects of the 5-HT agonists and antagonists on the phase of the circadian locomotor activity rhythm of animals kept in constant light conditions (LL) are rare. Therefore the influence of R-(+)-OH-DPAT (5-HT1A receptors agonist) and metergoline (5-HT1/2/7 receptors antagonist) on the phase shift of the locomotor-activity rhythm alone and when combined with dark pulses in mice kept in LL are examined. The results indicate that 8-OH-DPAT administered independently at 12.00CT (Circadian Time) shifted the phase of the circadian rhythm and reinforced the effect of dark pulses on this parameter. 12.00CT was defined arbitrarily as the onset of locomotor activity in constant conditions. Metergoline diminished the phase shifts after dark pulses compared to 8-OH-DPAT. The influence of the serotonin agonist showed that serotonin can reinforce the phase shifting effect of the locomotor activity rhythm after dark pulses in LL condition. 相似文献

RasGRF1 is a neuron-specific guanine nucleotide exchange factor for the small GTPases Ras and Rac. It is implicated in the regulation of memory formation and in the development of tolerance to drug abuse, although the mechanisms have been elucidated only in part. Here we report the isolation, by the yeast two-hybrid screen, of the microtubule-destabilizing factor SCLIP (SCG10-like protein) as a novel RasGRF1-interacting protein. This interaction requires the region spanning the Dbl-homology domain of RasGRF1, endowed with catalytic activity on Rac. In search for a possible function we found by biochemical means that SCLIP influences the signaling properties of RasGRF1, greatly reducing its ability to activate the Rac/p38 MAPK pathway, while the Ras/Erk one remains unaffected. Moreover, a potential role is suggested by transfection studies in neuronal PC12 cells in which RasGRF1 induces neurite outgrowth, and coexpression of SCLIP counteracts this effect, causing a dramatic decrease in the percentage of cells bearing neurites, which also appear significantly shortened. This study unveils a physical and functional interaction between RasGRF1 and SCLIP. We suggest that this novel interplay may have possible implications in mechanisms that regulate neuronal morphology and structural plasticity. 相似文献

Non-invasive imaging of transgene expression requires the appropriate combination of a reporter gene and a reporter probe. [18F]FEAU positron emission tomography (PET) is used for the assessment of herpes simplex virus type-1 thymidine kinase gene expression. Hybrid AAV phage (termed AAVP) can be adapted to transduce mammalian cells by targeting to a specific receptor. We evaluated a targeted AAVP vector using [18F]FEAU PET. This protocol describes [18F]FEAU production and dosing, micro-PET imaging and image analysis. 2-Deoxy-2-trifluoromethanesulfonyl-1,3,5-tri-O-benzoyl-alpha-D-ribofuranose is radio-fluorinated, converted into its 1-bromo derivative and coupled with protected 5-ethyl uracil. The coupled product is hydrolyzed and purified using HPLC. Tumor-bearing animals targeted with either retroviral or AAVP vectors are anesthetized and injected with [18F]FEAU (0.1 mCi per mouse); this is followed 2 h after injection by imaging on a micro-PET. Production of [18F]FEAU requires approximately 3.5 h from the end of bombardment. PET imaging studies require 2-3 h (depending on the number of animals) after synthesis of [18F]FEAU. 相似文献

The aim of this study was to assess the effect of carvacrol supplement as a dietary additive to rumen fermentors, fed a barley seed:alfalfa hay (70:30) ration and to compare its effect with monensin supplementation. The material was incubated with goat ruminal fluid and four different treatments were included: no additive (C), 7.5 mg/l monensin (M), 250 mg/l carvacrol (C250) and 500 mg/l carvacrol (C500). The addition of carvacrol reduced in vitro dry matter (DM), crude protein (CP) and neutral-detergent fibre (NDF) digestion. The effects induced by C250 on DM digestion at 72 h of incubation were comparable with those of M, whereas a greater reduction was obtained when carvacrol was supplemented at 500 mg/l concentration (68.9, 68.5 and 53.0 v. 76.1% for M, C250 and C500 v. C, respectively). The reduced CP potential degradability by supplements (51.2, 53.9 and 51.5 v. 72.8% for M, C250 and C500 v. C, respectively) was mainly caused by a reduction of the slowly degradable fraction. Volatile fatty acid (VFA) profiles determined after 48 h of incubation showed C250 increased butyrate and decreased acetate proportions, whereas M mainly stimulated propionate proportions, suggesting that the mechanism of action of carvacrol and M differs. C500 significantly reduced total VFA production. Carvacrol could be of great interest for its usage as a potential modulator of ruminal fermentation. Future research, including in vivo studies, in order to understand the factors that contribute to its antimicrobial activity and the selection of the optimal dose is required. 相似文献

