The marine red alga genus Laurencia is one of the richest producers of unique brominated compounds in the marine environment. The cDNAs for two Laurencia nipponica vanadium-dependent bromoperoxidases (LnVBPO1 and LnVBPO2) were cloned and expressed in Escherichia coli. Enzyme assays of recombinant LnVBPO1 and LnVBPO2 using monochlorodimedone revealed that they were thermolabile but their Km values for Br? were significantly lower than other red algal VBPOs. The bromination reaction was also assessed using laurediol, the predicted natural precursor of the brominated ether laurencin. Laurediol, protected by trimethylsilyl at the enyne, was converted to deacetyllaurencin by the LnVBPOs, which was confirmed by tandem mass spectrometry. Native LnVBPO partially purified from algal bodies was active, suggesting that LnVBPO is functional in vivo. These results contributed to our knowledge of the biosynthesis of Laurencia brominated metabolites.  相似文献

Avian malaria parasites (Plasmodium) have a worldwide distribution except for Antarctica. They are transmitted exclusively by mosquito vectors (Diptera: Culicidae) and are of particular interest to health care research due to their phylogenetic relationship with human plasmodia and their ability to cause avian malaria, which is frequently lethal in non-adapted avian hosts. However, different features of avian Plasmodium spp, including their taxonomy and aspects of their life-history traits, need to be examined in more detail. Over the last 10 years, ecologists, evolutionary biologists and wildlife researchers have recognized the importance of studying avian malaria parasites and other related haemosporidians, which are the largest group of the order Haemosporida by number of species. These studies have included understanding the ecological, behavioral and evolutionary aspects that arise in this wildlife host-parasite system. Molecular tools have provided new and exiting opportunities for such research. This review discusses several emerging topics related to the current research of avian Plasmodium spp and some related avian haemosporidians. We also summarize some important discoveries in this field and emphasize the value of using both polymerase chain reaction-based and microscopy-based methods in parallel for wildlife studies. We will focus on the genus Plasmodium, with an emphasis on the distribution and pathogenicity of these parasites in wild birds in Brazil.  相似文献

This study investigated whether surface hole counts could be used as a reliable estimate of density of the ghost shrimps Trypaea australiensis Dana 1852 and Biffarius arenosus Poore 1975 (Decapoda, Thalassinidea) in south eastern Australia. The relationship between the number of holes and the number of ghost shrimps was explored in two ways. Resin casts were used to document any changes in the number of burrow openings per shrimp burrow over time. Manual suction pumping (bait pumping) within a given mudflat area was used to directly compare the number of holes on the sediment surface with the number of ghost shrimps occupying the corresponding volume of sediment. Resin casting showed that throughout the year, the burrows of T. australiensis consistently had an average of two openings, whereas the burrows of B. arenosus showed much greater variability over time with two to four openings per burrow. Overall, a significant relationship between the number of holes and the number of ghost shrimps (mixed species populations) was found, with 2.1 burrow openings for each ghost shrimp. However, some temporal and spatial variation was seen in this relationship. We suggest that the hole count method may be reliable in estimating ghost shrimp densities with restricted use and site specific validation based on some limitations found in this study. We studied the seasonal and annual variation in diet composition of the fat dormouse (Glis glis) in Lithuania, a locality situated on the northern periphery of the dormouse range and outside of the range of the European beech (Fagus sylvatica). After emergence from hibernation, dormice fed on oak acorns (from the previous year), inflorescences of various trees, vegetative parts of plants and food of animal origin (birds, their eggs and insects). In June, soft mast and seeds of birches supplemented the dormouse diet, and diet composition was the most diverse during this period. In July, raspberries and fruits of glossy buckthorn constituted the bulk of dormouse diet, but seeds of birches dominated in a specific year. Hard mast (mainly acorns) dominated the diet of G. glis from August until the beginning of hibernation in October. A high prevalence of acorns, comparatively high proportion of birch seeds and low proportion of food of animal origin in the diet, as well as feeding on fruits of glossy buckthorn, are specific features of feeding by G. glis in Lithuania. The diet of G. glis on the northern periphery of its range resembles its diet on the eastern periphery of the range where beech trees are also absent. According to the composition of G. glis diet, feeding conditions in both of these peripheral regions are poorer in comparison to central or southern regions.  相似文献

Salmonellae are a major cause of food-borne outbreaks in Europe, with eggs and egg products being identified as major sources. Due to the low levels of salmonellae in eggs and egg products, direct quantification is difficult. In the present study, enrichment quantitative real-time PCR (qPCR) was employed for enumeration of salmonellae in different matrices: table eggs, pasteurized egg products, and egg-containing dishes. Salmonella enterica serovar Enteritidis and S. enterica serovar Tennessee were used to artificially contaminate these matrices. The results showed a linear regression between the numbers of salmonellae and the quantification cycle (Cq) values for all matrices used, with the exception of pasteurized egg white. Standard curves were constructed by using both stationary-phase cells and heat-stressed cells, with similar results. Finally, this method was used to evaluate the fate of salmonellae in two egg-containing dishes, long egg and tiramisu, at abused refrigeration temperatures, and results indicated the growth of bacteria over a 1-week period. In conclusion, enrichment qPCR was shown to be reliable for enumeration of salmonellae in different egg products.  相似文献

Objective(s)

We sought to prospectively study the association between antenatal emotional distress and gestational length at birth as well as preterm birth.

