Identification of the precise molecular pathways involved in oncogene-induced transformation may help us gain a better understanding of tumor initiation and promotion. Here, we demonstrate that SOX2+ foregut epithelial cells are prone to oncogenic transformation upon mutagenic insults, such as KrasG12D and p53 deletion. GFP-based lineage-tracing experiments indicate that SOX2+ cells are the cells-of-origin of esophagus and stomach hyperplasia. Our observations indicate distinct roles for oncogenic KRAS mutation and P53 deletion. p53 homozygous deletion is required for the acquisition of an invasive potential, and KrasG12D expression, but not p53 deletion, suffices for tumor formation. Global gene expression analysis reveals secreting factors upregulated in the hyperplasia induced by oncogenic KRAS and highlights a crucial role for the CXCR2 pathway in driving hyperplasia. Collectively, the array of genetic models presented here demonstrate that stratified epithelial cells are susceptible to oncogenic insults, which may lead to a better understanding of tumor initiation and aid in the design of new cancer therapeutics.  相似文献

The mycorrhizal status of plant species in north-west Patagonia was examined. Communities representative of Patagonian steppe and marshes were compared with respect to the mycorrhizal status of their species.Most of both native and exotic plant species sampled were arbuscular mycorrhizal (AM). The percentage of species with mycorrhizal association was higher for perennial herbs and shrubs than for annual herbs. The higher ratio of mycorrhizal/nonmycorrhizal (NM) species found for dicotyledons than for monocotyledons, could reflect the presence of a considerable number of NM monocotyledons in the marsh. The mycorrhizal status of plants differed slightly between the steppe and the marsh. In the steppe, native AM species were more frequent than in the marsh. In contrast, in the marsh, the NM species were proportionally more represented than in the steppe. The Juncaceae and Cyperaceae, which include hydrohytic NM plants, accounted for many of these differences. Moreover, the dominant species in the marshes, Juncus arcticus, is a NM species.In the present study, most of species belonging to the same taxonomic family tended to have the same mycorrhizal associations, in agreement with studies on plants from other regions. Exceptions to this general behaviour were observed in the families Cyperaceae, Scrophulariaceae, Berberidaceae and Amaryllidaceae. The most represented families in which mycorrhizal behaviour differed between species of the same family were Asteraceae, Fabaceae and Poaceae. Senecio neaei (Asteraceae) and Boopis australis (Calyceraceae) showed facultative mycorrhizal behaviour.  相似文献

The saprophytic fungi Wardomyces inflatus (Marchal) Hennebert, Paecilomyces farinosus (Holm & Gray) A. H. S. Brown & G. Sm., Gliocladium roseum Bain., sterile dark mycelium (SDM-54), Trichoderma pseudokoningii Rifai and Trichoderma harzianum Rifai were isolated from sporocarps of Glomus mosseae. The effect of saprophytic fungi on G. mosseae spore germination was tested on water agar. Wardomyces inflatus decreased the percent germination of G. mosseae spores; G. roseum, T. pseudokoningii and T. harzianum had no effect on germination; and P. farinosus and SDM-54 increased the percentage of spore germination of G. mosseae after 4 d. Wardomyces inflatus significantly decreased hyphal length of spores which germinated, but no other saprophytic fungi affected hyphal growth. Trichoderma pseudokoningii, T. harzianum, P. farinosus and SDM-54 increased the number of auxiliary cells formed by G. mosseae. The effect of saprophytic fungi on arbuscular mycorrhizal colonization of soybean was studied in a greenhouse trial. The percentage of soybean root length colonized was decreased by W. inflatus, unaffected by SDM-54 and T. harzianum, and increased by P. farinosus. Gliocladium roseum decreased root length colonized when plants were 12 wk old, and T. pseudokoningii increased colonization of roots when plants were 4 wk old. Antagonistic, synergistic and neutral actions of G. mosseae upon the saprophytic fungi were observed. The population of T. harzianum decreased and the populations of T. pseudokoningii and SDM-54 increased in the presence of G. mosseae. Our results indicate a complex interaction between G. mosseae and associated saprophytic fungi.  相似文献

