The relationship between protein synthesis and the incorporation of [3H]gibberellin A1 ([3H]GA1) into a 2,000xg pelletable (2KP) fraction from lettuce (Lactuca sativa L.) hypocotyl sections has been investigated. Concentrations of D-2-(4-methyl-2,6-dinitroanilino)-N-methylpropionamide (MDMP) between 10-7 M and 10-4 M caused increasing inhibition of growth, 2KP labelling and incorporation of [14C]leucine into soluble protein. Growth and 2KP radioactivity were highly correlated (r=0.996). Transfer to MDMP early or late in the course of GA response caused reductions in both growth and incorporation into the 2KP fraction. Exposure to the inhibitor had more effect at 4 h than at 20 h. The proportions of alkali-soluble and insoluble radioactivity in the 2KP fraction were also altered by this treatment. Summary The distribution of N-linked glycans in rat testis has been probed using a panel of lectins derived fromGalanthus nivalis (snowdrop, GNA),Canavalia ensiformis (jack bean, Con A),Lens culinaris (lentil, LCA),Pisum sativum (garden pea, PSA) andPhaseolus vulgaris, erythro- and leucoagglutinins (kidney bean, ePHA and IPHA). Several classes of N-linked glycan were identified in the spermatogenic series, and during differentiation into spermatozoa they altered in both their pattern of distribution and relative abundance. A population of tetra-antennary, non-bisected, complex glycans, detected by IPHA, was lost during the transition from spermatogonia to spermatocytes, while high-mannose structures were acquired; these were most abundant in spermatocytes, as were bi- and tri-antennary complex, non-bisected glycans, the latter becoming increasingly abundant on acrosomes and spermatozoa. Their bisected counterparts were more generally expressed throughout spermatogenic cells, although marked localization onto acrosomes and nuclear caps was again seen. Transition from spermatocytes to spermatids involved mainly changes of the acrosomal granule and nuclear cap, which were carried through to the final stages of differentiation. Sertoli cell surfaces and cytoplasmic granules showed a high level of N-glycan expression.  相似文献

Summary Rat testes have been examined with a panel of lectins that bind specifically to oligosaccharide sequences having terminal or subterminal -galactosyl residues in O-linked glycans, or in the outer chains of complex N-linked glycans:Arachis hypogaea (peanut, AHA),Erythrina cristagalli (coral tree, ECA),Ricinus communis (castor bean, RCA120) andAbrus precatorius (jequirity bean, APA) agglutinins. Pretreatment of sections with neuraminidase, -galactosidase and removal of alkali-labile O-linked sequences by -elimination allowed the structure of these glycans to be further explored. In spermatogonia and spermatocytes there was little evidence of glycans terminating in -galactosyl residues, although these were present at non-reducing terminals as sialylgalactosides. The acrosome contained two subsets of O-linked glycans terminating in sialylgalactosides, while the nuclear cap showed at least two subsets of N-linked sialylgalactosyl as well as O-linked glycans. Spermatozoa exhibited minor changes in the pattern of glycosylation, although the overall pattern of -galactosyl expression was similar. Binding to Sertoli cells showed the presence of some unsubstituted -galactosyl terminals on O-linked glycans but few such N-linked residues, while terminal -galactosides were scanty in tubular basement membranes.  相似文献

The binding of [3H]diazepam and [3H]3-carboethoxy-beta-carboline was examined in rat brain synaptosomal membranes treated with irazepine, an alkylating benzodiazepine. Under incubation conditions that resulted in a 25-33% reduction in the Bmax of [3H]diazepam binding, only modest (less than 8.5%) reductions in the Bmax of [3H]3-carboethoxy-beta-carboline were observed. The differential effects of irazepine on the binding of these two compounds may be explained by the presence of multiple areas or "domains" on the benzodiazepine receptor.  相似文献

Chlorophyll levels in l-cm sections of the youngest fully expanded leaves of normal (Y) Festuca pratensis L. declined almost to zero over a period of 6 days after excision. Chlorophyll in a mutant genotype (NY) remained near the initial level for the whole of this period. Abscisic acid promoted pigment loss in Y but had no significant effect on chlorophyll in NY. Kinetin retarded pigment loss in Y but was ineffective in NY. Other biochemical changes associated with leaf senescence—reduction in protein content and the appearance of novel isoenzymes of α-naphthyl acetate esterases—occurred in both genotypes. Abscisic acid accelerated protein breakdown, whereas kinetin inhibited the loss of protein in both genotypes. The mutation thus appears to be expressed as a highly specific lesion in pigment metabolism. We concluded that pigment breakdown, which is widely used as an index of leaf senescence, may not be an inevitable part of the aging process.  相似文献

A method of analysis is proposed based on the enzyme-catalysed transfer of tritium from [3H]NADH to suitable substrates. Its practicability is demonstrated on the examples of oestrone and progesterone with the respective use of the 3 beta, 17 beta- and 3 alpha, 20 beta-hydroxysteroid oxidoreductase. Specificity is tested by application to the analysis of the plasma of pregnant women and measurement of the 3H/14C ratios on purification of the enzymic reduction products.  相似文献

Wild house mice, Mus musculus L., were trapped, and their descendants reared, in permanently mated pairs, for a number of generations in two laboratory environments, at about 21°C (controls) and -3°C, respectively. All mice had sawdust and cottonwool for bedding; but the nests of those at -3°C were colder than those in the warm, and fluctuated greatly in temperature.
Reproductive performance was inferior in the cold environment: more pairs were barren, and the fecund pairs reared fewer young than the controls. Yet litters at birth were usually larger in the cold, and the young at three weeks were always heavier. Over ten generations nestling mortality declined at -3°C.
From generation 1 on, adult mice at -3°C were heavier than the controls, but there was no corresponding increase in body length. Tails were much shorter relative to body length in the first generations in the cold, but returned to control proportions by generation 10. Most of the structural changes in the cold accord with the "rules" of Bergmann and Allen.
The incidence of abnormal sixth lumbar vertebrae was low in all generations at both temperatures.
After nine generations, some mice were transferred from the cold to the warm environment, and bred for a further three generations. There they outstripped the controls both in reproductive performance and in growth. They also had more fat, and a heavier and longer small intestine; but the heart, stomach and kidneys were lighter than those of the controls. Adrenal weights at 21°C declined over the generations, but those of the mice at -3°C did not.
The secular changes observed, especially those in the cold environment, are attributed principally to differential selection of genotypes, not to inbreeding; but maternal effects may also have been involved.  相似文献
