Tylenchorhynchus dubius was observed to feed on ''Toronto'' creeping bentgrass and ''Merion'' Kentucky bluegrass, and was a key participant in reducing the vegetative growth of both grass species. The severity of foliar and root macrosymptoms incited by T. dubius to ''Toronto'' bentgrass was greater on plants grown at 16 C than on plants maintained at 21, 27 and 32 C. These parasitized plants exhibited a suppression of secondary stolon formation, shortened internodes and premature inflorescence initiation. Initial inoculum densities of 500 and 1000 nematodes/test pot produced similar pathogenic effects on the host. Feeding was primarily on root hairs and epidermal cells immediately behind the meristematic region. No necrotic lesions or other diagnostic symptoms were visible at the feeding sites. To provide a routine check for the presence of ferric iron in sections, Perls' method was combined with hematoxylin and eosin as follows. Deparaffinized sections of formalin-fixed tissues are stained in Perls' reagent (1:1 2%, w/v, of potassium ferrocyanide in distilled water and 2%, v/v, concentrated HCl in distilled water) for 20 min. After brief rinsing in distilled water stain sections in Mayer's hemalum, wash in tap water for 5 min, counterstain in 0.5% (w/v) eosin B in 50% ethyl alcohol for 15 sec. The hydraulic conductivity of the membranes surrounding the giant axon of the squid, Dosidicus gigas, was measured. In some axons the axoplasm was partially removed by suction. Perfusion was then established by insertion of a second pipette. In other axons the axoplasm was left intact and only one pipette was inserted. In both groups hydrostatic pressure was applied by means of a water column in a capillary manometer. Displacement of the meniscus in time gave the rate of fluid flowing across the axon sheath. In both groups osmotic differences across the membrane were established by the addition of a test molecule to the external medium which was seawater. The hydraulic conductivity determined by application of hydrostatic pressure was 10.6 ± 0.8.10-8 cm/sec cm H2O in perfused axons and 3.2 ± 0.6.10-8 cm/sec cm H2O in intact axons. When the driving force was an osmotic pressure gradient the conductivity was 4.5 ± 0.6 x 10-10 cm/sec cm H2O and 4.8 ± 0.9 x 10-10 cm/sec cm H2O in perfused and intact axons, respectively. A comparable result was found when the internal solution was made hyperosmotic. The fluid flow was a linear function of the hydrostatic pressure up to 70 cm of water. Glycerol outflux and membrane conductance were increased 1.6 and 1.1 times by the application of hydrostatic pressure. These increments do not give an explanation of the difference between the filtration coefficients. Other possible explanations are suggested and discussed. 相似文献

Plant defence mechanisms can reduce the digestive enzyme activity of insect pests. The aim of this study was to determine the relationship between the production of proteinase inhibitors, lipoxygenase and polyphenol oxidase activity in Coffea arabica (Catuai IAC 15) plants, and the digestive enzyme activity in the pest Leucoptera coffeella (Lepidoptera: Lyonetiidae) after feeding on the plant. The production of proteinase inhibitors was evaluated with L‐BApNA as a substrate. We studied lipoxygenase activity with linoleic acid and polyphenol oxidase activity with catechol substrates, in coffee plants damaged (T1) and not damaged (T2) by L. coffeella. L. coffeella digestive enzyme activity was verified by trypsin‐like (substrate l ‐BApNA and l ‐TAME), chymotrypsin‐like (BTpNA and ATEE), cysteine proteases (l ‐BApNA) and total protease (azocasein). Proteinase inhibitor production and lipoxygenase and polyphenol oxidase activity in C. arabica increases (P ≤ 0.05) with L. coffeella damage. Our results provide important information that these enzymatic activities may play a role in plant defence processes in C. arabica. Trypsin‐like activity increases, whereas chymotrypsin‐like and cysteine protease activity decrease in the midgut of L. coffeella, which acts as a defence mechanism. 相似文献

In the last decades, an increasing fishing effort and a decreasing trend in fish catches have been observed in southern Brazil. Considering that marine mammals and fisheries usually compete for the same resources, it is reasonable to presume that the feeding ecology of these predators is affected by the current scenario. To evaluate this hypothesis, long-term variation in the diet of the South American sea lion (Otaria flavescens) relative to fisheries exploitation was analyzed for two periods (1993–2003 versus 2004–2014). The degree of overlap between the relative biomass of the sea lions’ diet and the target species of six types of local fishery was analyzed. An increase in prey overlap between sea lions and fisheries was observed in the more recent sampling period, along with an increase in prey diversity, richness, and niche breadth of the sea lions’ diet. These results suggest that the overfishing scenario could partly explain the modified feeding ecology of the sea lions. In this context, we recommend a review and better regulation of the current fishing effort in the region, which we believe will be an important step to maintain the fish stocks and minimize the impact of fishing on marine top predators. 相似文献

