Identification of Cell Cycle Dependent Interaction Partners of the Septins by Quantitative Mass Spectrometry

The septins are a conserved family of GTP-binding proteins that, in the baker''s yeast, assemble into a highly ordered array of filaments at the mother bud neck. These filaments undergo significant structural rearrangements during the cell cycle. We aimed at identifying key components that are involved in or regulate the transitions of the septins. By combining cell synchronization and quantitative affinity-purification mass-spectrometry, we performed a screen for specific interaction partners of the septins at three distinct stages of the cell cycle. A total of 83 interaction partners of the septins were assigned. Surprisingly, we detected DNA-interacting/nuclear proteins and proteins involved in ribosome biogenesis and protein synthesis predominantly present in alpha-factor arrested that do not display an assembled septin structure. Furthermore, two distinct sets of regulatory proteins that are specific for cells at S-phase with a stable septin collar or at mitosis with split septin rings were identified.Complementary methods like SPLIFF and immunoprecipitation allowed us to more exactly define the spatial and temporal characteristics of selected hits of the AP-MS screen.     

Bacterial classification: an overview

Classification of bacteria evolved from limited subjective groupings to general, more objective arrangements based on overall phenotypic similarities. However, classifications based on phenotypic characters lack stability, whereas those based on genetic relatedness tend to be stable. DNA-DNA hybridization has proven to be extremely useful in resolving taxonomic problems at the species level. Broad relationships among bacteria have been identified by comparing ribosomal RNA cistrons; however, many groups based on ribosomal RNA analysis are not easily definable in terms of phenotypic similarities. Unless resolved, these problems could lead to the establishment of two separate classification systems, one phylogenetic and the other practical.     

Chemoreceptor function and metabolism of the carotid body in the rat

Using fluorimetric and polarographic determination of cell respiration, metabolic reactions of the carotid body of rats to adequate stimulation have been revealed. Functional relationships of the dose--effect type, which were found for these reactions, may be approximated by equations which are similar to those describing the unconditioned reflexes from the carotid chemoreceptors. However, reactions of the metabolics systems of the carotid body account for the perception of not all chemical stimuli, but only of acids and some alkaloids. These data reveal the heteregeneity of the carotid chemosensory system not only in the amphibians (which was known earlier), but in mammals as well.     

Transcriptional regulation of the antioxidant protein 2 gene, a thiol-specific antioxidant, by lens epithelium-derived growth factor to protect cells from oxidative stress.

Antioxidant protein 2 (AOP2), a member of the newly defined family of thiol-specific antioxidant proteins, has been shown to remove H(2)O(2) and protect proteins and DNA from oxidative stress. Here we report that LEDGF is one of the regulatory factors for the AOP2 gene. We found that LEDGF bound to the heat shock element and to stress-related elements in the AOP2 promoter. It trans-activated expression of AOP2-CAT in COS-7 cells and lens epithelial cells overexpressing LEDGF. Mutations in the heat shock element and stress-related elements of the AOP2 promoter reduced LEDGF-dependent trans-activation. Lens epithelial cells showed a higher level of AOP2 mRNA in the presence of LEDGF. Cells overexpressing LEDGF exhibited a higher level of AOP2 protein, the level of which was directly related to the increase in cellular protection. Thus, LEDGF, by activating the AOP2 gene, protected and enhanced the survival of cells under oxidative stress.     

Biosensors for assay of glycoalkaloids in potato tubers

The possibility of practical application of biosensors based on pH-sensitive field-effect transistors and butyrylcholinesterase to glycoalkaloid analysis in potato tubers was studied. The main analytical features of the designed biosensors and measurement conditions were optimized. The biosensor was applied to quantitative analysis of glycoalkaloids in tubers of different potato varieties. The results proved to be in good agreement with those obtained by conventional protocols. Experiments on glucose assay were performed. An inverse correlation between the contents of glucose and glycoalkaloids in potato tubers was demonstrated.     

Rotaviral enteritis in a raccoon

A hand-reared raccoon (Procyon lotor) kit had severe diarrhea and died within 24 hr. Gross and histopathologic findings were compatible with a diagnosis of viral enteritis. The immunoperoxidase test revealed rotavirus group A antigen in the intestinal mucosa. This is the first record of rotaviral enteritis in a raccoon.
