The placing of animal behaviour in an evolutionary context is one of the great achievements of biologists in the last century. Life history theory has been a powerful tool in explaining both adaptation and constraint in phenotypic evolution, but rarely addresses the mechanistic bases of the traits it discusses. Recent advances in molecular biology have begun to uncover these mechanisms, and provide a challenge to the traditional view that life history trade-offs are the result of the differential allocation of limiting resources. In particular, costs of reproduction in Caenorhabditis elegans appear to arise from molecular signals, which have been claimed to be arbitrary with respect to fitness. We review the evidence that costs of reproduction in C. elegans are not resource based, and find that this is not necessarily the case. However, we welcome the challenge to traditional thinking, and suggest that integrating an understanding of mechanisms into life history theory will be one of the most exciting tasks facing evolutionary biologists in the 21st century. Abstract. Floristic composition of 14 mesas (i.e. flat‐top mountains) in four different study areas was investigated in southern African Nama Karoo. The study areas were arranged along a latitude gradient. Two simple questions were asked: are mesas potential conservation islands (a) in terms of re‐colonization potential and (b) in terms of species richness? Detailed vegetation surveys along a transect from the plains to the top of each mesa were summarized to obtain species composition for the three main habitats: plain, slopes and plateau. Floristic similarities between plains and mesa habitats were used to answer question (a), and species richness was used to answer question (b). Geographic position, high within‐area variability and elevation were found to largely influence the findings, resulting in only few consistent trends. The main findings of this study were, however: (1) mesas can act as sources for re‐colonization as well as havens for species adapted to mountain habitats. (2) Considering total species richness, mesa habitats were richer in species than plains in the northern, but not the southernmost study area. (3) Scaled by area, mesa plateaus emerged as the most species‐rich in all study areas. (4) Smaller and medium‐sized mesas showed greater re‐colonization potential, i.e. shared more species with their surrounding plains. (5) Medium‐sized to larger mesas supported plant communities more distinct from the surrounding plains. (6) A cut‐off point of approximately 240 m above the surrounding plain was indicated for the shift from ‘re‐colonization’ to ‘remnant’ function. The effects of acetaldehyde concentration (0, 0.025, 0.05, 0.1, 0.2,0.3, 0.4 or 0.5%) in solution on the vase life and ethylene productionoftwo carnation (Dianthus caryophyllus L.) cultivarscontrasting in vase life (Sandrosa (long lasting) and Yellow Candy (shortlasting)) were studied. In Sandrosa, acetaldehyde solution (0.05%)increased the vase life by 3 days in the absence, or by 2 days in the presence,of exogenous ethylene; the increase in vase life was associated with a delay inthe ethylene climacteric peak. In Yellow Candy, acetaldehyde (0.05%)increased vase life by 3 days, in an ethylene free environment. In the presenceof exogenous ethylene there was no increase in vase life of Yellow Candy.Acetaldehyde failed to inhibit ethylene production in either of the cultivars.It is suggested that acetaldehyde is not involved in the inhibition of ethylenebiosynthesis or sensitivity to ethylene in these two carnation cultivars. 相似文献

Stainless steel coupons were treated with skim milk and subsequently challenged with individual bacterial suspensions of Staphylococcus aureus, Pseudomonas fragi, Escherichia coli, Listeria monocytogenes, and Serratia marcescens. The numbers of attached bacteria were determined by direct epifluorescence microscopy and compared with the attachment levels on clean stainless steel with two different surface finishes. Skim milk was found to reduce adhesion of S. aureus, L. monocytogenes, and S. marcescens. P. fragi and E. coli attached in very small numbers to the clear surfaces, making the effect of any adsorbed protein layer difficult to assess. Individual milk proteins α-casein, β-casein, κ-casein, and α-lactalbumin were also found to reduce the adhesion of S. aureus and L. monocytogenes. The adhesion of bacteria to samples treated with milk dilutions up to 0.001% was investigated. X-ray photoelectron spectroscopy was used to determine the proportion of nitrogen in the adsorbed films. Attached bacterial numbers were inversely related to the relative atomic percentage of nitrogen on the surface. A comparison of two types of stainless steel surface, a 2B and a no. 8 mirror finish, indicated that the difference in these levels of surface roughness did not greatly affect bacterial attachment, and reduction in adhesion to a milk-treated surface was still observed. Cross-linking of adsorbed proteins partially reversed the inhibition of bacterial attachment, indicating that protein chain mobility and steric exclusion may be important in this phenomenon. 相似文献

The mechanism for inducing leucocytosis (increase in white blood cells) and thrombocytosis (increase in platelets) during exercise is unclear. Because plasma osmolality (Osm) may influence T-cell proliferation, Osm and the number of leucocytes (WBC) and platelets in blood were measured periodically during a 90 min rest period, and were compared with those during upright sitting ergometer exercise in six untrained, healthy men who cycled for 70 min at 71% of their maximal oxygen uptake (VO2max). There were 6 experiments in which the subjects drank different fluid formulations (10 ml x kg(-1) of various ionic and osmotic concentrations intermittently during 60 min of the rest period and during the exercise period. Osmolality, and WBC and platelet counts increased significantly (p < 0.05) within the first 10 min of exercise, but the additional 60 min of exercise did not significantly change the leucocytosis or thrombocytosis. There were low but significant correlations between individual values of total WBC and total Osm during exercise (r0.001(2),284 = 0.39) and during rest plus exercise (r0.001(2),499 = 0.43). With combined data from the six experiments, mean Osm correlated highly and significantly with both mean WBC (r0.001(2),6 = 0.95, p < 0.001) and mean platelets (r0.001(2),6 = 0.94, p < 0.01) during the exercise phase. These data indicate that increases in leucocytes, thrombocytes, and osmolality occur primarily within the first 10 min of high-intensity exercise, but neither hypovolemia nor hyperthermia during exercise contributed to the leucocytosis, thrombocytosis, or hyperosmolality. The high correlations between plasma Osm and WBC or platelet counts suggest changes in osmolality may contribute to the mechanism of leucocytosis and thrombocytosis induced by exercise. 相似文献

The 2.1 A resolution crystal structure of flavin reductase P with the inhibitor nicotinamide adenine dinucleotide (NAD) bound in the active site has been determined. NAD adopts a novel, folded conformation in which the nicotinamide and adenine rings stack in parallel with an inter-ring distance of 3.6 A. The pyrophosphate binds next to the flavin cofactor isoalloxazine, while the stacked nicotinamide/adenine moiety faces away from the flavin. The observed NAD conformation is quite different from the extended conformations observed in other enzyme/NAD(P) structures; however, it resembles the conformation proposed for NAD in solution. The flavin reductase P/NAD structure provides new information about the conformational diversity of NAD, which is important for understanding catalysis. This structure offers the first crystallographic evidence of a folded NAD with ring stacking, and it is the first enzyme structure containing an FMN cofactor interacting with NAD(P). Analysis of the structure suggests a possible dynamic mechanism underlying NADPH substrate specificity and product release that involves unfolding and folding of NADP(H). 相似文献
