Character state‐based taxa erected to accommodate fossil and extant needle stoneflies (Leuctridae – Leuctrida tax.n.) and close relatives

In order to accommodate a newly discovered fossil specimen related to a subset of extant needle stoneflies (Leuctridae sensu Zwick, 2000), we provide a comparative analysis of the wing venation of these insects, and of their close relatives, primarily based on extant material. A number of character states potentially supporting monophyletic taxa are identified, and are used to erect taxon names under the cladotypic nomenclatural procedure. We introduce a new standard definition in which the mention of the antonym of the defining character state is applied. The proposed taxa, and taxa inclusiveness, are as follows: Nemouromorpha > Capniida/Leuctrida/‘Nemouridae s.l.’, and Leuctrida > Exeleuctrida > Mioleuctrida & Eleuctrida > Collaleuctrida. This scheme allowed us to tentatively accommodate several fossil species described earlier and the new specimen, left undetermined at the genus and species levels, but assigned to Collaleuctrida. To optimize retrieval of hierarchical information, we introduce the use of ‘nec’ (Latin for ‘but not’) into the taxonomic address to indicate exclusion from a taxon. For example, Mioleuctrida nec Collaleuctrida ?yehae indicates that ?yehae can be assigned to Mioleuctrida but can also be conclusively excluded from Collaleuctrida. Based on the investigated case, it is finally argued that the cladotypic nomenclatural procedure is optimal regarding name use and numbers, and allows adequate treatment of homoplasy.     

Organization and functional analysis of three T-DNAs from the vitopine Ti plasmid pTiS4

Summary The vitopine Ti plasmid pTiS4 of Agrobacterium vitis has an unusual T-DNA organization. The pTiS4 oncogenes, localized by screening selected pTiS4 clones for growth-inducing activity, are localized on three T-DNAs, whereas in all other characterized Ti plasmids one or two T-DNAs are found. The nucleotide sequences and predicted amino acid sequences of the pTiS4 oncogenes set them apart from the corresponding genes from other Ti or Ri plasmids. The oncogenes induce the same type of reaction on various test plants as the well-known pTiAch5 oncogenes but the pTiS4 ipt gene induces considerably more shoots than its Ach5 homologue. We have also identified the gene coding for vitopine synthase as well as a vitopine synthase pseudogene. Both sequences show homology to the octopine synthase gene. Analysis of intrapulmonary O(2) concentration by MR imaging of inhaled hyperpolarized helium-3.

Inhalation of hyperpolarized (3)He allows magnetic resonance imaging (MRI) of ventilated airspaces. (3)He hyperpolarization decays more rapidly when interacting with paramagnetic O(2). We describe a method for in vivo determination of intrapulmonary O(2) concentrations ([O(2)]) based on MRI analysis of the fate of measured amounts of inhaled hyperpolarized (3)He in imaged regions of the lung. Anesthetized pigs underwent controlled normoventilation in a 1.5-T MRI unit. The inspired O(2) fraction was varied to achieve different end-tidal [O(2)] fractions (FET(O(2))). With the use of a specifically designed applicator, (3)He (100 ml, 35-45% polarized) was administered at a predefined time within single tidal volumes. During subsequent inspiratory apnea, serial two-dimensional images of airways and lungs were acquired. At least once in each animal studied, the radio-frequency excitation used for imaging was doubled at constant FET(O(2)). Signal intensity measurements in regions of interest of the animals' lungs (volume range, 54-294 cm(3)), taken at two different radio-frequency excitations, permitted calculation of [O(2)] in these regions of interest. The [O(2)] fractions in the regions of interest correlated closely with FET(O(2)) (R = 0.879; P < 0.0001). O(2)-sensitive (3)He-MRI may allow noninvasive study of regional distribution of ventilation and alveolar PO(2) in the lung.     

A mathematical analysis of the relation between (bacterio) chlorophyll fluorescence yield and the concentration of reaction centre traps in photosynthesis

A random walk approach is used to discuss the relation between fluorescence yield and the concentration of reaction centre traps. For the matrix model as used for bacteria, non-negligible deviations are found from the Vredenberg-Duysens relation. An approximately linear re;ationship between the reciprocal of the fluorescence yield and the trap concentration remains. For the unit model as used for system 2, the deviation from Joliot's relation is small. The assumptions of the unit model are debatable. An alternative matrix model is presented.     

Properties of D(+)-lysopine dehydrogenase from crown gall tumour tissue.

D(+)-Lysopine dehydrogenase of an octopine-type Crown Gall tumour has been partially purified and a number of kinetic parameters have been determined. D(+)-Lysopine dehydrogenase catalyzes the reductive condensation of pyruvate and one of at least six different L-amino acids, as well as the reverse reactions, with preferential use of NADP(H) as a cofactor. The optimal pH for both reductive and oxidative reactions has been determined. At pH 6.8, L-lysine has of all the amino acids the lowest Km value, while at the same pH the highest V was found with L-arginine and L-histidine. The isoelectric point of D(+)-lysopine dehydrogenase is about 4.5.
