Ozone (O3) and nitrogen (N) deposition affect plant carbon (C) dynamics and may change ecosystem C‐sink/‐source properties. We studied effects of increased background [O3] (up to [ambient] × 2) and increased N deposition (up to +50 kg ha?1 a?1) on mature, subalpine grassland during the third treatment year. During 10 days and 13 nights, distributed evenly over the growth period of 2006, we measured ecosystem‐level CO2 exchange using a static cuvette. Light dependency of gross primary production (GPP) and temperature dependency of ecosystem respiration rates (Reco) were established. Soil temperature, soil water content, and solar radiation were monitored. Using Reco and GPP values, we calculated seasonal net ecosystem production (NEP), based on hourly averages of global radiation and soil temperature. Differences in NEP were compared with differences in soil organic C after 5 years of treatment. The high [O3] had no effect on aboveground dry matter productivity (DM), but seasonal mean rates of both Reco and GPP decreased ca. 8%. NEP indicated an unaltered growing season CO2–C balance. High N treatment, with a +31% increase in DM, mean Reco increased ca. 3%, but GPP decreased ca. 4%. Consequently, seasonal NEP yielded a 53.9 g C m?2 (±22.05) C loss compared with control. Independent of treatment, we observed a negative NEP of 146.4 g C m?2 (±15.3). Carbon loss was likely due to a transient management effect, equivalent to a shift from pasture to hay meadow and a drought effect, specific to the 2006 summer climate. We argue that this resulted from strongly intensified soil microbial respiration, following mitigation of nutrient limitation. There was no interaction between O3 and N treatments. Thus, during the 2006 growing season, the subalpine grassland lost >2% of total topsoil organic C as respired CO2, with increased N deposition responsible for one‐third of that loss.  相似文献

The pleiotropic mutant lethal(3)giant larvae [l(3)gl] of Drosophila hydei exhibits among other anatomical defects, hypertrophy of the larval brain and imaginal discs. Both hypertrophic tissues when transplanted into wild-type female flies behave as fast growing and lethal neoplasms. Implanted into mature wild-type larvae they fail to metamorphose. When l(3)gl neoplastic brain tissue or imaginal discs were mixed with normal imaginal discs, cultured in vivo in the abdomen of adult females and transplanted into mature wild-type larvae, the following results were obtained. The invasive l(3)gl brain neoplasm, while fatal for adult hosts, had no effect on the metamorphosis of normal imaginal disc tissue. On the other hand, the noninvasive l(3)gl imaginal disc neoplasms when mixed with normal imaginal disc tissue inhibited its development and metamorphosis in the wild-type host. This inhibitory effect was not observed when the tissues were injected as separate implants into the same host.  相似文献

The differentiation of high- and low-affinity postsynaptic gamma-aminobutyric acid (GABA) receptors was examined in a washed cortical membrane preparation of the rat. The selective elimination of the high- and low-affinity GABA sites by the chaotropic anion thiocyanate and diazotization by p-diazobenzenesulfonic acid (DSA), respectively, offered two model systems for the separate sites. The [3H]GABA displacing potencies of some GABA agonists [GABA, 4,5,6,7-tetrahydro- isoxazole [4,5c]pyridine-3-ol (THIP), and muscimol] and antagonists [bicuculline methiodide (BCM), 3-alpha-hydroxy-16-imino-5 beta-17-aza-androstan-11-one (R-5135), and d-tubocurarine] and their slope factors were examined in these model systems and in control membranes. The displacing potency of the agonists was increased in the DSA-pretreated membranes and decreased in the presence of thiocyanate. The displacing potency of the antagonists was shifted in an opposite manner. The chaotropic effect of thiocyanate was reversible and not additive with the inhibitory effect of diazotization on the specific binding of GABA. Inhibition of specific GABA binding by pyridoxal-5-phosphate (PLP) could not be protected by GABA antagonists (BCM and R-5135) but only by agonists. The results can be interpreted in the framework of a dual (agonist-antagonist) receptor model, postulating a hydrophobic accessory site at the low-affinity GABA receptor. The effect of thiocyanate on the GABA receptor may result in the exposure of the hydrophobic accessory sites.  相似文献

