A functional detection assay was developed for Escherichia coli secreted shiga-like toxin based on antibody capture and visualization with a multivalent galabiose ligand. It was possible to detect verotoxin in medically relevant E. coli samples in a dose dependent fashion. This method is a new step towards measuring functional protein levels in complex mixtures, which can be used for diagnostic purposes in a clinical setting.  相似文献

Recent studies have concluded that release from native soil pathogens may explain invasion of exotic plant species. However, release from soil enemies does not explain all plant invasions. The invasion of Ammophila arenaria (marram grass or European beach grass) in California provides an illustrative example for which the enemy release hypothesis has been refuted. To explore the possible role of plant–soil community interactions in this invasion, we developed a mathematical model. First, we analyzed the role of plant–soil community interactions in the succession of A. arenaria in its native range (north-western Europe). Then, we used our model to explore for California how alternative plant–soil community interactions may generate the same effect as if A. arenaria were released from soil enemies. This analysis was carried out by construction of a 'recovery plane' that discriminates between plant competition and plant–soil community interactions. Our model shows that in California, the accumulation of local pathogens by A. arenaria could result in exclusion of native plant species. Moreover, this mechanism could trigger the rate and spatial pattern of invasive spread generally observed in nature. We propose that our 'accumulation of local pathogens' hypothesis could serve as an alternative explanation for the enemy release hypothesis to be considered in further experimental studies on invasive plant species.  相似文献

Maximum quantum yield for leaf CO2 assimilation under limiting light conditions (Φ CO2LL) is commonly estimated as the slope of the linear regression of net photosynthetic rate against absorbed irradiance over a range of low-irradiance conditions. Methodological errors associated with this estimation have often been attributed either to light absorptance by non-photosynthetic pigments or to some data points being beyond the linear range of the irradiance response, both causing an underestimation of Φ CO2LL. We demonstrate here that a decrease in photosystem (PS) photochemical efficiency with increasing irradiance, even at very low levels, is another source of error that causes a systematic underestimation of Φ CO2LL. A model method accounting for this error was developed, and was used to estimate Φ CO2LL from simultaneous measurements of gas exchange and chlorophyll fluorescence on leaves using various combinations of species, CO2, O2, or leaf temperature levels. The conventional linear regression method under-estimated Φ CO2LL by ca. 10–15 %. Differences in the estimated Φ CO2LL among measurement conditions were generally accounted for by different levels of photorespiration as described by the Farquhar-von Caemmerer–Berry model. However, our data revealed that the temperature dependence of PSII photochemical efficiency under low light was an additional factor that should be accounted for in the model.  相似文献

The distribution and proliferation of CFUs from bone marrow and spleen cell suspensions were followed after injection in lethally irradiated isogeneic mice. It was found that a larger proportion of the injected bone marrow CFUs than of the spleen derived CFUs could be recovered from the recipient's spleen and femur. This consistently higher recovery points to the conclusion that a larger fraction of bone marrow-derived CFUs than of spleen-derived CFUs is capable of producing daughter CFUs, most likely due to a commitment to early differentiation of many spleen CFUs.  相似文献

Marine fisheries science is a broad field that is fundamentally concerned with sustainability across ecological, economic, and social dimensions. Ensuring the delivery of food, security, equity, and well-being while sustaining ecosystems in the face of rapid change is, by far, the main challenge facing marine fisheries. A tighter integration of modeling and empiricism is needed to confront this challenge. In particular, improved incorporation of empirically grounded and realistic representation of human behaviors into models will greatly enhance our ability to predict likely outcomes under alternative adaptive strategies. Challenges to this integration certainly exist, but many of these can be overcome via improved professional training that reduces cultural rifts between empiricists and modelers and between natural and social sciences, ideally ending the presumption that there is a divide between empiricism and modeling.  相似文献
