Coated microvesicle fractions isolated from ox forebrain cortex by the ultracentrifugation procedure of Pearse (1) and by the modified, less time consuming method of Keen et al (2) had comparable Ca2++Mg2+ dependent ATPase activities (about 9 μmol/h per mg protein). The Na++K++Mg2+ dependent ATPase activity was 3.2 μmol/h per mg (±1.0, S.D., n=3) when microvesicles were prepared according to (1) and 1.5 μmol/h per mg (±1.0, S.D., n=3) when prepared according to (2).Oligomycin, ruthenium red, and trifluoperazine, inhibitors of Ca2+ transport in mitochondria and erythrocyte membranes had no effect on Ca2++Mg2+ dependent ATPase from any of the preparations.As demonstrated both by ATPase assays and electron microscopy, coated microvesicles could be bound to immunosorbents prepared with poly-specific antibodies against a coated microvesicle fraction obtained by the method of Pearse (1). The binding could be inhibited by dissolved coat protein using partially purified clathrin. The fraction of coated vesicles eluted from the immunosorbent was purified relative to the starting material as judged by electron microscopy.The Ca2++Mg2+ ATPase activity and calmodulin content was copurified with the coated microvesicles and the specific activity of Na++K++Mg2+ ATPase was decreased.Na++K++Mg2+ dependent ATPase activity in the coated microvesicle fraction could be ascribed to membranes with the appearance of microsomes. These membranes were also bound to the immunosorbents, but the binding was not influenced by clathrin. The capacity of the immunosorbents for these membranes was less than for the coated microvesicles, resulting in a decrease of Na++K++Mg2+ dependent ATPase activity in the eluted coated microvescile fraction.It was concluded that Ca2++Mg2+ ATPase activity is not a contamination from plasma membrane vesicles or mitochondrial membranes but seems to be an integral part of the coated vesicle membrane.  相似文献

The coprodeum is a very efficient Na+-retaining epithelium. Coprodeum from birds on a high Na+ diet has virtually no ion transport, while an Amiloride-sensitive Na+ absorption of 10–12 μ equiv·cm?2·h?1 is induced in the coprodeal epithelium from birds on a low Na+ diet. Both measurements of the Na+ influx and Na+-diffusion potentials across the luminal cell membrane have revealed a selective opening of this membrane to Na+ in birds on a low Na+ diet. Freeze-fracture P faces of the luminal membrane in coprodea taken from birds on a low Na+ diet have rod-shaped particles, 100 × 240 A?, in more than 20% of the principal cells. Rod-shaped particles appear in less than 1% of these cells in coprodea from high Na+-diet birds. Thus a low Na+ diet induces rod-shaped particles in the luminal cell membrane of the hen coprodeum. These new particles may function as Na+-channels mediating the increased Na+-influx across the apical cell membrane.  相似文献

On typing of 90 strains of bovine Group-B streptococci (B-str.) from 21 herds, NT1, NT2 and NT3 antigens were found in 30 %, viz., NT1 in 14.4 %, NT2 in 3.3 %, and NT3 in 12.2 %. In 9 of the 21 herds examined (43 %) one or other of these antigens was found. Among 8 strains from 4 herds of Herd type NT the NT1 antigen was demonstrated once. In 5 herds of Herd type X, 20 strains were typed, and 9 isolates (45 %) from 3 herds (60 %) carried one or other of the three antigens. Among herds in which the herd type was referable to polysaccharide antigens of the accepted set, NT3 antigen was present in 1 herd of Herd type Ia, namely along with IaX antigens in two strains, and with Ia antigen in one. In 3 herds of Herd type Ibc and in 2 herds of Herd type Ib no NT1, NT2, or NT3 antigen was demonstrated. On the other hand, such antigens were found in all of 4 herds of Herd type III, being present in 50 % of the strains (14/28) examined, and the frequency of NT1, NT2, and NT3 antigens was only slightly higher among NT and X strains (10/19 or 53 %) than among III and IIIX strains (4/9 or 44 %). The NT2 reference strain carried the Ibc protein antigen. In addition to strong homologous reactions, NT1 antiserum reacted with B-str. group antigen, NT2 antiserum with Ibc antigen, and NT3 antiserum with the NT1 reference strain. It is concluded that the NT1, NT2, and NT3 antigens are of doubtful value in epidemiological studies on bovine infections with group-B streptococci.  相似文献

