A thermophilic bacterium, identified as a neighboring species to Geobacillus thermocatenulatus, having a growth optimum at 55 °C and, capable of degrading nylon 12, was isolated from soil by enrichment culture technique at 60 °C. At this temperature, the strain grew on 5 g nylon 12 l–1 with a decrease in its molecular weight from 41000 to 11000 over 20 d. The degradation was assumed to be due to endogenous hydrolysis of amide bond in nylon 12. The strain degraded also nylon 66 with a decrease in its molecular weight from 43000 to 17000 in 20 d at 60 °C. Nylon 6 was not degraded.  相似文献

C-type natriuretic peptide (CNP) has been shown to play an important role in the regulation of vascular tone and remodeling. However, the physiological role of CNP in the lung remains unknown. Accordingly, we investigated whether CNP infusion attenuates bleomycin (BLM)-induced pulmonary fibrosis in mice. After intratracheal injection of BLM or saline, mice were randomized to receive continuous infusion of CNP or vehicle for 14 days. CNP infusion significantly reduced the total number of cells and the numbers of macrophages, neutrophils, and lymphocytes in bronchoalveolar lavage fluid. Interestingly, CNP markedly reduced bronchoalveolar lavage fluid IL-1beta levels. Immunohistochemical analysis demonstrated that CNP significantly inhibited infiltration of macrophages into the alveolar and interstitial regions. CNP infusion significantly attenuated BLM-induced pulmonary fibrosis, as indicated by significant decreases in Ashcroft score and lung hydroxyproline content. CNP markedly decreased the number of Ki-67-positive cells in fibrotic lesions of the lung, suggesting antiproliferative effects of CNP on pulmonary fibrosis. Kaplan-Meier survival curves demonstrated that BLM mice treated with CNP had a significantly higher survival rate than those given vehicle. These results suggest that continuous infusion of CNP attenuates BLM-induced pulmonary fibrosis and improves survival in BLM mice, at least in part by inhibition of pulmonary inflammation and cell proliferation.  相似文献

Previous reports indicated the significance of the TGF-β signaling in the pathogenesis of systemic sclerosis. We tried to evaluate the possibility that microRNAs (miRNAs) play a part in the type I collagen upregulation seen in normal fibroblasts stimulated with exogenous TGF-β and systemic sclerosis (SSc) fibroblasts. miRNA expression profile was evaluated by miRNA PCR array and real-time PCR. The protein expression of type I collagen was determined by immunoblotting. In vivo detection of miRNA in paraffin section was performed by in situ hybridization. Several miRNAs were found to be downregulated in both TGF-β-stimulated normal fibroblasts and SSc fibroblasts compared with normal fibroblasts by PCR array. Among them, miR-196a expression was decreased in SSc both in vivo and in vitro by real-time PCR or in situ hybridization. In SSc fibroblasts, miR-196a expression was normalized by TGF-β small interfering RNA. miR-196a inhibitor leads to the overexpression of type I collagen in normal fibroblasts, whereas overexpression of the miRNA resulted in the downregulation of type I collagen in SSc fibroblasts. In addition, miR-196a was detectable and quantitative in the serum of SSc patients. Patients with lower serum miR-196a levels had significantly higher ratio of diffuse cutaneous SSc:limited cutaneous SSc, higher modified Rodnan total skin thickness score, and higher prevalence of pitting scars than those without. miR-196a may play some roles in the pathogenesis of SSc. Investigation of the regulatory mechanisms of type I collagen expression by miR-196a may lead to new treatments using miRNA.  相似文献

