Induction of chromosome banding by trypsin/EDTA for gene mapping by in situ hybridization 总被引:10,自引:0,他引:10
We describe an easy and reproducible procedure that utilizes trypsin/EDTA for the induction of chromosome banding in conjunction with in situ hybridization. The high quality banding resolution required for grain localization is obtained on both elongated and contracted chromosomes derived from synchronized or nonsynchronized human lymphocytes or fibroblasts. Through the use of a cDNA probe, the human erbB-2 gene was localized by in situ hybridization of normal human chromosomes at 17q11-q21. In situ hybridization of chromosomes derived from fibroblasts carrying a constitutional 15;17t(q22.3;q11.21) translocation showed that the erbB-2 gene was relocated on the rearranged chromosome 15. These results as well as grain localization on prophase chromosomes locate the erbB-2 gene at 17q12-q21.32. This localization may facilitate the search for human malignancies with chromosome changes involving the erbB-2 gene. 相似文献

1. Immediately after hatching, the pectoral muscle has a low glycogen content, and exhibits very low glycogen and high glucose and oxygen consumptions. 2. The protein kinase C inhibitor H-7 (2-20 microM) inhibited dose-dependently the infectivity of the vesicular stomatitis virus on cultured human fibroblasts. Electron microscopy showed that H-7 inhibited the viral entry. H-7 also inhibited the infectivity of four other enveloped viruses, herpes simplex I, turkey herpes, vaccinia and Sindbis. Similar results were obtained using staurosporine (2.5 nM), tamoxifen (40 microM), phloretin (140 microM), or W-7 (40 microM). However, the infectivity of non-enveloped viruses (e.g. poliomyelitis I) was not inhibited by H-7. These results show that protein kinase C is critically involved in the infectivity of enveloped viruses, most probably at the level of viral entry (receptor-mediated endocytosis). 相似文献
