Surface wettability and microtopography can either enhance or deter larval settlement of many sessile marine organisms. This study quantifies the effect of these surface properties on the settlement of pediveligers of Mytilus galloprovincialis, using polymers spanning a range of wettability and microtextured polydimethylsiloxane (PDMS). Furthermore, the adhesion strength of settled pediveligers on microtextured PDMS surfaces was quantified using a flow chamber. Settlement was enhanced at the hydrophilic end of the wettability spectrum, where mean settlement on nylon reached 33.5 ± 13.1%. In contrast, mean settlement on the most hydrophobic polymer (PDMS) was 4.2 ± 3.2%. Microtopography had a much stronger effect compared to wettability, where 400 μm textured PDMS enhanced settlement above 90%. Settlement preferences were also positively correlated to adhesion strength at flow rates of 4 knots, with all initially settled pediveligers on smooth PDMS detaching, while 79.9 ± 5.7% of pediveligers remained on the 400 μm texture. 相似文献

Abstract Inelastic neutron scattering spectroscopy is used to investigate dynamic changes in lysozyme powder at two different low D,0 hydrations (0.07g D2,O/g protein and 0,20 g D2,O/g protein). In the higher hydration sample, the inelastic scattering between 0.8 and 4.0 cm?1 energy transfer is increased and the elastic scattering is decreased. The decreased elastic scattering suggests increased atomic amplitudes of motion and the increased 0.8 to 4.0 cm?1 scattering suggests increased motions in this frequency range. Comparison with normal mode models of lysozyme dynamics shows that the inelastic difference occurs in the frequency region predicted for the lowest frequency, largest amplitude, global modes of the molccule[M. Summary Chromate-reducing bacteria were isolated from the cooling water of an electricity generating station where reduction of chromate had caused blockage of pipes by precipitation of chromium(III) oxide. Isolates identified included the generaAlcaligenes, Vibrio, Bacillus, Micrococcus, Staphylococcus andCorynebacterium. Isolate VMC-2 with the highest chromate-reducing activity was tentatively identified asComanonas testosteroni. The concentration of added chromate (K2CrO4, 20 M)_decreased by 95% during 45 min incubation with whole cells of VMC-2. In comparison, two Fe(III)-reducing isolates,Vibrio metschnikovii andAeromonas hydrophila, from lake sediments, showed similarly high chromate-reducing activities, and were able to reduce 99% of added chromate (20 M) in 45 min. Moderate Cr(VI)-reducers included strains ofBacillus, Vibrio andCorynebacterium. Micrococcus andStaphylococcus did not reduce Cr(VI). Sulfate (0.5 and 1.0 mM) inhibited the reduction of chromate by VMC-2 suggesting competition between the two oxyanions. Chromate-reducing activity was located in the soluble fraction of this isolate. The intermediacy of Cr(V)_in the reduction of chromate was confirmed by EPR spectroscopy. The bactericidal activity of hypochlorite towards isolate VMC-2 was determined. 相似文献