Paddy fields surrounded by forests are characteristic of the rural landscapes in East Asia. These characteristic landscapes are maintained by agricultural activities; recently, however, local and regional changes in economic and social systems have resulted in their deterioration. In order to conserve these landscapes, the diversity of land-use systems should be documented, and the human impact on landscape structure should be analyzed. In this study, landscape structure, stand structure, and influence of human activity on the landscape were examined for a hilly agricultural area in Fenghua City, Zhejiang Province, China. Eleven types of landscape elements were recognized that include evergreen broad-leaved forests, evergreen coniferous (Pinus massoniana) forests, tall bamboo (Phyllostachys pubescens) forests, small bamboo (Phyllostachys praecox f. prevernalis) plantations, tea gardens, grasslands, dry arable fields, paddy fields, residential areas, bare ground, and open water. In the study plots, which were established in the main forest types, the species name, cover abundance, and sociability of vascular plants were recorded, and tree height and diameter at breast height were measured. In addition, the amount of fuelwood collected from the forests was determined. There were many component species common to both secondary and natural forests, but the stand structure for these forests was considerably different. Cyclobalanopsis (Quercus) glauca, which had adapted to being frequently cut, was found to be the preferred species for fuelwood.  相似文献

The greatest mass extinction occurred at the end of the Permian. Most records of the mass extinction are not from pelagic sediments, but from shallow-marine and terrestrial sediments. Although several pelagic sections that span the end-Permian mass extinction have been found, these sections contain few index fossils and are often discontinuous because of small faults. We found the index fossils Albaillella cf. triangularis (Radiolaria) in siliceous claystone beds, Hindeodus parvus (Conodont) in the overlying black claystone beds, and Neospathodus cf. cristagalli and Ns. waageni (Conodont) in the subsequent siliceous claystone beds in Akkamori section-2 in northern Japan. These fossils suggest that this section ranges from the late Permian to the Early Triassic, including the early Induan and Olenekian stages. Furthermore, the lithological changes in the section, i.e., starting from bedded chert through siliceous claystone and black claystone to siliceous claystone, are concordant with those of well-known Permian–Triassic pelagic sequences in Japan. There is no gap between each lithofacie of the Akkamori section-2. Critical lithological continuity between Upper Permian siliceous claystone beds and uppermost Permian to lowermost Triassic black claystone beds of the Akkamori section-2 was recognized by observing hand-polished specimens and thin sections. Such paleontological and sedimentological evidence implies that the Akkamori section-2 is a continuous pelagic section that records the end-Permian mass extinction event. The carbonaceous black claystone beds have high total organic carbon (TOC) concentrations (1.06–3.31 wt.%), suggesting oceanic anoxia at least deep and probably stable primary productivity. A decrease in radiolarian abundance from 26–563 to 0.27–20 specimens/cm2 coincided with an increase in TOC content from 0.01–0.16 to 1.06–3.31 wt.% at the boundary of the siliceous claystone and the overlying black claystone beds near the top of the Permian, implying that radiolarian extinction occurred at the end of the Permian coinciding with oceanic anoxia. Although TOC contents decreased in the early Olenekian (Smithian), radiolarian abundance did not increase at that time, indicating that radiolarian recovery was delayed by > 1.5 m.y.  相似文献

Monoclonal antibodies (MAbs) are important for in-depth antigenic characterization and diagnosis of infections with human caliciviruses that cause almost all outbreaks of nonbacterial gastroenteritis. We compared different routes of immunization with nonreplicating virus-like particles (VLPs) from recombinant Norwalk virus (rNV) and recombinant Mexico virus (rMX) administered to BALB/c mice to determine the efficiency of hybridoma production. Oral immunization with VLPs without adjuvant resulted in high yields of MAb-secreting hybridomas (90%) to these VLPs of IgG (61%), IgM (29%) and IgA (10%) isotypes. Fusions with mesenteric lymph node lymphocytes yielded MAbs of various subclasses including IgG2a, IgG3, IgM and IgA. These results suggest that an immunization route that mimics the natural route of viral infection pathway may facilitate MAb technology by increasing the yields of antibody secreting hybridoma cells.  相似文献

