The KefB and KefC systems of Escherichia coli cells are activated by iodoacetate (IOA) and chlorodinitrobenzene (CDNB), leading to a rapid drop in the intracellular pH. However, survival of exposure to IOA or CDNB was found to be essentially independent of KefB and KefC activation. No correlation was found between the toxicity of the compound and its ability to elicit protective acidification via activation of KefB and KefC. 相似文献

The initial steps in taste and olfaction result from the activation by
chemical stimuli of taste receptor cells (TRCs) and olfactory receptor
neurons (ORNs). In parallel with these two pathways is the chemosensitive
trigeminal pathway whose neurons terminate in the oral and nasal cavities
and which are activated by many of the same chemical stimuli that activate
TRCs and ORNs. In a recent single unit study we investigated the responses
of rat chorda tympani and glossopharnygeal neurons to a variety of
bitter-tasting alkaloids, including nicotine, yohimbine, quinine,
strychnine and caffeine, as well as capsaicin, the pungent ingredient in
hot pepper. Here we apply many of these same compounds to cultured rat
trigeminal ganglion (TG) neurons and measure changes in intracellular
calcium [Ca2+]i to determine whether TG neurons will respond to these same
compounds. Of the 89 neurons tested, 34% responded to 1 mM nicotine, 7% to
1 mM caffeine, 5% to 1 mM denatonium benzoate, 22% to 1 mM quinine
hydrochloride, 18% to 1 mM strychnine and 55% to 1 microM capsaicin. These
data suggest that neurons from the TG respond to the same bitter-tasting
chemical stimuli as do TRCs and are likely to contribute information sent
to the higher CNS regarding the perception of bitter/irritating chemical
stimuli.

The histological effect on and stress response of post juvenile Clarias gariepinus exposed to Qua Iboe crude oil and rig wash were investigated. Fish weighing 60–90 g and measuring 16–18 cm were exposed for 7–28 days to 8.00 ml?1 Qua Iboe crude oil and 0.0018 ml–1 rig wash, both being 0.1 of the 96 hr LC50. Blood samples of C. gariepinus were collected every seven days and evaluated for stress by measuring cortisol concentration. The gills and liver were studied and scored for Gill Alteration Index (GAI) and Hepatic Alteration Index (HAI), respectively. There was an increase in cortisol level up to the 7th and 14th day among the group exposed to Qua Iboe crude oil, with a decrease on the 21st and 28th day. The rig wash group increased in cortisol level up to the 7th day and decreased slightly on the 14th day, after which the trend became irregular. The toxic effects of the Qua Iboe crude oil and rig wash were time dependent, as shown by the histopathological alteration index (HAI) of gill and liver. After 28 days of exposure, the gills had irreparable damage due to high frequency of cellular necrosis and degeneration, whereas the liver had from moderate to severe damage due to the high frequency of cellular degeneration and inflammation. Qua Iboe crude oil and rig wash are both toxic to C. gariepinus, therefore their indiscriminate discharge to the environment must be discouraged. 相似文献

Plastid sequencing is an essential tool in the study of plant evolution. This high‐copy organelle is one of the most technically accessible regions of the genome, and its sequence conservation makes it a valuable region for comparative genome evolution, phylogenetic analysis and population studies. Here, we discuss recent innovations and approaches for de novo plastid assembly that harness genomic tools. We focus on technical developments including low‐cost sequence library preparation approaches for genome skimming, enrichment via hybrid baits and methylation‐sensitive capture, sequence platforms with higher read outputs and longer read lengths, and automated tools for assembly. These developments allow for a much more streamlined assembly than via conventional short‐range PCR. Although newer methods make complete plastid sequencing possible for any land plant or green alga, there are still challenges for producing finished plastomes particularly from herbarium material or from structurally divergent plastids such as those of parasitic plants. 相似文献

Evolutionary transitions from outcrossing to selfing occur commonly in heterostylous genera. The morphological polymorphisms that characterize heterostyly provide opportunities for different pathways for selfing to evolve. Here, we investigate the origins and pathways by which selfing has evolved in tristylous Eichhornia paniculata by providing new evidence based on morphology, DNA sequences and genetic analysis. The primary pathway from outcrossing to selfing involves the stochastic loss of the short-styled morph (S-morph) from trimorphic populations, followed by the spread of selfing variants of the mid-styled morph (M-morph). However, the discovery of selfing variants of the long-styled morph (L-morph) in Central America indicates a secondary pathway and distinct origin for selfing. Comparisons of multi-locus nucleotide sequences from 27 populations sampled from throughout the geographical range suggest multiple transitions to selfing. Genetic analysis of selfing variants of the L- and M-morphs demonstrates recessive control of the loss of herkogamy, although the number of factors appears to differ between the forms. Early stages in the establishment of selfing involve developmental instability in the formation of flowers capable of autonomous self-pollination. The relatively simple genetic control of herkogamy reduction and frequent colonizing episodes may often create demographic conditions favouring transitions to selfing in E. paniculata . 相似文献

Ant‐dispersed herbs (myrmecochores) can account for more than one‐third of the stems in the temperate deciduous forests of eastern North America. Because many ant species have been observed collecting the seeds, this interaction is often described as a generalized mutualism. Here, we combine fieldwork and meta‐analyses to test this assumption. Our meta‐analysis demonstrated that Aphaenogaster ants (predominantly A. rudis) collect approximately 74±26% (mean±SD) of the myrmecochorous seeds in eastern North American forests where any encounters with Aphaenogaster were reported, and approximately 61±37% of the seeds in all the eastern forests where any seed collection has been monitored. This remarkable monopolization of seeds is due to at least two factors: 1) Aphaenogaster are significantly more likely to collect the ant‐adapted seeds they discover than are ten other ant genera found in these forests and 2) the densities of Aphaenogaster and myrmecochorous plants are positively correlated at three nested spatial scales (within 20×20 m patches, among patches within a forest, and among 41 forests in the eastern United States). Although other ants can collect seeds, our analyses demonstrate that A. rudis is the primary seed dispersal vector for most of this rich temperate ant‐dispersed flora. The low levels of plant partner diversity for myrmecochores demonstrated here rivals that of tropical ant‐plants (myrmecophytes) and well exceeds that typically observed in temperate plant–frugivore and plant–pollinator mutualisms and myrmecochory in other biomes. 相似文献