Neohelice granulata provides an interesting animal model for studying behavioural process because it is widely distributed, ensuring variability related to different environmental conditions. The aim of this study was to analyse variation in site fidelity with relation to landscape heterogeneity. Field observations were carried out in three geographically distant marshes in Argentina (Mar Chiquita, San Antonio Oeste, and Riacho San José), which differ in their environmental characteristics and where crabs display different reproductive strategies. We analysed potential variation in burrow fidelity with relation to body size, sex and presence of vegetation (mudflat and saltmarsh) at all study sites. In addition, we analysed the influence of tidal flooding on fidelity in the Mar Chiquita saltmarsh. To achieve these goals, we used a mark–recapture method in which we tagged approximately 100 crabs during mid-summer for each zone at each site (a total of 668 crabs) for geographical comparison and approximately 370 crabs to evaluate the influence of tidal flooding. We found more faithful individuals in Mar Chiquita than in San Antonio Oeste and Riacho San José. For the San Antonio Oeste and Riacho San José populations and for Mar Chiquita previous to flooding samples we also found differences in site fidelity related to crab body size. At San Antonio Oeste and Riacho San José the relationship between size and proportion of faithful crabs was negative (smaller crabs were more faithful than larger crabs). In Mar Chiquita, a higher proportion of fidelity previous to flooding and a size-dependent response to flooding were detected, suggesting that fidelity may be modulated by tides having different effects on crabs with different body sizes. Equal proportions of males and females displaying site fidelity were observed at all study zones and sites. Our study suggests that N. granulata burrow fidelity behaviour changes with latitude and landscape (mudflat or saltmarsh) and can be sensitive to variables such as body size and frequency of flooding.  相似文献

 Chickpea (Cicer arietinum L.) ranks third in the world, and first in the Mediterranean basin, for production among pulses. Despite its importance as a crop and considerable research effort, traditional breeding methods have so far been unable to produce cultivars with a large impact on chickpea production. Interspecific hybridization is known to improve yield in many crops. Therefore, an attempt was made to increase the seed yield in chickpea through the introgression of genes from wild relatives at the International Center for Agricultural Research in the Dry Areas (ICARDA), Syria, from 1987 to 1995. Four crosses, ILC 482 (C. arietinum)×ILWC 179 (C. echinospermum) and ILC 482×ILWC 124 (C. reticulatum) and their reciprocals, were made. Pedigree selection was used to advance the material. Heterosis was recorded visually in F1s, and single plant measurements for seed yield were recorded in F2 populations. Promising and uniform progenies were bulked in the F5 generation. Out of 96 F6 lines, 22 were selected on the basis of seed yield and other agronomic characters, and evaluated in a replicated trial for seed yield and 14 agronomical, morphological and quality characters. A high level of heterosis was observed in F1s. Several F2 plants produced two to three times more seed yield than the best plant from the cultigen. Nine F7 lines out-yielded the cultigen parent by up to 39%. Over 2 years, 12 lines had a higher yield than the cultigen parent. These lines were not only high yielding but also free of any known undesirable traits from the wild species, such as spreading growth habit, pod dehiscence, and non-uniform maturity. Quality traits, such as seed shape, type, colour, weight, and testa texture, protein content, cooking time and an organoleptic test of a Middle East dish, Homos Bi-Tehineh, were also similar to the cultigen parent. Summary Roots, stems, or leaves of American (Panax quinquefolium) and Korean (Panax ginsing) ginseng were grown as callus or supension tissue cultures. Tissue cultures ofP. ginseng would occasionally form plantlets. The fundamental chemical composition, inorganic analysis, and saponin (panaquilin) content of American and Korean ginseng plants and tissue cultures were determined. The crude saponin content is very similar to, but approximately one-half (1.3%, fresh weight) of that present in ginseng roots. Two-dimensional thin layer chromatographic analysis revealed minor differences in the panaquilins present in American and Korean ginseng tissue cultures. The sapogenin, panaxadiol, was isolated from Korean ginseng callus.  相似文献

The endemic New Zealand ground wētā (Hemiandrus sp. ‘promontorius’) has a Naturally Uncommon conservation status. This is because of the paucity of information on its density and distribution. Here, the biology, density and distribution of a population of this wētā found in and around vineyards in the Awatere Valley, Marlborough was studied. Wētā density was assessed in vineyards, paddocks and shrublands in this valley. Soil moisture, penetration resistance, pH and organic matter were recorded at locations with and without wētā. Wētā density in vineyards was significantly higher than in either paddocks or shrub habitats. In vineyards, the density of this insect was significantly higher under-vines than in the inter-rows. Higher numbers of this wētā were found in moist soils that required lower force to burrow. Females laid an average of 55 eggs between March and April, which hatched in September. These findings highlight the intersection between agriculture and conservation.  相似文献
