A simple and convenient method for the one-pot three-component synthesis of 3-pyranyl indoles has been accomplished by tandem Knoevenagel–Michael reaction of 3-cyanoacetyl indole, various aromatic aldehydes and malononitrile catalyzed by InCl3 in ethanol under reflux conditions. The newly synthesized 3-pyranyl indoles were evaluated for anti-microbial, antioxidant, and anticancer activities. Some of the compounds showed good anticancer activity against MCF-7 breast cancer cell lines on comparison with of standard drug.  相似文献

A deficiency in major seed storage proteins is associated with a nearly two-fold increase in sulfur amino acid content in genetically related lines of common bean (Phaseolus vulgaris). Their mature seed proteome was compared by an approach combining label-free quantification by spectral counting, 2-DE, and analysis of selective extracts. Lack of phaseolin, phytohemagglutinin and arcelin was mainly compensated by increases in legumin, α-amylase inhibitors and mannose lectin FRIL. Along with legumin, albumin-2, defensin and albumin-1 were major contributors to the elevated sulfur amino acid content. Coordinate induction of granule-bound starch synthase I, starch synthase II-2 and starch branching enzyme were associated with minor alteration of starch composition, whereas increased levels of UDP-glucose 4-epimerase were correlated with a 30% increase in raffinose content. Induction of cell division cycle protein 48 and ubiquitin suggested enhanced ER-associated degradation. This was not associated with a classical unfolded protein response as the levels of ER HSC70-cognate binding protein were actually reduced in the mutant. Repression of rab1 GTPase was consistent with decreased traffic through the secretory pathway. Collectively, these results have implications for the nutritional quality of common bean, and provide information on the pleiotropic phenotype associated with storage protein deficiency in a dicotyledonous seed.  相似文献

Field, greenhouse, and growth chamber experiments were conducted to determine the effects of different burial treatments on photosynthesis (carbon dioxide exchange rate), chlorophyll-a fluorescence, leaf area, biomass, leaf thickness, total chlorophyll content and chlorophyll a/b ratio of ten sand dune species: Agropyron psammophilum, Cakile edentula, Cirsium pitcheri, Corispermum hyssopifolium, Elymus canadensis, Oenothera biennis, Panicum virgatum, Strophostyles helvola, Tusilago farfara, and Xanthium strumarium. Although there were significant differences between species, all of them exhibited stimulation in growth following burial in sand. Generally, buried plants of these species showed an increase in biomass, photosynthetic efficiency, and chlorophyll-a fluorescence because of higher energy content in their roots, rhizomes, and underground stems. The main reasons for the stimulation in growth were an increase in leaf area, leaf thickness, and root biomass. The total chlorophyll content of leaves of buried plants of A. psammophilum, E. canadensis, and P. virgatum was higher than controls, but there were no significant differences for Cirsium pitcheri, O. biennis, and S. helvola. The similarities and differences exhibited by the test species in their responses to burial would be ecologically adaptive to survive the harsh environmental conditions of foredunes. All species showed a clear compensatory response following recovery from the burial episode and surpassed control by enhancing the vital physiological, morphological and growth functions.  相似文献

The interaction between stromal and epithelial cells is critical for the initiation and progression of prostate cancer, but the molecular determinants responsible for the cross-talk between these two cell types remain largely unknown. Here, we used a co-culture cell assay to identify messengers involved in the cross-talk between human prostate stromal PS30 and epithelial LNCaP cells. Stimulation with lysophosphatidic acid (LPA) activates the mitogenic ERK signaling pathway in PS30, but not LNCaP, cells. The co-culture of PS30 and LNCaP cells results in the activation of ERK in LNCaP cells and that is further increased in response to stimulation with LPA. Physiologic relevance of the interaction between PS30 and LNCaP cells is demonstrated using LNCaP xenograft tumor assays. Animals implanted with a mixture of both cell types develop larger tumors with higher frequency compared with those injected with LNCaP cells alone. Conditioned medium transfer experiments reveal the PS30-derived inducing factor is soluble and promotes mitogenic ERK and STAT3 signaling pathways in LNCaP cells. Protein analysis demonstrates that treatment of the PS30 cells with LPA induces synthesis of interleukin 6 (IL-6). Antibody neutralization experiments reveal that IL-6 is responsible for the LPA-induced mitogenic signaling and growth of the LNCaP cells. Our findings reveal that the LPA-regulated secretion of IL-6 is an important messenger linking stromal and epithelial prostate cells, which may be exploited for the effective treatment of patients with advanced prostate cancer.  相似文献

A series of novel 2-(4-substituted piperazin-1-yl)-1,8-naphthyridine-3-carbonitrile 6 were prepared by microwave irradiation and conventional heating. The intermediate, 2-oxo-1,2-dihydro-1,8-naphthyridine-3-carbonitrile 3, was prepared from 2-aminonicotinaldehyde 1 and ethyl cyanoacetate 2 in the presence of piperidine under solvent free condition. The synthesized compounds were evaluated for 5-HT3 antagonisms in longitudinal muscle-myenteric plexus (LMMP) preparation from Guinea pig ileum against 5-HT3 agonist, 2-methyl-5-HT. Among the compounds tested, 2-(4-allylpiperazin-1-yl)-1,8-naphthyridine-3-carbonitrile 6d showed most favorable 5-HT3 receptor antagonism in the Guinea pig ileum.  相似文献

Parasitic disease in fishes is one of the most important factors limiting aquaculture production and its economic viability. Cymothoa indica, a cymothoid isopod, is reported here for the first time parasitizing cultured larvae of the Asian seabass Lates calcarifer in India. Fourteen-day-old L. calcarifer larvae of mean weight 8.73 +/- 0.03 mg were fed with wild zooplankton in the laboratory. On Day 14 of rearing, larvae were found parasitized by cymothoids. Infected larvae reached a mean (+/- SE) weight of 98.86 +/- 0.30 mg, while uninfected specimens weighed 117 +/- 0.43 mg at the end of the experiment (Day 21). C. indica occurred in the branchial and anterodorsal regions of infected fish, where resultant skin lesions were red, hemorrhagic, without scales and with abundant secreted mucus. The cumulative mortality over the 3 wk period was 16.54 %. These parasites are transferred to the host via the zooplankton used as food; this could easily be overcome, either by filtering wild zooplankton to remove the infectious swimming larvae of C. indica or by using cultured copepods.  相似文献
