Electrophoretically homogenous preparations of malate dehydrogenase (MDH) isoforms of the bacteria Sphaerotilus natans D-507 with specific activity 7.46 U/mg and 5.74 U/mg with respect to protein concentration have been obtained. The dimeric
isoform of the enzyme was shown to function under organotrophic growth conditions, whereas the tetrameric isoform was induced
under mixotrophic cultivation conditions. PCR-analysis revealed a single gene encoding the malate dehydrogenase molecule.
The topography of the MDH isoform surface was studied by atomic-force microscopy, and a 3D-structure of the enzyme was obtained.
Spectraphotometric analysis data allowed us to suggest that stabilization of the tetrameric form of MDH is due to additional

The Krebs cycle is one of the major metabolic pathways in a cell, which includes both catabolic and anabolic reactions. The first enzyme of the Krebs cycle, citrate synthase, catalyzes one of a few irreversible reactions of the cycle, citrate formation from acetyl-CoA and oxaloacetate. Expression of the CIT1 gene encoding the mitochondrial form of this enzyme inSaccharomyces cerevisiae is repressed on glucose- and glutamate-containing medium and activated on the raffinose-containing medium. In this work, the dependence of glucose repression of the CIT1 gene on the content of phosphate in the medium was studied. On the phosphate-deficient medium, the level of the CIT1 gene expression was increased twice. A low-molecular-weight (about 34 kDa) protein was identified and shown to interact with a region of the CIT1gene promoter (from –367 to –348 bp), which controls the glucose repression. The results obtained suggest that the Pho4 protein is involved in regulation of the CIT1gene expression on the glucose-containing and phosphate-deficient medium. Disruption of the PHO85 gene encoding phosphoprotein kinase (Pho4p is the substrate of this enzyme) leads to alleviation of glucose repression of the CIT1 gene. Thus, in yeast cells grown in the presence of glucose, the PHO85gene mediates downregulation of theCIT1expression. 相似文献

The efficiency of neuston sampling from freshwater Lake Baikal with Garrett’s metal screen and polycarbonate membrane filters has been estimated. It is revealed that both sampling methods demonstrate similar results. Method selection is determined by climatic and hydrological factors. The results of the present work demonstrate that sampling using a metal screen can be recommended for the complex characterization of neuston biofilm and estimation not only microbiological, but also molecular and chemical parameters, because it allows collecting the water volume required for all analyses. 相似文献

Serum samples from 641 workers of large poultry and meat-packing plants were studied in the passive hemagglutination test with the use of Salmonella complex and serogroup diagnostica. A specific increase in the level of anti-Salmonella antibodies in 60.7% of poultry plant workers and in 9.8% of meat-packing plant workers was established. Among the workers of the poultry plants the most pronounced immune shifts were detected in persons having contacts with sick poultry and pathological material and among the employees of the meat-packing plant, in those who ate raw sausage meat. A high level of antibodies in the professional groups under study was observed as early as in the first year of work at the plant and persisted over the whole period of this work. Under the conditions of constant contamination of the workers of poultry and meat-packing plants with small doses of salmonellae specific immunity to this infection was seemingly induced, which inhibited the development of the manifest forms of infection, but did not prevent the formation of chronic carrier state. 相似文献

The immunological activity of specific immunoglobulins in different forms of cholera infection was studied with the use of the indirect immunofluorescent method. The forms of cholera infection were shown to be related to the physico-chemical spectrum of antibodies: patients with the pronounced clinical symptoms of cholera produced serum IgM, including antigen -- antibody complex, more actively and in a greater amount; patients with faintly pronounced and asymptomatic infection produce highly active IgG. The competitive action of immunoglobulins of different classes in their simultaneous reaction with O antigen of Vibrio cholerae is shown. The mechanism of the competitive action of different immunoglobulin classes is supposed to play some role in the process of the sanation (with highly active IgM) or detoxication (with highly active IgG) of the organism. 相似文献
