1. The interaction between [(14)C]carboxymethylated cytochrome c and monolayers of egg phosphatidylethanolamine at the air/water interface has been investigated by measurements of surface radioactivity, pressure and potential. 2. On adding (14)C-labelled cytochrome c to the subphase under monolayers with a surface pressure below 24dynes/cm. there was an initial surface pressure increment as the protein penetrated, followed by an adsorption that could be detected only by a continued increase in the surface radioactivity. 3. Above film pressures of 24dynes/cm. only adsorption was observed, i.e. an increment in surface radioactivity with none in surface pressure. 4. The changes in surface parameters with penetration of cytochrome c added to the subphase were indirectly proportional to the initial pressure of the monolayer. With hydrogenated phosphatidylethanolamine the constant of proportionality was increased but penetration again ceased at 24dynes/cm. 5. On compressing a phosphatidylethanolamine film containing penetrated cytochrome c to 40dynes/cm. only a proportion of the protein was ejected on a subphase of 10mm-sodium chloride, whereas on a subphase of m-sodium chloride nearly all the protein was lost. 6. With both penetration and adsorption only a small proportion of the added cytochrome c interacted with the phospholipid films, and initially the amount bound was proportional to the added protein concentration. There was no evidence of a stoicheiometric relationship between the protein and phospholipid or the build-up of multilayers. The bonded protein was not released by removing cytochrome c from the subphase. 7. The addition of m-sodium chloride to the subphase delays the rate of protein penetration into low-pressure films, but the final surface-pressure increment is not appreciably decreased. In contrast, m-sodium chloride almost completely stops adsorption on to films at all pressures. 8. When sodium chloride is added to the subphase below cytochrome c adsorbed to monolayers at high pressures, so that the final concentration is 1m, only a proportion of the protein is desorbed and this decreases as the time of the interaction increases. This indicates that adsorption is initially electrostatic, followed by the formation of non-ionic bonds. 9. Alteration of the subphase pH under a high-pressure film leads to a steady increase in adsorption from pH3 to 8.5 followed by a rapid fall to zero adsorption at pH11. 10. The penetration into phospholipid monolayers at 10dynes/cm. shows a rate that is consistent with the relative electrostatic status of the two components of the interaction as the subphase pH is varied between 3 and 10.5. The final equilibrium penetration shows a pronounced peak in the increments of surface pressure at pH9.0 although a similar peak is not observed in the surface radioactivity. This indicates that more residues of the protein are penetrating into the film at about this pH. 11. Determinations were made of the electrophoretic mobilities of phosphatidylethanolamine particles both alone and after interaction with cytochrome c. 12. The electrophoretic mobilities of cytochrome c adsorbed on lipid particles showed an isoelectric point below that of cytochrome c. This and the observations on the monolayers suggest that, with cytochrome c, protein-protein interactions are weak compared with other proteins.  相似文献

The marine natural product, halistanol trisulfate, has a relatively low critical micelle concentration of 0.001% m/v (14.5 microM) and strong hemolytic potency with an EC50 of 0.00046% m/v (6.67 microM). As expected of a detergent, it inhibits the growth of gram-positive but not gram-negative bacteria. The hemolytic activity of halistanol trisulfate and other detergents has been shown to correlate with critical micelle concentration. This correlation may have important implications in the mechanism of membranolytic bioactivity.  相似文献

Pacific salmon are noted for their lengthy foraging migrations and for their precise homing ability. Extensive sampling has documented the general migratory patterns of the major populations, but many basic aspects of their marine ecology are still poorly understood. Their life history pattern has been interpreted as an adaptation to exploit the higher productivity of the marine environment over that in fresh water. The adaptive significance of homing is implied by the specializations of populations for their natal habitat and the competitive superiority of locally adapted populations over transplants from other rivers. However, the establishment of new populations by strays and the levels of gene flow between natural populations have only recently received much attention. Research on salmon migrations has also focused on the mechanisms that guide homing at sea and in fresh water. While salmon have highly developed sensory systems, the ways in which inputs are integrated to guide migration through diverse and complex habitats are still being investigated.  相似文献

