A novel β-mannanase gene, man5XZ7, was cloned from thermophilic fungus Thielavia arenaria XZ7, and successfully expressed in Pichia pastoris. The gene (1,110 bp) encodes a 369-amino acid polypeptide with a molecular mass of approximately 40.8 kDa. The deduced sequence of Man5XZ7 consists of a putative 17-residue signal peptide and a catalytic module belonging to glycoside hydrolase (GH) family 5, and displays 76 % identity with the experimentally verified GH 5 endo-β-1,4-mannanase from Podospora anserina. Recombinant Man5XZ7 was optimally active at 75 °C and pH?5.0 and exhibited high activity at a wide temperature range (>50.0 % activity at 50–85 °C). Moreover, it had good adaptability to acidic to basic pH (>74.1 % activity at pH?4.0–7.0 and 25.6 % even at pH?9.0) and good stability from pH?3.0 to 10.0. These enzymatic properties showed that Man5XZ7 was a new thermophilic and alkali-tolerant β-mannanase. Further amino acid composition analysis indicated that Man5XZ7 has several characteristic features of thermophilic enzymes. 相似文献

本研究旨在通过转录组分析预测的方法,由地衣芽孢杆菌中筛选获得一种新型双向启动子,鉴定其启动强度。以已知强组成型启动子pShuttle-09为对照,检测其对克劳氏芽孢杆菌碱性蛋白酶基因的表达活性。成功构建了3种重组碱性蛋白酶表达载体及对应的工程菌株。在新型启动子pLA和其反向启动子pLB调控转录下,克劳氏芽孢杆菌碱性蛋白酶表达活性达到164 U/mL和111 U/mL。结果表明,pLA的启动强度明显高于pShuttle-09和pLB,pLA启动子与pLB启动子均可表达碱性蛋白酶。从而为枯草芽孢杆菌表达系统中异源基因的表达提供一个新的方向,也为原核生物中共同表达两种基因提供了新的思路。 相似文献

The multidrug exporter AcrB is the inner membrane component of the AcrAB-TolC drug efflux system in Escherichia coli and is responsible for the resistance of this organism to a wide range of drugs. Here we describe the crystal structure of the trimeric AcrB in complex with a designed ankyrin-repeat protein (DARPin) inhibitor at 2.5-Å resolution. The three subunits of AcrB are locked in different conformations revealing distinct channels in each subunit. There seems to be remote conformational coupling between the channel access, exit, and the putative proton-translocation site, explaining how the proton motive force is used for drug export. Thus our structure suggests a transport pathway not through the central pore but through the identified channels in the individual subunits, which greatly advances our understanding of the multidrug export mechanism. 相似文献

Populations of northern small rodents have previously been observed to fluctuate in spatial synchrony over distances ranging from tens to hundreds of kilometers between sites. It has been suggested that this phenomenon is caused by common environmental perturbations, mobile predators or dispersal movements. However, very little focus has been given to how the physical properties of the geographic area over which synchrony occurs, such as landscape composition and climate, affect spatial population dynamics. This study reports on the spatial and temporal properties of vole population fluctuations in two areas of western Finland: one composed of large interconnected areas of agricultural farmland interspersed by forests and the other highly dominated by forest areas, containing more isolated patches of agricultural land. Furthermore, the more agricultural area exhibits somewhat milder winters with less snow than the forested area. We found the amplitude of vole cycles to be essentially the same in the two areas, suggesting that the relative amount of predation on small rodents by generalist versus specialist predators is similar in both areas. No seasonal differences in the timing of synchronization were observable for Microtus voles, whereas bank vole populations in field habitats appeared to become synchronized primarily during winter. Microtus populations in field habitats exhibited smaller spatial variation and a higher degree of synchrony in the more continuous agricultural landscape than in the forest-dominated landscape. We suggest that this inter-areal difference is due to differences in the degree of inter-patch connectivity, with predators and dispersal acting as the primary synchronizing agents. Bank vole populations in field habitats were more synchronized within the forest-dominated landscape, most likely reflecting the suitability of the inter-patch matrix and the possibility of dispersal. The single large or several small (SLOSS) problem has been addressed in a large number of empirical and theoretical studies, but no coherent conclusion has yet been reached. Here I study the SLOSS problem in the context of metapopulation dynamics. I assume that there is a fixed total amount A(0) of habitat available, and I derive formulas for the optimal number n and area A of habitat patches, where n=A(0)/A. I consider optimality in two ways. First, I attempt to maximize the time to metapopulation extinction, which is a relevant measure for metapopulation viability for rare and threatened species. Second, I attempt to maximize the metapopulation capacity of the habitat patch network, which corresponds both with maximizing the distance to the deterministic extinction threshold and with maximizing the fraction of occupied patches. I show that in the typical case, a small number of large patches maximizes the metapopulation capacity, while an intermediate number of habitat patches maximizes the time to extinction. The main conclusion stemming from the analysis is that the optimal number of patches is largely affected by the relationship between habitat patch area and rates of immigration, emigration and local extinction. Here this relationship is summarized by a single factor zeta, termed the patch area scaling factor. 相似文献