Genotyping of HBV is generally used for determining the epidemiological relationship between various virus strains and origin of infection mostly in research studies. The utility of genotyping for clinical applications is only beginning to gain importance. A total of 295 patients with HBsAg positive were selected from the Pakistan Medical Research Council's (PMRC) out patient clinics. Two hundred and twenty six (77%) were males, sixty nine (23%) were females (M to F ratio 3.3:1).Results
Out of 295 patients, 156 (53.2%) had Acute(CAH), 71 (24.2%) were HBV Carriers, 54 (18.4%) had Chronic liver disease (CLD) Hepatitis. 14 (4.7%) were Cirrhosis and HCC patients. Genotype D was the most prevalent genotype in all categories of HBV patients, Acute (108), Chronic (39), and Carrier (53). Cirrhosis/HCC (7) were HBV/D positive. Genotype A was the second most prevalent with 28 (13%) in acute cases, 12 (22.2%) in chronics, 14 (19.7%) in carriers and 5 (41.7) in Cirrhosis/HCC patients. Mixed genotype (A/D) was found in 20 (12.8%) of Acute patients, 3 (5.6%) of Chronic and 4 (5.6%) of carriers, none in case of severe liver conditions.Conclusion
Mixed HBV genotypes A, D and A/D combination were present in all categories of patients except that no A/D combination was detected in severe conditions. Genotype D was the dominant genotype. Antarctic subglacial lakes have, over the past few years, been hypothesised to house unique forms of life and hold detailed
sedimentary records of past climate change. Testing this hypothesis requires in situ examinations. The direct measurement
of subglacial lakes has been considered ever since the largest and best-known lake, named Lake Vostok, was identified as having
a deep water-column. The Subglacial Antarctic Lake Environments (SALE) programme, set up by the Scientific Committee on Antarctic
Research (SCAR) to oversee subglacial lakes research, state that prior exploration of smaller lakes would be a “prudent way
forward”. Over 145 subglacial lakes are known to exist in Antarctica, but one lake in West Antarctica, officially named Ellsworth
Subglacial Lake (referred to hereafter as Lake Ellsworth), stands out as a candidate for early exploration. A consortium of
over 20 scientists from seven countries and 14 institutions has been assembled to plan the exploration of Lake Ellsworth.
An eight-year programme is envisaged: 3 years for a geophysical survey, 2 years for equipment development and testing, 1 year
for field planning and operation, and 2 years for sample analysis and data interpretation. The science experiment is simple
in concept but complex in execution. Lake Ellsworth will be accessed using hot water drilling. Once lake access is achieved,
a probe will be lowered down the borehole and into the lake. The probe will contain a series of instruments to measure biological,
chemical and physical characteristics of the lake water and sediments, and will utilise a tether to the ice surface through
which power, communication and data will be transmitted. The probe will pass through the water column to the lake floor. The
probe will then be pulled up and out of the lake, measuring its environment continually as this is done. Once at the ice surface,
any water samples collected will be taken from the probe for laboratory analysis (to take place over subsequent years). The
duration of the science mission, from deployment of the probe to its retrieval, is likely to take between 24 and 36 h. Measurements
to be taken by the probe will provide data about the following: depth, pressure, conductivity and temperature; pH levels;
biomolecules (using life marker chips); anions (using a chemical analyzer); visualisation of the environment (using cameras
and light sources); dissolved gases (using chromatography); and morphology of the lake floor and sediment structures (using
sonar). After the probe has been retrieved, a sediment corer may be dropped into the lake to recover material from the lake
floor. Finally, if time permits, a thermistor string may be left in the lake water to take time-dependent measurements of
the lake’s water column over subsequent years. Given that the comprehensive geophysical survey of the lake will take place
in two seasons during 2007–2009, a two-year instrument and logistic development phase from 2008 (after the lake’s bathymetry
has been assessed) makes it possible that the exploration of Lake Ellsworth could take place at the beginning of the next
