Repeated sequences cloned from the DNA of the sea urchin S. purpuratus were used as probes to measure the lengths of individual families of repeats. Some probes reassociated much more rapidly with preparations of long repeats than with short repeats while others reassociated more rapidly with short repeats than with long repeats. In this way two of five cloned repeats were shown to represent families with a great majority of sequences in the long class. One represented a family with similar numbers of long and short class members. Two were members of predominantly short class families. — The cloned repeats representing long class families, formed more precise duplexes than those representing short class families. Thermal stability measurements using S. purpuratus or S. franciscanus driver DNA showed that precise repetitive sequences have as great an interspecies sequence difference as the less precise repeats. Thus the precision of many families may result from recent multiplication rather than from selective pressure on the DNA sequences. Measurements of evolutionary frequency change show a clear correlation between the frequency change and the size of families of repeats in S. purpuratus. Comparison with S. franciscanus indicates that many of the large size families in S. purpuratus are those that have grown in size since these two species diverged.  相似文献

Energy flow (A = R + P; assimilation = respiration + production) and life-history/density-biomass cycles were followed for 1 year in a mayfly (Brachycercus sp.) population and two chironomid (Procladius sp. and Tendipes decorus) populations in a 0.94 ha pond located in north-central Texas. Temperature (5, 10, 15, 20, 25 and 30°C) effects on R and patterns of metabolic compensation/acclimatization were determined for the mayfly and chironomids. Chironomid and mayfly populations dominated the nine benthic macroinvertebrates identified in the pond, with greatest abundances in winter and winter-spring respectively. Annual energy flow was 51 kcal m–2 (R = 40 kcal, P = 11 kcal or 1.9 g) in Brachycercus sp., 23 kcal m–2 (R = 5 kcal, P = 18 kcal or 2.4 g) in Procladius sp. and 74 kcal m–2 (R = 17 kcal, P = 57 kcal or 6.0 g) in T. decorus. Each species metabolically compensated partially to temperatures en countered during their life histories.  相似文献

The glycoprotein of pig gastric mucus has been isolated free of non-covalently bound protein as judged by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis and equilibrium density-gradient centrifugation. After reduction with 0.2 M-mercaptoethanol, protein was released from the glycoprotein, which consisted of a major 70000-mol.wt. component and a minor 60000-mol.wt. component. The 70000-mol.wt. protein fraction was separated from the reduced glycoprotein by either density-gradient centrifugation in CsCl or by gel filtration. Analysis of the 70000-mol.wt. protein fraction showed that, within the limits of the analysis, it was non-glycosylated, and its amino acid analysis was quite different from that of the reduced glycoprotein, which is high in serine, threonine and proline. There was a ratio of one 70000-mol.wt. protein per native glycoprotein molecule of 2 X 10(6) mol.wt. Dissociation of the native glycoprotein into glycoprotein subunits (5 X 10(5) mol.wt.) by reduction or proteolysis results in the release or hydrolysis respectively of the 70000-mol.wt. protein. A similar 70000-mol.wt. protein is demonstrated in human gastric mucus glycoprotein. A structural role for the proteins in these mucus glycoproteins is proposed.  相似文献

The Large-billed Reed Warbler Acrocephalus orinus is known only from the type specimen, collected in Himachal Pradesh, India, in 1867. The specimen is poorly prepared, and it has been suggested that it could represent an isolated form of the Clamorous Reed Warbler A. stentoreus or an aberrant Blyth's Reed Warbler A. dumetorum . We tested the affinity of A. orinus by (1) re-examining the morphology of the type specimen and (2) amplifying and sequencing a portion of its mitochondrial cytochrome b gene. Both the morphological and the mitochondrial analyses showed the specimen to be similar to dumetorum , but distinct enough to qualify as a species of its own. Relative to dumetorum , it has a more rounded wing, longer bill, longer and more graduated tail with more pointed tail feathers, and larger claws. The divergence in mitochondrial DNA between orinus and dumetorum was 7.8%, well above the value expected between subspecies. A. orinus is smaller than any of the forms of A. stentoreus or the related Australian Reed Warbler A. australis . It has a somewhat longer first primary, more pointed tail feathers and paler, less robust feet and claws. DNA comparison places it in the clade of small unstreaked Acrocephalus warblers, and apart from the clade of large unstreaked warblers that contains stentoreus and australis .  相似文献

Fibrous long spacing collagen (FLS) fibrils are collagen fibrils in which the periodicity is clearly greater than the 67-nm periodicity of native collagen. FLS fibrils were formed in vitro by the addition of alpha1-acid glycoprotein to an acidified solution of monomeric collagen and were imaged with atomic force microscopy. The fibrils formed were typically approximately 150 nm in diameter and had a distinct banding pattern with a 250-nm periodicity. At higher resolution, the mature FLS fibrils showed ultrastructure, both on the bands and in the interband region, which appears as protofibrils aligned along the main fibril axis. The alignment of protofibrils produced grooves along the main fibril, which were 2 nm deep and 20 nm in width. Examination of the tips of FLS fibrils suggests that they grow via the merging of protofibrils to the tip, followed by the entanglement and, ultimately, the tight packing of protofibrils. A comparison is made with native collagen in terms of structure and mechanism of assembly.  相似文献

