We found a dramatic upregulation in the expression of LC3 in the hippocampus of rats upon status epilepticus (SE). However, the enhancement in LC3 expression might be caused by a reduction in lysosomal activity or by alterations in autophagosome-lysosome fusion leading to a cytosolic vesicular retention. In order to dissect this aspect, we monitored the spatial and temporal expression of LC3 and LAMP1 in the hippocampus of rats with SE. The Western blot analysis showed that the expression of LAMP1 was slightly increased in hippocampal cells at 6, 24, and 48 h post-SE. However, immunofluorescence analysis showed dramatic spatial changes in LAMP1 distribution within the hippocampus. LAMP1 in controls was localised only in cytosol as dot like staining, however at 24 h post-SE LAMP1 was not only highly expressed, but accumulated in mossy fibers of dentate gyrus. In parallel, we found few scattered LC3-positive-dots in neurites of dentate gyrus which co-localise with LAMP1-positive structures. We conclude that SE not only increased autophagosomal abundance, but also lysosomal activities and a massive accumulation of LAMP1 in axons of dentate gyrus. This could support the hypothesis that the marked increased autophagosomal abundance in cytosol reflects an increase in the autophagic activity more than an inhibition of autophagosomal clearance. Although LAMP1 may have contributed to cell damage in the selective vulnerable hippocampal CA1-subfield, it is also possible that lysosomal/autophagic mechanisms in mossy fibers were compensatory and reflected an attempt to survive the epileptic insult by breaking down non-essential components.  相似文献

The ECM is composed of various cell-adhesive glycoproteins, such as, fibronectin (FN), laminin (LN), and different types of glycosaminoglycans and proteoglycans. These building blocks of the ECM are linked together to form a dense and complex tissue that fills the interstitial spaces and comprises the boundaries between cells and tissues. The ECM is the major milieu in which immune cells function during inflammatory processes (Shimizu and Shaw, 1991; Yamada, 1991). Recognition of ECM-glycoproteins by immune cells is mediated by very late activation (VLA) receptors, also referred to as integrins of the β1-subfamily (Hynes, 1992). A prerequisite of lymphocyte-ECM interactions is activation of the cells by mitogens, or via their CD3-T cell receptor complex, either of these types of activation modulates the affinity of otherwise inactive β1-integrins (Shimizu, et al., 1990).  相似文献

Examination of 904 skulls of wild caught marmosets revealed 92 with diseased teeth and alveolar infections. The number of affected teeth and alveoli was 259 or 0.9% of the total number of teeth including alveoli. Dental abnormalities such as super-numeraries, congenital assymetries, and postpartum injuries were noted in 14 skulls. The diseased individuals appeared in 83 of the 207 collecting localities, and constitute nearly 22% of the 488 specimens from those sites. Difference between the proportion of diseased upper teeth (9.4%), and lower teeth (8.3%) of all teeth examined, is insignificant. There is indication, however, that the lower the proportion of diseased antepremolar teeth, the higher the proportion of diseased postcanine teeth, and vice versa. Incidence of diseases is practically nil in juveniles and sub-adults, but rises rapidly in late maturity and old age. Fine line cracking or crazing of the enamel in marmoset teeth is briefly described and compared with same phenomenon in other mammals including man. Crazing is attributed to the cumulative effects of bite impact. It is absent in deciduous and unworn permanent primate teeth and absent or uncommon in mammalian teeth adapted to an insectivorous diet.  相似文献

The present study aimed to investigate the effect of walnut consumption on lipid profile, steroid hormones and inflammation in trained elderly men performing concurrent (resistance and endurance) training. Twenty healthy elderly males were divided into two matched groups, in a randomized controlled trial, that trained three sessions per week: concurrent training + dietary walnut consumption (15 g/day for six weeks, CTW: n = 10); concurrent training + control diet (CT: n = 10). Fasting blood samples were taken 48 hours before and after intervention for biochemical assessments. levels of high-density lipoprotein (HDL) increased only in CTW compared to baseline (19.8%, p < 0.01). Total cholesterol (TC), low-density lipoprotein (LDL) and triglyceride (TG) levels significantly decreased only for CTW (i.e., 13%, 18%, and 18.5% at p < 0.01 for all). Testosterone (T) increased after the training compared to pre-training for CTW and CT (10.3%, p < 0.01, 4.27% p < 0.05, respectively), but the increase was significantly higher in CTW (p < 0.05). Serum cortisol (C) was lower for CTW compared to CT (p < 0.01). C-reactive protein (CRP) decreased in CTW in comparison with CT. The present study revealed that 6-week moderate walnut supplementation (15 g/day) improved lipid profile, steroid hormones and systematic inflammation in aged men performing concurrent training. These findings could be attributable to the potential effect of polyunsaturated fatty acids (PUFA) contained in walnut (linoleic acid, n-6; linolenic acid, n-3).  相似文献

Abstract: Specific stress polypeptides were detected in three drought-resistant cyanobacteria ( Phormidium autumnale , LPP4 and Chroococcidiopsis sp.) subjected to matric- and osmotic-water stress. Drought stress caused the induction of at least 2–3 new polypeptides of apparent molecular masses of 30 kDa and 40–45 kDa. The polypeptide of 30 kDa was located in the thylakoid membranes, and the 45-kDa polypeptide in the cytoplasm. When these cyanobacteria were exposed to salt stress polypeptides of similar size appeared.  相似文献