Nitrous oxide (N2O) fluxes from soil under mown grassland were monitored using static chambers over three growing seasons in intensively and extensively managed systems in Central Switzerland. Emissions were largest following the application of mineral (NH4NO3) fertilizer, but there were also substantial emissions following cattle slurry application, after grass cuts and during the thawing of frozen soil. Continuous flux sampling, using automatic chambers, showed marked diurnal patterns in N2O fluxes during emission peaks, with highest values in the afternoon. Net uptake fluxes of N2O and subambient N2O concentrations in soil open pore space were frequently measured on both fields. Flux integration over 2.5 years yields a cumulated emission of +4.7 kgN2O‐N ha?1 for the intensively managed field, equivalent to an average emission factor of 1.1%, and a small net sink activity of ?0.4 kg N2O‐N ha?1 for the unfertilized system. The data suggest the existence of a consumption mechanism for N2O in dry, areated soil conditions, which cannot be explained by conventional anaerobic denitrification. The effect of fertilization on greenhouse gas budgets of grassland at the ecosystem level is discussed.  相似文献

Melanoma differentiation‐associated protein 5 (MDA5) mediates the innate immune response to viral infection. Polymorphisms in IFIH1, the gene coding for MDA5, correlate with the risk of developing type 1 diabetes (T1D). Here, we demonstrate that MDA5 is crucial for the immune response to enteric rotavirus infection, a proposed etiological agent for T1D. MDA5 variants encoded by minor IFIH1 alleles associated with lower T1D risk exhibit reduced activity against rotavirus infection. We find that MDA5 activity limits rotavirus infection not only through the induction of antiviral interferons and pro‐inflammatory cytokines, but also by promoting cell death. Importantly, this MDA5‐dependent antiviral response is specific to the pancreas of rotavirus‐infected mice, similar to the autoimmunity associated with T1D. These findings imply that MDA5‐induced cell death and inflammation in the pancreas facilitate progression to autoimmune destruction of pancreatic β‐cells.  相似文献

Production of haploid plants has been restricted to only a few ornamental species. In this paper an efficient anther culture protocol has been devised for production of haploid plants of Phlox drummondii, a garden ornamental. Anthers with microspores at early- to late-uninucleate stages were inoculated on MS (Murashige and Skoog, Physiol Plant 15:473–479, 1962) basal medium containing 9% sucrose, 10 μM 2,4-D + 5 μM BA in the dark for callus induction. The callus (~2 mm) was transferred to MS medium containing 3% sucrose + 10 μM BA + 5 μM NAA under a 16 h photoperiod for multiplication. Anther-derived callus showed the greatest shoot differentiation (60% with greater than 3 shoots per culture) at 13 weeks after culture initiation when maintained on MS medium supplemented with 3% sucrose and cytokinin (7.5 μM BA). At 68 weeks, only 4.6% of cultures differentiated with less than one shoot per callus. Anther-derived shoots rooted readily on MS medium containing 7.5 μM IAA. Of 60 plants that regenerated from anther callus, 50% were haploid, 30% diploid, and 20% aneuploid. Developed protocol could be useful for the haploid induction of outcrossing ornamental plants for production of their homozygous double haploids.  相似文献

Triploid plants of neem were obtained by immature endosperm culture. Immature seeds, at the early dicotyledonous stage of embryo development, is the best explant to raise endosperm callus on MS + NAA (5 mumol/L) + BAP (2 mumol/L) + CH (500 mg L-1). Maximum shoot bud differentiation from the endosperm callus occurred on MS + 5 mumol/L BAP. Shoots were multiplied by forced axillary branching and rooted in vitro. The plants were established in soil. Over 66% of the plants were triploid with chromosome number 2n = 3x = 36. A characteristic feature of the shoots of endosperm origin is the presence of a large number of multi-cellular glands.  相似文献