The balance between prostacyclin and thromboxane A2 (TXA2) plays an important role in pulmonary homeostasis. However, little information is available regarding the therapeutic potency of these prostanoids for pulmonary fibrosis. We have recently developed ONO-1301, a novel long-acting prostacyclin agonist with thromboxane synthase inhibitory activity. Thus we investigated whether repeated administration of ONO-1301 attenuates bleomycin-induced pulmonary fibrosis in mice. After intratracheal injection of bleomycin or saline, mice were randomized to receive repeated subcutaneous administration of ONO-1301 or vehicle. Bronchoalveolar lavage (BAL) and histological analyses were performed at 3, 7, and 14 days after bleomycin injection. In vitro studies using mouse lung fibroblasts were also performed. ONO-1301 significantly attenuated the development of bleomycin-induced pulmonary fibrosis, as indicated by significant decreases in Ashcroft score and lung hydroxyproline content. ONO-1301 significantly reduced total cell count, neutrophil count, and total protein level in BAL fluid in association with a marked reduction of TXB2. A single administration of ONO-1301 significantly increased plasma cAMP level for >2 h. In vitro, ONO-1301 and a cAMP analog dose-dependently reduced cell proliferation in mouse lung fibroblasts. The reduction in cell proliferation by ONO-1301 was attenuated by a protein kinase A (PKA) inhibitor. Furthermore, bleomycin mice treated with ONO-1301 had a significantly higher survival rate than those given vehicle. These results suggest that repeated administration of ONO-1301 attenuates the development of bleomycin-induced pulmonary fibrosis and improves survival in bleomycin mice, at least in part by inhibition of TXA2 synthesis and activation of the cAMP/PKA pathway.  相似文献

Guadalupian–Lopingian sedimentary rocks are widely distributed in accretionary complexes in Japan, but the Guadalupian–Lopingian boundary (G–LB) is not well documented from these pelagic sediments. To identify the G–LB and to better correlate an extinction event that occurred around the Guadalupian–Lopingian boundary, we examined the conodont biostratigraphy from a Permian pelagic chert sequence in the Gujo-hachiman section, Gifu, southwest Japan. Age-diagnostic conodonts, including Clarkina postbitteri postbitteri, were found in this section. The biostratigraphic occurrences of these age-diagnostic conodonts can pinpoint the “G–L transitional zone” in the Gujo-hachiman section by comparison with well-studied sections from south China, including the GSSP section. The transitional zone was recognized by the first occurrence horizons of both Clarkina postbitteri hongshuiensis and C. p. postbitteri. The G–LB has been placed at or above the first occurrence horizon of the radiolarians Albaillella yamakitai or Albaillella cavitata in previous studies from China and Japan. We detected the first occurrence horizon of A. yamakitai below the base of the “G–L transitional zone,” in the Upper Capitanian. The conodont biostratigraphy is consistent with the radiolarian biostratigraphy in this section, which can be correlated to relevant sections in China.  相似文献

A method enabling the object-oriented image analysis of landscape elements incorporating topographic data was designed and tested on a Japanese countryside target area. IKONOS data (four multispectral bands with a spatial resolution of 4 m and a panchromatic band with a spatial resolution of 1 m) acquired on 23 April 2001 were used. Definiens v.5 software (Definiens AG, München, Germany) was employed for the classification. The initial segmentation was multiresolution and bottom-up, and each segment identified was considered to be one object. Two classifications employing the same landscape elements and ground truth data were implemented. One classification adopted an object-based image analysis classification method based on spectral characteristics; the other utilized an object-oriented image analysis (OOIA) that allows for a suitable scale parameter to be selected independently for each landscape element. In addition, topographic data derived from field surveys (walking surveys) and topographic maps were used to create a topographic database delineating the boundary between valley bottoms and the adjacent slopes (elevation: about 10 m). These data were then integrated into the OOIA analysis. The accuracies of the two classifications were assessed by comparing the results to a master landscape map produced directly from aerial photographs and on-site observations. The object-oriented method using the topographic data resulted in a higher overall kappa coefficients (0.63–0.47) than the object-based method. These results indicate that object-oriented image analysis of very high resolution data used in combination with topographic data can be an effective tool for landscape classification in Japan, where historical land-use patterns have resulted in finely dissected landscapes.  相似文献
