RNA interference (RNAi) is an RNA degradation process that involves short, double-stranded RNAs (dsRNA) as sequence specificity factors. The natural function of the RNAi machinery is to generate endogenous short double-stranded RNAs to regulate gene expression. It has been shown that treatment of Plasmodium falciparum, the etiologic agent of malaria, with dsRNA induces degradation of the corresponding microRNA (miRNA), yet typical RNAi-associated genes have not been identifiable in the parasite genome. To clarify this discrepancy we set out to clone short RNAs from P. falciparum-infected red blood cells and from purified parasites. We did not find any short RNA that was not a rRNA or tRNA fragment. Indeed, only known human miRNAs were isolated in parasite preparations indicating that very few if any short RNAs exist in P. falciparum. This suggests a different mechanism than classical RNAi in observations of dsRNA-mediated degradation. Of the human miRNAs identified, the human miRNA mir-451 accumulates at a very high level in both infected and healthy red blood cells. Interestingly, mir-451 was not detectable in a series of immortalised cell lines representing progenitor stages of all major blood lineages, suggesting that mir-451 may play a role in the differentiation of erythroid cells. 相似文献

Studies of the influence of fish aquaculture on benthic freshwater nematode assemblages are scarce, but could provide a way of gauging environmental effects. The abundance and diversity of nematode assemblages in response to Oreochromis niloticus aquaculture were investigated in Kafr El-Sheikh Governorate, Egypt, from July to November 2014 under conditions of irrigation (reference), fish farm pond with high Tilapia density, and fish farm pond effluent canal without fish. The nematode genera Adoncholaimus, Punctodora, Labronema, Oncholaimus and Odontolaimus were present at all sites. Environmental factors were not related to nematode distribution patterns. Tilapia predation and/or disturbance may explain reduced nematode abundance, especially of the largest genera, Adoncholaimus, Punctodora and Labronema at the fish farm site. The absence of fish from the drainage site allowed intergeneric nematode competitive exclusion, benefitting the largest nematodes and reducing diversity indices. 相似文献

Summary The effect of frequency-dependent advantage in wheat was investigated by growing F1. hybrid seeds of the crosses (Warimek X Halberd) and (Wariquam x Halberd) in a stand of Halberd at 5 frequencies: 4 %, 6.25 %, 11.11 %, 25 % and 50 %. A reduction of 35 % to 40 % in grain yield of individual plants was observed with both hybrids as their frequencies changed from 4% to 50 %. A similar trend with frequency was noted for several other plant characteristics, including total grain number, particularly with those measured towards the end of the growing season. Halberd plants did not show a corresponding increase as their frequency declined from 96 % to 50%. — The following season, 76 F4 lines from the cross (Warimek X Halberd) and 70 F4 lines from (Wariquam x Gabo) were grown at frequencies of 6.25% and 18.75% in machine sown stands of Wariquam and Halberd, respectively. Again grain yield decreased as genotypic frequency increased. Furthermore, there was a positive correlation between frequency-dependent advantage and relative grain yield, suggesting that high yielding genotypes show a greater advantage at low frequencies than lower yielding ones. 相似文献

The recently developed MAC (morphology-antibody-chromosome) method allows simultaneous immunophenotype and karyotype analysis in the same cell. To date, application of this new method has been hampered by the poor quality of chromosome banding. In this paper, we describe a modified simultaneous immunofluorescence and Q-banding technique, as well as a new combination of immunohistochemical and fluorescent R-banding methods. By further modifying the MAC method, we were able not only to achieve unequivocal results with weakly expressed antigens but also to improve the quality of the banding techniques, so that even structural chromosome abnormalities were well defined. 相似文献