The chaperone calreticulin is a highly conserved eukaryotic protein mainly located in the endoplasmic reticulum. It contains a free cysteine SH group but does not form disulfide-bridged dimers under physiological conditions, indicating that the SH group may not be fully accessible in the native protein. Using PAGE, urea gradient gel electrophoresis, capillary electrophoresis and MS, we show that dimerization through the SH group can be induced by lowering the pH to 5-6, heating, or under conditions that favour partial unfolding such as urea concentrations above 2.6 m or SDS concentrations above 0.025%. Moreover, we show that calreticulin also has the ability to self-oligomerize through noncovalent interactions at urea concentrations above 2.6 m at pH below 4.6 or above pH 10, at temperatures above 40 degrees C, or in the presence of high concentrations of organic solvents (25%), conditions that favour partial unfolding or an intramolecular local conformational change that allows oligomerization, resulting in a heterogeneous mixture of oligomers consisting of up to 10 calreticulin monomers. The oligomeric calreticulin was very stable, but oligomerization was partially reversed by addition of 8 m urea or 1% SDS, and heat-induced oligomerization could be inhibited by 8 m urea or 1% SDS when present during heating. Comparison of the binding properties of monomeric and oligomeric calreticulin in solid-phase assays showed increased binding to peptides and denatured proteins when calreticulin was oligomerized. Thus, calreticulin shares the ability to self-oligomerize with other important chaperones such as GRP94 and HSP90, a property possibly associated with their chaperone activity.  相似文献

Synthetic peptides based on amino-acid residues 27-38 of human serum amyloid P component represent a novel type of heparin binders as they do not contain clusters of basic amino acids or other known features associated with protein or peptide heparin binding. Here, we characterize the binding using capillary electrophoresis (CE), surface plasmon resonance (SPR), and isothermal titration calorimetry (ITC). By CE, heparin-binding activity was readily apparent for both a regular peptide and a slightly N-terminally modified form, while a sequence-scrambled peptide had no measurable binding. Dissociation constants in the 1-15 microm range were estimated, but only a minor part of the binding isotherm was covered by the experiments. SPR measurements using immobilized peptides verified heparin binding, the range of the binding constants, and the reduced binding of the sequence-scrambled peptide. Structurally defined heparin oligosaccharides were used to establish that while the tetrasaccharide is too small to exhibit strong binding, little difference in binding strength is observed between hexa- and tetradeca-saccharides. These experiments also confirmed the almost complete lack of activity of the sequence-scrambled peptide. The amino-acid sequence-dependent binding and the importance of a disulfide bond in the peptide were verified by ITC, but the experimental conditions had to be modified because of peptide precipitation and ITC yielded significantly weaker binding constants than the other methods. While the precise function of the peptide in the intact protein remains unclear, the results confirm the specificity of the glycosaminoglycan interaction with regard to peptide sequence by applying two additional biophysical techniques and showing that the N-terminal part of the peptide may be modified without changing the heparin binding capabilities.  相似文献

Waste paper is suitable for recycling back into paper or for incineration for energy recovery. If waste paper is used for recycling, secondary pulp replaces virgin pulp. Fiber recycling is limited, however, because of physical constraints—particularly the breakage of fiber in the recycling process—and a permanent input of virgin fiber to the system is required. Therefore one can expect that the relationship between recycling rates and resource requirements is represented by a curved line rather than a straight one. In this article, we present a mathematical model which confirms that the relationship between recycling rates and primary pulp requirements can be described as nonlinear. Furthermore, we show that this nonlinear relationship leads to an optimal recycling rate with regard to energy consumption: 93 persent for paper produced from chemical pulp, and 81 persent for paper produced from mechanical pulp. Sensitivity testing additionally reveals that at low recycling rates increasing waste paper recycling is energy efficient, but it becomes less efficient at higher recycling rates. Close to the optimum recycling rates (within 10 persent), increasing or decreasing the rate affects the total energy requirement less than 0.3%  相似文献

Bladder function is dependent upon cellular metabolism of substrates and the adequate generation of high-energy phosphate compounds. Partial outlet obstruction induces a marked decrease in bladder function which is associated with a significant decrease in the oxidative metabolism of glucose.The current investigation was designed to determine whether the time course of the decrease in mitochondrial oxidation in the hypertrophied urinary bladder is similar to the time course of the contractile dysfunction observed. In these studies we determined: 1) the rate of 14C-pyruvate metabolism to 14CO2 in control and obstructed tissue (1, 3, 5 and 7 days), and 2) the mitochondrial enzymatic activities of malate dehydrogenase and citrate synthase.The results can be summarized as follows: 1) The rate of pyruvate metabolism decreases by over 50% within one day following partial outlet obstruction, and remains at this level for the seven day period of study. 2) Kinetic analysis demonstrates that the change in enzymatic activity is related to a decrease in Vmax; the Kd for pyruvate is similar for control and after all time periods of obstruction. 3) The enzymatic activity of malate dehydrogenase and citrate synthase is reduced by over 50% within one day following partial obstruction, and remains at this level throughout the 7 day study period. These metabolic results correlate in time and duration with the decreased ability of the bladder to empty following partial outlet obstruction.  相似文献