Study Design

We followed up 40,077 primiparous women in the Norwegian Mother and Child Cohort Study. Emotional distress was reported in a short form of the Hopkins Symptom Checklist-25 (SCL-5) at 17 and 30 weeks of gestation. Gestational length at birth, obtained from the Medical Birth Registry of Norway, was used as continuous (gestational length in days) and categorized (early preterm (22–31 weeks) and late preterm (32–36 weeks) versus term birth (≥37 weeks)) outcome, using linear and logistic regression analysis, respectively. Births were divided into spontaneous and provider-initiated.

Results

Of all women, 7.4% reported emotional distress at 17 weeks, 6.0% at 30 weeks and 5.1% had a preterm birth. All measurements of emotional distress at 30 weeks were significantly associated with a reduction of gestational length, in days, for provider-initiated births at term. Emotional distress at 30 weeks showed a reduced duration of pregnancy at birth of 2.40 days for provider-initiated births at term. An increase in emotional distress from 17 to 30 weeks was associated with a reduction of gestational length at birth of 2.13 days for provider-initiated births at term. Sustained high emotional distress was associated with a reduction of gestational length at birth of 2.82 days for provider-initiated births. Emotional distress did not increase the risk of either early or late preterm birth.

Conclusion

Emotional distress at 30 weeks, an increase in emotional distress from 17 to 30 weeks and sustained high levels of emotional distress were associated with a reduction in gestational length in days for provider-initiated term birth. We found no significant association between emotional distress and the risk of preterm birth.  相似文献

Urine samples from 14 mares, belonging to five breeds, were collected at estrus and diestrus to search for estrous specific volatile compounds which could be used to determine ovulation time. Around 150 volatiles were collected from urine head-space samples by solid phase micro extraction technique, and analyses were conducted by gas chromatography-mass spectrometry methods. Comparison of chromatographic profiles of volatile substances revealed that concentrations of m- and p-cresols were significantly greater during estrus while diethylphthalate was more abundant at diestrus. Monitoring of m- and p-cresols during the period of estrus and a few days before and after estrus revealed irregular changes in amounts of cresols until 3-4 days before ovulation when the concentration of the compounds began to increase with peaks 1 day before ovulation. On the day when ovulation occurred, amounts of the metabolites decreased sharply, almost to basal concentrations, and remained at these concentrations for 6 days - when sampling was finished. In four of the mares changes in the concentration of diethylphthalate were less pronounced and more temporally variable compared with those of cresols. Based on reproducible temporal changes in concentrations of m- and p-cresols, with respect to the time of ovulation, a noninvasive test to determine a precise insemination time could occur. This would save time, reduce costs and simplify the procedure.  相似文献

Changes in plasmalemma K+Mg2+-ATPase dephosphorylating activity and H+ transport were examined in freezing-tolerant and non-tolerant genotypes of the perennial grass species Festuca pratensis Huds. Enzyme activity and ΔμH+ were measured in plasmalemma fractions isolated from basal nodes and roots. Three types of experiments were undertaken: (i) a field experiment, utilizing the seasonal growth and cessation cycle of a perennial plant; (ii) a cold acclimation experiment in hydroponics; and (iii) an instant freezing test. A specific fluctuation in K+Mg2+-ATPase activity was found throughout the seasonal growth of the plants (i). The K+Mg2+-ATPase activity peaks for both the basal node and the root plasmalemma were determined early in the spring before the renewal of growth. The lowest activity values in roots occurred at the time approaching flowering, and in basal nodes at the transition into the growth cessation. The K+Mg2+-ATPase activity was approximately 50% lower in the basal node plasmalemma of freezing-tolerant plants than of non-tolerant ones, when assessed at the optimal growth stage in hydroponics. In hydroponics (ii) and in the freezing test (iii), temperature stress was followed by a more pronounced change in the level of K+Mg2+-ATPase activity than in that of H+ transport, and this change was more clearly differentiated in the basal node plasmalemma of contrasting genotypes than in the roots. Stress response was manifested differently in freezing-tolerant and non-tolerant plants at cold acclimation (4–2 °C) and at freezing (−8 °C) temperatures. Proton transport regulation via coupled changes in the hydrolysed ATP/transported proton ratio, as an attribute of freezing-tolerant plants, is discussed.  相似文献
