A 19 kDa soluble protein was purified from human nasal mucus. Its N-terminal amino-acid sequence appeared to be identical to that of a lipocalin synthesised both in lachrymal glands and in von Ebner's glands (VEG) of circumvallate papillae. In order to verify whether this protein was synthesised in the nasal cavity or was the result of tear contamination, we adopted an immunohistochemical approach. Polyclonal antibodies, raised against a primate VEG protein, were used on sections of human nasal mucosa obtained from surgery. The results clearly indicate that the protein is synthesised in sero-mucous glands underlying the respiratory ciliated epithelium. Although ligand-binding experiments with some odorant molecules have given negative results, we cannot exclude a role of odorant solubiliser and carrier for this protein.  相似文献

Visual pigments are a class of receptor proteins that absorb light and trigger sensory signals. Retinal-containing proteins are used in nature as photoreceptors mainly in animals vision. Mammalian rhodopsin is the best studied example of a light sensor which couples photon absorption to a cascade of biochemical reactions amplifying the input signal. A surprising discovery was to find rhodopsin also in Archaebacteria and in unicellular eukaryotes. On the basis of absorption microspectroscopic measurements and of inhibition experiments on pigment biosynthetic pathways, we have recently suggested that a rhodopsin could be the functional receptor of the visual process in Euglena gracilis, a flagellate which can use light directly to promote photosynthetic reactions, or as an incident flux of information to adjust its swimming orientation. We here report purification and identification of all-trans-retinal by column chromatography, HPLC and GC-MS in E. gracilis; these findings indicate with absolute certainty that rhodopsin is the photoreceptor molecule of this microorganism.  相似文献

Cultivation of microalgae in closed photobioreactors suffers from high temperatures, whereas in open raceway ponds the optimal temperature is seldom reached. With an integrated device coupling a raceway pond with a suitably sized and positioned closed system (alveolar panel), the heat accumulated in the panel was efficiently transferred to the pond and a near-optimum temperature regimen for Arthrospira platensis was maintained, with no need for any additional cooling device. The productivity obtained in the integrated system was higher than the sum of the productivities (g reactor−1 day−1) of the pond and panel systems operating separately. The effect of diets supplemented with three different fats (olive oil, sunflower oil, pork fat) on the susceptibility of the rat heart to oxidative stress and on the rate of eicosanoid release were studied. Our results show that when fatty-acid unsaturation of heart lipids is increased or vitamin E is decreased, even to a low degree, a marked enhancement of the susceptibility to hydroperoxide-induced oxidative stress (measured by chemiluminescence emission) occurs, which is associated with an increase of eicosanoid release from the heart.  相似文献

1. Two soluble proteins, with good affinity to tritiated 2-isobutyl-3-methoxypyrazine, have been purified from mouse nasal mucosa. 2. The first protein is a heterodimer with subunits of apparent M(r) 18 and 19 kDa and isoelectric point of 4.9; the second is a monomer of M(r) 21 kDa and isoelectric point of 4.8. 3. Three new ligands for odorant-binding proteins (OBPs) have been prepared, to be used in protein structure studies: the photoaffinity label 2-azido-4-butyl-5-propylthiazole, the NMR probe 2-trifluoromethyl-4-butyl-5-propylthiazole and the X-ray probe 2-amino-4-butyl-5-propylselenazole. All three compounds bind the bovine OBP with dissociation constants in the micromolar range.  相似文献

Human stretch reflexes (SRs) are often too weak and ineffectual to provide adequate postural regulation or rhythmic movement boosting (e.g. in ankle pushoff at the end of stance phase in fast running). Recent improvements in methods of artificially enhancing skeletomotor responses, especially in therapeutic regimens, should not be widely employed until the clonus-resisting stability properties of SRs are better understood. We formulate an idealized linear servo model of a segmentally-mediated SR system which includes the often ignored electromechanical coupling delay. For typical closed-loop (delay/gain) ratios, the model is shown to be unstable for all values of loop gain when operating as a position servo, but maximally stable when operating as a velocity servo. We claim that the velocity servo or more of its nonlinear relatives is a better model for some well studied SRs than, e.g., Houk's stiff muscle hypothesis. We also present evidence that even feeble and quickly saturating monosynaptic postural servos are always unstable if operated as pure position regulators.  相似文献