We review recent developments in spatially realistic metapopulation theory, which leads to quantitative models of the dynamics of species inhabiting highly fragmented landscapes. Our emphasis is in stochastic patch occupancy models, which describe the presence or absence of the focal species in habitat patches. We discuss a number of ecologically important quantities that can be derived from the full stochastic models and their deterministic approximations, with a particular aim of characterizing the respective roles of the structure of the landscape and the properties of the species. These quantities include the threshold condition for persistence, the contributions that individual habitat patches make to metapopulation dynamics and persistence, the time to metapopulation extinction, and the effective size of a metapopulation living in a heterogeneous patch network. 相似文献

Background
Etiological assessment of stroke is essential for accurate treatment decisions and for secondary prevention of recurrence. There is evidence that interleukin-10 (IL-10) associates with ischemic stroke. The aim of this prospective study was to assess the levels of IL-10 in ischemic stroke with unknown or suspected cardiogenic etiology, and evaluate the correlation between IL-10 plasma concentration and the number of diagnosed high risk sources for cardioembolism.Methods
A total of 141 patients (97 males; mean age 61±11 years) with acute ischemic stroke with unknown etiology or suspected cardiogenic etiology other than known atrial fibrillation (AF) underwent imaging investigations to assess high risk sources for cardioembolic stroke established by the European Association of Echocardiography (EAE). IL-10 was measured on admission to the hospital and on a three month follow-up visit.Results
Acute phase IL-10 concentration was higher in patients with EAE high risk sources, and correlated with their number (p<0.01). In patients with no risk sources (n = 104), the mean IL-10 concentration was 2.7±3.1 ng/L (range 0.3–16.3 ng/L), with one risk source (n = 26) 3.7±5.5 ng/L (0.3–23.6 ng/L), with two risk sources (n = 10) 7.0±10.0 ng/L (1.29–34.8 ng/L) and with three risk sources (n = 1) 37.2 ng/L. IL-10 level was not significantly associated with cerebral infarct volume, presence of previous or recent myocardial infarction, carotid/vertebral artery atherosclerosis, paroxysmal AF registered on 24-hour ECG Holter monitoring or given intravenous thrombolytic treatment.Conclusion
IL-10 plasma concentration correlates independently with the number of EAE cardioembolic risk sources in patients with acute stroke. IL-10 may have potential to improve differential diagnostics of stroke with unknown etiology. 相似文献9.
Species distributions often show an aggregated pattern, which can be due to a number of endo- and exogenous factors. While autologistic models have been used for modelling such data with statistical rigour, little emphasis has been put on disentangling potential causes of aggregation. In this paper we ask whether it is possible to infer sources of aggregation in species distributions from a single set of occurrence data by comparing the performance of various autologistic models. We create simulated data sets, which show similar occupancy patterns, but differ in the process that causes the aggregation. We model the distribution of these data with various autologistic models, and show how the relative performance of the models is sensitive to the factor causing aggregation in the data. This information can be used when modelling real species data, where causes of aggregation are typically unknown. To illustrate, we use our approach to assess the potential causes of aggregation in data of seven bird species with contrasting statistical patterns. Our findings have important implications for conservation, as understanding the mechanisms that drive population fluctuations in space and time is critical for the development of effective management actions for long-term conservation. 相似文献