Species distribution models for Amazonian trees have mostly been produced at scales and resolutions that are too broad and coarse for practical use in either conservation or forestry. On the other hand, several studies have shown that elevation and the medium‐resolution remote sensing data available via Landsat imagery can be successfully used to detect differences in plant species composition in Amazonia. Therefore, it seems likely that the same data can also be used to predict geographical distributions of individual taxa. Here we use remotely sensed data and a maximum entropy algorithm (MaxEnt) to generate landscape‐scale distribution models at 30‐m‐resolution for five economically important timber tree genera (Apuleia, Amburana, Crepidospermum, Dipteryx, and Manilkara). Individual Landsat Thematic Mapper bands and normalized difference vegetation index yielded acceptable model performance, and the use of averaging filters (3 × 3 and 5 × 5 pixel low‐pass filters) improved model performance further. Including elevation as a predictor also improved model performance for all the genera. Our results suggest that it is possible to use Landsat bands and elevation as predictors for modeling the potential distribution of tree species in lowland Amazonia at a fine enough resolution to facilitate the practical management of forest resources. 相似文献

Hosts counteract infections using two distinct defence strategies, resistance (reduction in pathogen fitness) and tolerance (limitation of infection damage). These strategies have been minimally investigated in multi-host systems, where they may vary across host species, entailing consequences both for hosts (virulence) and parasites (transmission). Comprehending the interplay among resistance, tolerance, virulence and parasite success is highly relevant for our understanding of the ecology and evolution of infectious and parasitic diseases. Our work investigated the interaction between an insect parasite and its most common bird host species, focusing on two relevant questions: (i) are defence strategies different between main and alternative hosts and, (ii) what are the consequences (virulence and parasite success) of different defence strategies? We conducted a matched field experiment and longitudinal studies at the host and the parasite levels under natural conditions, using a system comprising Philornis torquans flies and three bird hosts – the main host and two of the most frequently used alternative hosts. We found that main and alternative hosts have contrasting defence strategies, which gave rise in turn to contrasting virulence and parasite success. In the main bird host, minor loss of fitness, no detectable immune response, and high parasite success suggest a strategy of high tolerance and negligible resistance. Alternative hosts, on the contrary, resisted by mounting inflammatory responses, although with very different efficiency, which resulted in highly dissimilar parasite success and virulence. These results show clearly distinct defence strategies between main and alternative hosts in a natural multi-host system. They also highlight the importance of defence strategies in determining virulence and infection dynamics, and hint that defence efficiency is a crucial intervening element in these processes. 相似文献

In order to develop BiodiversityKnowledge, a Network of Knowledge working at the European science–policy interface for biodiversity and ecosystem services, we conducted three trial assessments. Their purpose was to test structure and processes of the knowledge synthesis function and to produce knowledge syntheses. The trial assessments covered conservation and management of kelp ecosystems, biological control of agricultural pests, and conservation and multifunctional management of floodplains. Following the BiodiversityKnowledge processes, we set up expert consultations, systematic reviews, and collaborative adaptive management procedures in collaboration with requesters, policy and decision-makers, stakeholders, and knowledge holders. Outputs included expert consultations, systematic review protocols, a group model and a policy brief. Important lessons learned were firstly that the scoping process, in which requesters and experts iteratively negotiate the scope, scale and synthesis methodology, is of paramount importance to maximize the scientific credibility and policy relevance of the output. Secondly, selection of a broad array of experts with diverse and complementary skills (including multidisciplinary background and a broad geographical coverage) and participation of all relevant stakeholders is crucial to ensure an adequate breath of expertise, better methodological choices, and maximal uptake of outcomes: Thirdly, as the most important challenge was expert and stakeholder engagement, a high visibility and reputation of BiodiversityKnowledge, supported by an incentive system for participation, will be crucial to ensure such engagement. We conclude that BiodiversityKnowledge has potential for a good performance in delivering assessments, but it requires adequate funding, trust-building among knowledge holders and stakeholders, and a proactive and robust interface with the policy and decision making community. 相似文献