Incorporation of [3H]leucine and [3H]valine into proteins of freshwater bacteria was studied in two eutrophic lakes. Incorporation of both amino acids had a saturation level of about 50 nM external concentration. Only a fraction of the two amino acids taken up was used in protein synthesis. At 100 nM, the bacteria respired 91 and 78% of leucine and valine taken up, respectively. Respiration of 3H and 14C isotopes of leucine gave similar results. Most of the nonrespired leucine was recovered in bacterial proteins, while only up to one-half of the nonrespired valine occurred in proteins. In intracellular pools of the bacteria, [3H]leucine reached an isotope saturation of 88 to 100% at concentrations of >40 nM. For [3H]valine, an isotope equilibrium of about 90% was obtained at concentrations of >80 nM. Within an incubation period of typically 1 h, tritiated leucine and valine incorporated into proteins of the bacteria reached an isotope saturation of 2 to 6%. In a 99-h batch experiment, bacterial protein synthesis calculated from incorporation of leucine and valine corresponded to 31 and 51% (10 nM) and 89 and 97% (100 nM), respectively, of the chemically determined protein production. Measured conversion factors of 100 nM leucine and valine were 6.4 × 1016 and 6.6 × 1016 cells per mol, respectively, and fell within the expected theoretical values. The present study demonstrates that incorporation of both valine and leucine produces realistic measurements of protein synthesis in freshwater bacteria and that the incorporation can be used as a measure of bacterial production.  相似文献

Nitrogen and oxygen transformations were studied in a bioturbated (reworked by animals) estuarine sediment (Norsminde Fjord, Denmark) by using a combination of 15N isotope (NO3-), specific inhibitor (C2H2), and microsensor (N2O and O2) techniques in a continuous-flow core system. The estuarine water was NO3- rich (125 to 600 μM), and NO3- was consistently taken up by the sediment on the four occasions studied. Total NO3- uptake (3.6 to 34.0 mmol of N m-2 day-1) corresponded closely to N2 production (denitrification) during the experimental steady state, which indicated that dissimilatory, as well as assimilatory, NO3- reduction to NH4+ was insignificant. When C2H2 was applied in the flow system, denitrification measured as N2O production was often less (58 to 100%) than the NO3- uptake because of incomplete inhibition of N2O reduction. The NO3- formed by nitrification and not immediately denitrified but released to the overlying water, uncoupled nitrification, was calculated both from 15NO3- dilution and from changes in NO3- uptake before and after C2H2 addition. These two approaches gave similar results, with rates ranging between 0 and 8.1 mmol of N m-2 day-1 on the four occasions. Attempts to measure total nitrification activity by the difference between NH4+ fluxes before and after C2H2 addition failed because of non-steady-state NH4+ fluxes. The vertical distribution of denitrification and oxygen consumption was studied by use of N2O and O2 microelectrodes. The N2O profiles measured during the experimental steady state were often irregularly shaped, and the buildup of N2O after C2H2 was added was much too fast to be described by a simple diffusion model. Only bioturbation by a dense population of infauna could explain these observations. This was corroborated by the relationship between diffusive and total fluxes, which showed that only 19 to 36 and 29 to 62% of the total O2 uptake and denitrification, respectively, were due to diffusion-reaction processes at the regular sediment surface, excluding animal burrows.  相似文献

Summary The seasonal variation in frequency of birth of 297 patients with karyotypes 47,XXY, 47,XYY, 47,XXX and 45,X registered in the Danish Cytogenetic Central Register until January 1st, 1971, was significantly different from the variation expected. The frequency of conception was higher than expected in June, July and August and lower in September, October and December. The variation in frequency of birth of the 791 patients with trisomy 21 was not significantly different from the expected value.There was no coincidence between the peak frequency of conception of children with aneuploid sex chromosome abnormalities and the peak frequency of a number of infectious diseases.It is concluded that the cause of the seasonal variation in frequency of conception and birth of children with aneuploid sex chromosome abnormalities is unknown. A number of possible factors are mentioned.
Zusammenfassung Bei den 297 Patienten mit Karyotypen 47,XXY, 47,XYY, 47,XXX und 45,X, die bis zum 1. Januar 1971 im Dänischen Cytogenetischen Centralergister registriert worden sind, unterschied sich die jahreszeitliche Variation in der Geburtenhäufigkeit signifikant von der erwarteten Variation. Die Konzeptionshäufigkeit war in den Monaten Juni, Juli und August erhöht und im September, Oktober und Dezember erniedrigt, wohingegen die Variation in der Geburtenhäufigkeit von 791 Patienten mit Karyotypen 47,XX order XY,+21 Abnormitäten nicht signifikant unterschiedlich von der erwarteten Variation war.Es fand sich kein Zusammentreffen zwischen dem Häufigkeitsgipfel der Konzeption von Kindern mit aneuploiden Geschlechtschromosomenabnormitäten und dem Gipfel einer Reihe von Infektionskrankheiten.Daraus wird geschlossen, daß die Ätiologie der jahreszeitlichen Variation der Konzeptions-häufigkeit und Geburten von Kindern mit aneuploiden Geschlechtschromosomenabnormitäten unbekannt ist. Eine Reihe möglicher Faktoren wird erwähnt.