A laboratory study was conducted to determine the effects of defoliation and denodulation on compensatory growth of Medicago sativa (L.). Plants grown hydroponically in clear plastic growth pouches were subjected to 0 and 50% nodule pruning, and 0, 25, 50, and 75% defoliation by clipping trifoliate leaves. An additional experiment was conducted to determine if clipping leaves simulated herbivory by Hypera postica (Gyllenhal) larvae. Previously, we determined that nodule pruning accurately simulated herbivory by Sitona hispidulus (L.) larvae (Quinn & Hall, 1992). Results indicated that denodulation stimulated nodule growth and caused exact compensation in standing and total number of nodules per plant within 15 days and in standing nodule biomass within 22 days of treatment. Denodulation caused a significant reduction (13%) in final shoot biomass, but did not affect significantly final root biomass. Percentage of change in number of trifoliate leaves per plant increased with the level of defoliation. Within 22 days of treatment, total number of trifoliate leaves per plant was similar to controls. However, final standing shoot biomasses were significantly less that controls, indicating undercompensatory growth. Shoot biomasses of the 25-, 50-, and 75%-defoliated plants were 18, 20, and 36% lower than controls, respectively. Nodule biomass per plant was reduced by 24 and 32% in 50- and 75%-defoliated plants, respectively, but was not affected significantly by 25% defoliation. Root biomass was affected by all levels of defoliation. Clipping trifoliate leaves accurately simulated defoliation by H. postica larvae. Our results indicated that partial defoliation affected shoot, root, and nodule biomass of M. sativa, but that partial denodulation only affected shoot biomass.  相似文献

A number of methods have been used for quantifying the sizes of the geographic ranges of species. The consequences of different levels of sampling (the proportion of actual spatial occurrences) are explored for eight of these, using data on the occurrences of butterfly species on a 10 × 10 km grid across Britain. For all methods, the percentage error of estimation (PEE) decreases with the number of 10 × 10 km squares which a species occupies, most rapidly for extent measures, and more rapidly for area measures than for measures of numbers of units occupied. The rate of decline in PEE itself falls as sampling effort increases. At a given sampling level, rank correlations between range sizes measured by different methods are generally high, but there is no consistent change in the magnitude of these correlations as the level of sampling increases. The composition of the set of species with the smallest range sizes changes with the level of sampling.  相似文献

We review the recent history of the Mute Swan in Great Britain and discuss the factors known to be affecting the population. Following a rise in the national population during the 1950s, numbers decreased sharply during the 1960s, and changed relatively little between 1970/71 and 1984/85. However, there has been considerable regional variation in the fortunes of Mute Swan populations during this period, with dramatic declines in some areas. Although several factors were thought to be contributing to such declines, poisoning from the ingestion of lead fishing weights was shown to be the largest single cause of death amongst swans in a number of areas. Voluntary measures to address this problem were initiated in 1982 and culminated in the banning by law of use of lead weights in 1987.Winter counts were used to investigate the current status and distribution of the Mute Swan in Great Britain and to examine long-term regional trends. The maximum total count reached 12600 birds in January 1990, which compares with an average of 9550 for the previous five winters. However, accounting for birds missed, the population may now number at least 25 000. Peak total numbers have mostly occurred in September, after which numbers remain approximately stable until December and then decline. Patterns of seasonal abundance vary between regions and habitats and these are discussed.The British population has increased dramatically since 1986/87 and reached its highest level for 27 years in 1987/88. There have been recent increases in most regions with record levels being reached mostly in 1987 or 1988, and there has been growth in the numbers on all habitat types, especially on reservoirs, gravel extraction pits and freshwater marshes. The timing of these increases corresponds very closely with the introduction of legislation against the use of lead fishing weights, and the incidence of lead poisoning is known to have been considerably reduced by such measures.  相似文献

Huperzine A, a potential agent for therapy in Alzheimer's disease and for prophylaxis of organophosphate toxicity, has recently been characterized as a reversible inhibitor of cholinesterases. To examine the specificity of this novel compound in more detail, we have examined the interaction of the 2 stereoisomers of Huperzine A with cholinesterases and site-specific mutants that detail the involvement of specific amino acid residues. Inhibition of fetal bovine serum acetylcholinesterase by (-)-Huperzine A was 35-fold more potent than (+)-Huperzine A, with KI values of 6.2 nM and 210 nM, respectively. In addition, (-)-Huperzine A was 88-fold more potent in inhibiting Torpedo acetylcholinesterase than (+)-Huperzine A, with KI values of 0.25 microM and 22 microM, respectively. Far larger KI values that did not differ between the 2 stereoisomers were observed with horse and human serum butyrylcholinesterases. Mammalian acetylcholinesterase, Torpedo acetylcholinesterase, and mammalian butyrylcholinesterase can be distinguished by the amino acid Tyr, Phe, or Ala in the 330 position, respectively. Studies with mouse acetylcholinesterase mutants, Tyr 337 (330) Phe and Tyr 337 (330) Ala yielded a difference in reactivity that closely mimicked the native enzymes. In contrast, mutation of the conserved Glu 199 residue to Gln in Torpedo acetylcholinesterase produced only a 3-fold increase in KI value for the binding of Huperzine A.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