Two regions in the gene coding for 16S rRNA in Rhodococcus equi were selected as species-specific primer sequences for the polymerase chain reaction (PCR). PCR using these primers was tested against 10 strains of R. equi (including the type strain) and gave positive results for all but was negative for all other tested species of Rhodococcus ; representatives of the most closely related genera and a number of other bacterial species. Abstract: Susceptibility to NMDA neurotoxicity peaks in the early postnatal period in rats. Although indirect evidence suggests that interleukin-1β is a mediator of NMDA neurotoxicity in perinatal rats, direct confirmation of NMDA-induced interleukin-1β production in the brain has not been reported previously. The primary goal of this study was to determine if intracerebral injection of a neurotoxic dose of NMDA stimulates interleukin-1β production acutely. We used a rat-specific interleukin-1β ELISA to quantify brain tissue homogenate interleukin-1β content, and an immunocytochemical assay with a monoclonal anti-rat interleukin-1β antibody to visualize its distribution. NMDA (10 nmol) was injected stereotaxically into 7-day-old rats, using coordinates that targeted the striatum and overlying dorsal hippocampus. Interleukin-1β concentrations were measured in samples from the injected and contralateral cerebral hemispheres 0–12 h later; in addition, the impact of treatment with the noncompetitive NMDA antagonist MK-801 on interleukin-1β production was assessed. We found marked increases in tissue content of interleukin-1β in the lesioned hemisphere; values peaked at 6 h post injection. Treatment with MK-801 (1 mg/kg) blocked NMDA-induced increases in interleukin-1β. Preliminary immunocytochemical analysis demonstrated high concentrations of interleukin-1β-immunoreactive cells in the lesioned hippocampus, and concurrent increases in interleukin-1β immunoreactivity diffusely in the ependyma at 6 h after NMDA administration. Variations in stomatal density and index: implications for palaeoclimatic reconstructions 总被引:12,自引:2,他引:10
The variation in stomatal characters in leaves from one Alnus glutinosa (L.) Gaertn. tree is analysed. Measurements were taken from over 70 sites on the abaxial surfaces of representative ‘sun’ and ‘shade’ leaves having the same insertion point. The mean values of stomatal density and index in the shade leaf were significantly lower (71 and 93%, respectively) than those for the sun leaf. Within leaves, up to 2.5-fold differences in stomatal density values were observed. Contour maps derived from the data reveal non-random trends over the leaf surface. Correlations between stomatal density, epidermal cell density and stomatal index indicate that the variation in stomatal density within leaves arose primarily from local differences in stomatal differentiation, rather than from local differences in leaf expansion. This research demonstrates that a high level of variation in stomatal characters occurs both within and between leaves. We conclude that a well-defined sampling strategy should be used when estimating stomatal characters for (tree) leaves. Furthermore, the leaf's insertion point and situation within the tree crown should be taken into account. We discuss the implications of these findings for palaeoclimatic interpretations and emphasize the need for great caution when drawing conclusions based solely on stomatal characters. 相似文献

Bacterial flavohaemoglobins: a consensus sequence and identification of a discrete enterobacterial group and of further bacterial globins 总被引:1,自引:0,他引:1
The amino acid sequences of haemoglobin-like proteins from the bacteria Alcaligenes eutrophus , Bacillus subtilis , Erwinia chrysanthemi , Escherichia coli , Vibrio parahaemolyticus , Vitreoscilla sp. and the yeast Saccharomyces cerevisiae were studied. Phylogenies based on distance and parsimony analysis showed that the eubacterial group can be easily distinguished from the other haemoglobin-like proteins. The construction of a consensus bacterial flavohaemoglobin based on the alignment of six bacterial and one yeast globins allowed the design of consensus primers to search for haemoglobin-like genes in other bacteria. PCR products of the expected size were found in Campylobacter jejuni , Salmonella typhimurium , Listeria monocytogenes , Rhizobium leguminosarum , Klebsiella pneumoniae , Pseudomonas aeruginosa and Staphylococcus aureus . 相似文献

To utilize the rat spinotrapezius muscle as a model to investigate the microcirculatory consequences of exercise training, it is necessary to design an exercise protocol that recruits this muscle. There is evidence that the spinotrapezius is derecruited during standard treadmill exercise protocols performed on the uphill treadmill (i.e., 6 degrees incline). This investigation tested the hypothesis that downhill running would effectively recruit the spinotrapezius muscle as assessed by the presence of an exercise hyperemia response. We used radioactive 15-microm microspheres to determine blood flows in the spinotrapezius and selected hindlimb muscles of female Sprague-Dawley rats at rest and during downhill (i.e., -14 degrees incline; 331 +/- 5 g body wt, n = 7) and level (i.e., 0 degrees incline; 320 +/- 11 g body wt, n = 5) running at 30 m/min. Both level and downhill exercise increased blood flow to all hindlimb muscles (P < 0.01). However, in marked contrast to the absence of a hyperemic response to level running, blood flow to the spinotrapezius muscle increased from 26 +/- 6 ml.min(-1).100 g(-1) at rest to 69 +/- 8 ml.min(-1).100 g(-1) during downhill running (P < 0.01). These findings indicate that downhill running represents an exercise paradigm that recruits the spinotrapezius muscle and thereby constitutes a tenable physiological model for investigating the adaptations induced by exercise training (i.e., the mechanisms of altered microcirculatory control by transmission light microscopy). 相似文献
