大山雀对巢箱颜色的识别和繁殖功效

为了确定大山雀对巢箱颜色是否能够识别和对繁殖功效的影响,以利于更好的进行保护,于2007—2010年3—7月,在吉林省左家自然保护区,通过悬挂黑、蓝、绿、白和红色巢箱,对大山雀的入住状况和繁殖参数进行了调查和分析。结果显示:2007、2008、2010年红色巢箱的入住率最高,2009年低于平均值;窝卵数除2010年红色巢箱组略低于其它颜色组平均值外,其它年份略高于其它组,而各年份的出飞数红色组均高于其它组。将其它组的合并均值与红色组进行了方差分析,结果表明,窝卵数之间无显著差异,而出飞数之间则有显著差异(F=17.65,df=1,P=0.04),表明红色组的出飞数高于其他组的平均值。     

Functional and structural characteristics of NY-ESO-1-related HLA A2-restricted epitopes and the design of a novel immunogenic analogue

NY-ESO-1, a commonly expressed tumor antigen of the cancer-testis family, is expressed by a wide range of tumors but not found in normal adult somatic tissue, making it an ideal cancer vaccine candidate. Peptides derived from NY-ESO-1 have shown preclinical and clinical trial promise; however, biochemical features of these peptides have complicated their formulation and led to heterogeneous immune responses. We have taken a rational approach to engineer an HLA A2-restricted NY-ESO-1-derived T cell epitope with improved formulation and immunogenicity to the wild type peptide. To accomplish this, we have solved the x-ray crystallographic structures of HLA A2 complexed to NY-ESO (157-165) and two analogues of this peptide in which the C-terminal cysteine residue has been substituted to alanine or serine. Substitution of cysteine by serine maintained peptide conformation yet reduced complex stability, resulting in poor cytotoxic T lymphocyte recognition. Conversely, substitution with alanine maintained complex stability and cytotoxic T lymphocyte recognition. Based on the structures of the three HLA A2 complexes, we incorporated 2-aminoisobutyric acid, an isostereomer of cysteine, into the epitope. This analogue is impervious to oxidative damage, cysteinylation, and dimerization of the peptide epitope upon formulation that is characteristic of the wild type peptide. Therefore, this approach has yielded a potential therapeutic molecule that satiates the hydrophobic F pocket of HLA A2 and exhibited superior immunogenicity relative to the wild type peptide.     

Energy transfer kinetics of phycoerythrocyanin trimer from cyanobacterium Anabaena variabilis (II)

The excitation energy transfer processes in trimeric PEC have been studied by using steady-state and time-resolved fluorescence spectra techniques in detail. The results indicate that the energy transfer processes should take place between α₈₄-PVB and β₈₄-or β₁₅₅-PCB chromophores with the time constants 34.7 ps and 175–200 ps individually; in contrast with monomeric PEC, from time-resolved fluorescence anisotropic spectrum technique, the decay constant of 45 ps which was assigned to the energy transfer time among three β₈₄-PCB chromophores was observed and the energy levels of β₈₄-and/or β₁₅₅-PCB chromophores were confirmed to turn over in trimeric PEC. 利用猪血清短期培养人体外周血淋巴细胞的研究

用猪血清代替小牛血清作人体外周血淋巴细胞短期培养,进行染色体分析及淋巴细胞转化的研究。在94例姐妹染色单体差别染色的培养中,培养液分别加入小牛血清与猎血清,细胞有丝分裂指数分别为3.91％和3.78％;21例常规法培养中,分裂指数分别为9.10％和9.21％;5例淋巴细胞转化试验,小牛血清和猪血清培养的转化率分别为69.52％和69.59％;16例阉割后公、母猪血清加入培养基中,细胞分裂指数(SCD法)分别为2.96％和3.14％。以上各项对照试验,均无显著性差异(P>0.05)本研究证实,猪血清可用于人体外周血淋巴细胞短期培养。     

Knockdown of von Hippel-Lindau protein decreases lung cancer cell proliferation and colonization

Although von Hippel-Lindau protein (pVHL) is known as a tumor suppressor in kidney and other organs, it remains unclear whether pVHL plays a role in lung cancer development. We investigated the role of pVHL in lung cancer cell proliferation, migration, and colonization using stable A549 cells with knockdown of pVHL. We found that knockdown of pVHL promotes epithelial-mesenchymal transition (EMT) in lung cancer cells. Knockdown of pVHL decreased tumor colonization in a tail-vein injection model and decreased cell proliferation, whereas overexpression of constitutive active HIF increased tumor colonization, suggesting a HIF-independent function of pVHL in lung. Knockdown of pVHL decreased phosphorylation of FAK and expression of integrin, suggesting that pVHL regulates lung cancer development via integrin/FAK signaling pathway.     

Prediction of drug efficacy for cancer treatment based on comparative analysis of chemosensitivity and gene expression data

The NCI60 database is the largest available collection of compounds with measured anti-cancer activity. The strengths and limitations for using the NCI60 database as a source of new anti-cancer agents are explored and discussed in relation to previous studies. We selected a sub-set of 2333 compounds with reliable experimental half maximum growth inhibitions (GI(50)) values for 30 cell lines from the NCI60 data set and evaluated their growth inhibitory effect (chemosensitivity) with respect to tissue of origin. This was done by identifying natural clusters in the chemosensitivity data set and in a data set of expression profiles of 1901 genes for the corresponding tumor cell lines. Five clusters were identified based on the gene expression data using self-organizing maps (SOM), comprising leukemia, melanoma, ovarian and prostate, basal breast, and luminal breast cancer cells, respectively. The strong difference in gene expression between basal and luminal breast cancer cells was reflected clearly in the chemosensitivity data. Although most compounds in the data set were of low potency, high efficacy compounds that showed specificity with respect to tissue of origin could be found. Furthermore, eight potential topoisomerase II inhibitors were identified using a structural similarity search. Finally, a set of genes with expression profiles that were significantly correlated with anti-cancer drug activity was identified. Our study demonstrates that the combined data sets, which provide comprehensive information on drug activity and gene expression profiles of tumor cell lines studied, are useful for identifying potential new active compounds.     

Sequence analysis and expression of a cDNA clone encoding tropomysin in <Emphasis Type="Italic">Sinonovacula constricta</Emphasis>

Shellfish can cause severe anaphylactic reactions. Tropomyosin has been assumed partly responsible for the cross-reactivity among shellfish and other invertebrates. In this study, cDNA of Sinonovacula constricta was amplified by RT-PCR and 3′-RACE from total RNA. The obtained tropomyosin cDNA included an open reading frame coding for 284 amino acids. The deduced amino acid sequence of the corresponding protein shared high identity with other allergenic tropomyosins. Expression of the recombinant tropomyosin was carried out in Escherichia coli BL21(DE3) using vector PET28a and the purification of the recombinant protein was performed via affinity chromatography. IgE reactivity of recombinant tropomyosin was investigated by immunoblot and the sensized precentage was 36% which indicated that tropomyosin was the minor allergens in S. constricta.
