Autoradiographic model experiments with 67Ga and 99mTc 总被引:1,自引:0,他引:1
Summary In order to evaluate the feasibility to localize correctly 67Ga citrate and 99mTc pertechnetate in tissues, the resolution of these radioactive compounds were measured in a model system using four different autoradiographic techniques, wet as well as dry.Wet autoradiographic techniques gave an almost complete loss of 67Ga. In deparaffinized sections of fixed and paraffin-embedded tissue the remaining 67Ga, which was probably bound to proteins, gave a resolution of 2.6 . 99mTc was either completely lost in wet techniques or the procedure could not be performed at all because of the very short half life of 99mTc.The resolution of 67Ga in a dry autoradiographic technique (according to Stumpf) was 6.9 and the resolution of 99mTc 22.6 .The technique in which frozen sections are thawed on dry film and consequently dryed, gave slightly better resolutions than the dry technique (according to Stumpf) with 67Ga as well as 99mTc.It is concluded that for the histological localization of 67Ga and 99mTc a dry technique is required. However, the use of a wet technique can be considered, provided a loss of the radioisotope is acceptable and the procedure is controlled by a dry technique. 相似文献

Female guinea pigs were exposed prenatally (Day 28–58) to the propionates of testosterone (TP), dihydrotestosterone (DHTP), or androstanediol (ADP). Only TP females failed to display lordosis in adulthood after estrogen and progesterone treatment. When given TP in adulthood, females in all groups mounted, but TP and DHTP females showed augmented intromission frequencies and higher percentages of correctly oriented mounts relative to controls. Moreover, TP females responded more quickly to TP injections in adulthood and had higher over-all mounting frequencies than other groups, while DHTP females displayed mounting frequencies intermediate to controls and TP females. ADP females were not different from controls for any measure of mounting behavior.No female in any group mounted when given DHTP in adulthood, even after 7 wk of daily injections. Since male guinea pigs do mount in response to DHTP given in adulthood, the results raise the possibility that mechanisms determining sensitivity to specific steroids may not be mediated exclusively by steroids during critical periods of embryological differentiation. 相似文献

The divalent cation requirement for mitochondrial cardiolipin biosynthesis has been further investigated. The relative order of divalent cation activity was Co-2+ greater than Mn-2+ greater than Mg-2+. Cardiolipin was not formed in the incubations with Zn-2+, Fe-2+, Cu-2+, Hg-2+, and Ca-2+. Cardiolipin synthesis in the presence of optimal cincentration of Co-2+ was inhibited by Ca-2+. A series of CDP-diglycerides was synthesized having differences in fatty acid chain lenth and degree of unsaturation. These compounds were tested in mitochondrial cardiolipin and phosphatidylglycerol synthesis. Although there were some minor differences between phosphatidylglycerol and cardiolipin synthesis, in general, saturated shorter chain CDP-diglycerides (dilauroyl and dimyristoyl) were better substrates than the longer chain dipalmitoyl and distearoyl homologues. Introduction of double bonds into distearoyl CDP-diglyceride resulted in more rapid rates of synthesis (e.g. dioleoyl and dilinoleoyl CDP-diglyceride). The rate of muscle cell fusion increases between 28 °C and 40 °C by a factor of 15 to 20. The formal activation energy of the fusion process changes abruptly at about 35 °C. This change is discussed in terms of a phase transition of the membrane lipids at 35 °C. In the presence of cholesterol or dipalmitoyllecithin the fusion rate decreases markedly. Increasing the temperature reverses the effects of cholesterol and dipalmitoyllecithin. These results are discussed in terms of interactions between membrane lipids. 相似文献

The group III metabotropic glutamate receptor subtype 7 (mGlu7) is an important regulator of glutamatergic and GABAergic neurotransmission and known to mediate emotionality and male social behavior. However, a possible regulatory role in maternal behavior remains unknown to date. Adequate expression of maternal behavior is essential for successful rearing and healthy development of the young. By understanding genetic and neural mechanisms underlying this important prosocial behavior, we gain valuable insights into possible dysregulations. Using genetic ablation as well as pharmacological modulation, we studied various parameters of maternal behavior in two different mouse strains under the influence of mGlu7. We can clearly show a regulatory role of mGlu7 in maternal behavior. Naïve virgin female C57BL/6 mGlu7 knockout mice showed more often nursing postures and less spontaneous maternal aggression compared to their heterozygous and wildtype littermates. In lactating C57BL/6 wildtype mice, acute central activation of mGlu7 by the selective agonist AMN082 reduced arched back nursing and accelerated pup retrieval without affecting maternal aggression. In addition, in lactating CD1 wildtype mice the selective mGlu7 antagonist XAP044 increased both pup retrieval and maternal aggression. With respect to receptor expression levels, mGlu7 mRNA expression was higher in lactating vs virgin C57BL/6 mice in the prefrontal cortex, but not hypothalamus or hippocampus. In conclusion, these findings highlight a significant role of the mGlu7 receptor subtype in mediating maternal behavior in mice. Region‐dependent studies are warranted to further extend our knowledge on the specific function of the brain glutamate system in maternal behavior. 相似文献

Genomic approaches permit direct estimation of inbreeding and its effect on fitness. We used genomic‐based estimates of inbreeding to investigate their relationship with eight adult traits in a captive‐reared Pacific salmonid that is released into the wild. Estimates were also used to determine whether alternative broodstock management approaches reduced risks of inbreeding. Specifically, 1,100 unlinked restriction‐site associated (RAD) loci were used to compare pairwise relatedness, derived from a relationship matrix, and individual inbreeding, estimated by comparing observed and expected homozygosity, across four generations in two hatchery lines of Chinook salmon that were derived from the same source. The lines are managed as “integrated” with the founding wild stock, with ongoing gene flow, and as “segregated” with no gene flow. While relatedness and inbreeding increased in the first generation of both lines, possibly due to population subdivision caused by hatchery initiation, the integrated line had significantly lower levels in some subsequent generations (relatedness: F2–F4; inbreeding F2). Generally, inbreeding was similar between the lines despite large differences in effective numbers of breeders. Inbreeding did not affect fecundity, reproductive effort, return timing, fork length, weight, condition factor, and daily growth coefficient. However, it delayed spawn timing by 1.75 days per one standard deviation increase in F (~0.16). The results indicate that integrated management may reduce inbreeding but also suggest that it is relatively low in a small, segregated hatchery population that maximized number of breeders. Our findings demonstrate the utility of genomics to monitor inbreeding under alternative management strategies in captive breeding programs. 相似文献