The loss of Bacillus subtilis penicillin-binding protein (PBP) 2a, encoded by pbpA, was previously shown to slow spore outgrowth and result in an increased diameter of the outgrowing spore. Further analyses to define the defect in pbpA spore outgrowth have shown that (i) outgrowing pbpA spores exhibited only a slight defect in the rate of peptidoglycan (PG) synthesis compared to wild-type spores, but PG turnover was significantly slowed during outgrowth of pbpA spores; (ii) there was no difference in the location of PG synthesis in outgrowing wild-type and pbpA spores once cell elongation had been initiated; (iii) outgrowth and elongation of pbpA spores were dramatically affected by the levels of monovalent or divalent cations in the medium; (iv) there was a partial redundancy of function between PBP2a and PBP1 or -4 during spore outgrowth; and (v) there was no difference in the structure of PG from outgrowing wild-type spores or spores lacking PBP2a or PBP2a and -4; but also (vi) PG from outgrowing spores lacking PBP1 and -2a had transiently decreased cross-linking compared to PG from outgrowing wild-type spores, possibly due to the loss of transpeptidase activity.  相似文献

Amino acid deprivation induces adaptive changes in amino acid transport and the intracellular amino acid pool in cultured cells. In this study intracellular amino acid levels were determined in cultured bovine aortic endothelial cells (EC) deprived of L-arginine or total amino acids for 1, 3, 6 and 24 h. Amino acid concentrations were analyzed by reverse phase HPLC after precolumn derivatisation. Under normal culture conditions levels of L-arginine L-citrulline, total essential and non-essential amino acids were 840 +/- 90 microM, 150 +/- 40 microM, 11.4 +/- 0.9 mM and 53.3 +/- 3.4 mM (n = 9), respectively. In EC deprived of L-arginine or all amino acids for 24 h L-arginine and L-citrulline levels were 200 microM and 50 microM, and 670 microM and 100 microM Deprivation of L-arginine or total amino acids induced rapid (1 h) decreases (30 - 50%) in the levels of other cationic (lysine, ornithine) and essential branched-chain (valine, isoleucine, leucine) and aromatic (phenylalanine, tryptophan) amino acids. L-glutamine was reduced markedly in EC deprived of total amino acids for 1 h - 6 h but actually increased 3-fold in EC deprived of L-arginine for 6 h or 24 h. Arginine deprivation resulted in a rapid decrease in the total intracellular amino acid pool, however concentrations were restored after 24 h. Increased amino acid transport and/or reduced protein synthesis may account for the restoration of amino acid levels in EC deprived of L-arginine. The sustained reduction in the free amino acid pool of EC deprived of all amino acids may reflect utilization of intracellular amino acids for protein synthesis.  相似文献

A single-chain antibody fragment has been constructed for an antibody that binds to the Chlamydia specific carbohydrate structure of the lipopolysaccharide. Single-chain protein was expressed and secreted into the periplasmic space of E. coli as a fusion protein with the maltose binding protein. The fusion protein was purified in one step by virtue of its ability to bind to maltose. In a sandwich ELISA, the eluted protein bound Chlamydia lipopolysaccharide, which demonstrates that the single-chain protein domain will function as part of a fusion protein. The expression of maltose binding fusion proteins into the periplasmic space could be used for production of other single-chain antibodies or protein fragments requiring appropriate folding and disulfide bond formation.  相似文献

We have increased the methionine content of the seed proteins of a commercial winter variety of canola by expressing a chimeric gene encoding a methionine-rich seed protein from Brazil nut in the seeds of transgenic plants. Transgenic canola seeds accumulate the heterologous methionine-rich protein at levels which range from 1.7% to 4.0% of the total seed protein and contain up to 33% more methionine. The precursor of the methionine-rich protein is processed correctly in the seeds, resulting in the appearance of the mature protein in the 2S protein fraction. The 2S methionine-rich protein accumulates in the transgenic seeds at the same time in development as the canola 11S seed proteins and disappears rapidly upon germination of the seed. The increase in methionine in the canola seed proteins should increase the value of canola meal which is used in animal feed formulations.  相似文献

Previous investigations have shown that the flight motor pattern of the mature locust (Locusta migratoria L.) relies heavily on the input of the hindwing tegulae. Removal of the hindwing tegulae results in an immediate change in the motor pattern: the wingbeat frequency (WBF) decreases and the interval between the activity of depressor and elevator muscles (D-E interval) increases. In contrast, removal of the forewing tegulae has little effect on the motor pattern. Here we report adaptive modifications in the flight system that occur after the removal of the hindwing tegulae. Over a period of about 2 weeks following hindwing tegula removal, the flight motor pattern progressively returned towards normal, and in about 80% of the animals recovery of the flight motor pattern was complete. We describe the changes in the activity pattern of flight muscles and in the patterns of depolarizations in flight motoneurons and flight interneurons associated with this recovery. In contrast to the situation in the intact animal, the activity of the forewing tegulae is necessary in recovered animals for the generation of the motor pattern. Removal of the forewing tegulae in recovered animals resulted in similar changes in the flight motor pattern as were observed in intact animals after the removal of the hindwing tegulae. Furthermore, electrical stimulation of forewing tegula afferents in recovered animals produced similar resetting effects on the motor pattern as electrical stimulation of the hindwing tegulae afferents in intact animals. From these observations we conclude that recovery is due to the functional replacement of the removed hindwing tegulae by input from the forewing tegulae.  相似文献
