In addition to the characterized mechanisms responsible for many direct effects of plant growth promoting bacteria (PGPB) on plants, it has been suggested that a number of PGPB contain the enzyme ACC deaminase that catalyzes degradation of 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of ethylene, into α-ketobutyrate and ammonia. As part of an effort to obtain an ACC deaminase encoding gene from a collection of soil samples, only one bacterial isolate, Pseudomonas fluorescens FY32 was capable of growing on ACC as a sole source of nitrogen. The ACC deaminase gene was amplified from the above isolate by polymerase chain reaction (PCR) giving an expected DNA fragment, 1017 bp. Sequence analysis of the fragment showed that it was highly homologous (94% and 98% identities at nucleotide and amino acid levels, respectively) to the previously characterized acdS gene from Pseudomonas sp. 6G5. Furthermore, fusion of the ACC deaminase ORF with lacZ gene resulted in the expression of active enzyme in Escherichia coli. In addition, further analyses revealed that the acdS gene was plasmid-encoded so that a large plasmid (pFY32) with almost 50 kb in size was identified from this bacterium. Furthermore, transfer of pFY32 into E. coli DH5α proved its ACC deaminase activity. This result was in accordance with previous reports suggesting horizontal transfer of the acdS gene. However, it needs more investigation to identify whether this pFY32 plasmid has undergone lateral gene transfer during the evolutionary process.  相似文献

Beauveria bassiana is a filamentous ascomycete that is pathogenic towards a broad host range of insect targets and is increasingly serving as a model for examining fungal development and host-pathogen interactions. B. bassiana displays a prohibitive level of resistance against many current fungal and/or yeast selection markers including hygromycin, neomycin, and zeocin. A genetic transformation system for B. bassiana based upon the use of a sulfonylurea resistance cassette derived from the Magnaporthe grisea, acetolactate synthase gene (sur) was developed. The transformation frequency ranged from 100–150 transformants per microgram DNA/108 cells and Southern blot analysis indicated that the plasmid vector was randomly integrated into the genome of B. bassiana. In addition, a construct bearing the sur gene and the enhanced green fluorescent protein gene egfp as a visual marker was used to successfully transform B. bassiana. Over 95% of the transformants retained the sulfonylurea resistance phenotype under non-selective conditions. The described transformation method increases opportunities for the genetic manipulation of B. bassiana.  相似文献

Modulation of TRPV1 by nonreceptor tyrosine kinase, c-Src kinase   总被引:5,自引:0,他引:5  
The capsaicin receptor TRPV1 is a nonselective cation channel that is expressed in sensory neurons. In this study, we examined the role of the nonreceptor cellular tyrosine kinase c-Src kinase in the modulation of the rat TRPV1. Capsaicin-induced currents in identified colonic dorsal root ganglion neurons were blocked by the c-Src kinase inhibitor PP2 and enhanced by the tyrosine phosphatase inhibitor sodium orthovandate. PP2 also abolished currents in human embryonic kidney-293 cells transfected with rat TRPV1, whereas cotransfection of TRPV1 with v-Src resulted in fivefold increase in capsaicin-induced currents. In cells transfected with dominant-negative c-Src and TRPV1, capsaicin-induced currents were decreased by approximately fourfold. TRPV1 co-immunoprecipitated with Src kinase and was tyrosine phosphorylated. These studies demonstrate that TRPV1 is a potential target for cellular tyrosine kinase-dependent phosphorylation.  相似文献

Background  

Tryptophan-pathway genes that exist within an apparent operon-like organization were evaluated as examples of multi-genic genomic regions that contain phylogenetically incongruous genes and coexist with genes outside the operon that are congruous. A seven-gene cluster in Xylella fastidiosa includes genes encoding the two subunits of anthranilate synthase, an aryl-CoA synthetase, and trpR. A second gene block, present in the Anabaena/Nostoc lineage, but not in other cyanobacteria, contains a near-complete tryptophan operon nested within an apparent supraoperon containing other aromatic-pathway genes.  相似文献

In Egypt, the chalk brood (CHB) disease caused by the fungus, Ascosphaera apis Maassen started again infesting the honeybee, Apis melifera L (Hymenoptera: Apidae) colonies after a cessation pause of seven to nine years. For the first time, an attempt has been made to look into the problem of the CHB disease from the view point of assessing losses in both clover (Trifolium alexandrinum) and chinus (Chinus molus) honey yields. In this regard, two techniques were adopted. Under the natural conditions of the experimental apiary (first technique), loss in clover honey was 18.412 +/- 0.663%, on average, whereas the average loss in chinus honey was 18.332 +/- 1.536%. In the second technique of loss appraisal (artificial infection). The tested honeybee colonies were artificially infected with four sources of A. apis infection, I.e. black mummies, white mummies, pollen grains and water, in addition to controls to create the so-called "different levels of infection". The mean percentages of losses in clover honey were 30.06 +/- 1.807, 27.95 +/- 1.062; 21.13 +/- 0.987; 16.96 +/- 0.672 and 0.00 +/- 0.00 for black mummies, white mummies, pollen grains, water and control, respectively. Taking into account the relationship between number of resulted mummies in each source of infection and % loss in clover honey, it could be concluded that as the resulted mummies increased, the corresponding clover honey yield decreased in each source of infection. It was noticed that the % loss in clover honey differed as the used technique differed. For example, the percentage loss in clover honey produced from colonies exposed to the natural conditions was relatively less than that of the artificially infected ones. This has been discussed in the text. However, the causative pathogen of CHB disease resulted in serious decrease in honey production. Loss appraisal is a perquisite step for the determination of the economic injury levels (EILs). By regressing % loss in clover honey yield against the total number of fallen mummies, the linear equation was worked out to be: Y = 0.242 + 0.040 x (r2 = 0.99, F = 62408.865). Depending on the total costs of controlling CHB disease, the EIL values were 31.875 and 11.250 mummies/colony for cedar oil and thymol granules, subsequently. As to the chinus honey the corresponding EILs were 18.940 and 6.683 mummies/colony for cedar oil and thymol granules, in respect, as a result of adopting the regression formula: Y = -6.762 + 0.101 X. As for the clover honey (artificial infection) the EIL values were 39.844 and 14.063 mummies/ colony in case of using cedar oil and thymol grains, subsequently. These values were worked out using the formula Y = 5.871 + 0.032 X. From the practical point of view, apiarists should not use thymol or cedar oil as fallen mummies don't reach the suggested EILs values to minimize control costs as much as possible.  相似文献

Background

Despite evidence supporting the importance of breastfeeding to child health, breastfeeding practices remain suboptimal in Vietnam. There is currently little evidence on the importance of breastfeeding in the prevention of morbidity during infancy in Vietnam. In order to provide country specific data for policy makers to support breastfeeding friendly policies and programs, analysis was undertaken on a cross-sectional dataset to investigate the association between breastfeeding practices and prevalence of diarrhea and acute respiratory infection (ARI) among infants aged 0–5 months.

Methods

Data on socio-demographic characteristics, infant feeding practices and prevalence of diarrhea and ARI were obtained from 6,068 mother-child dyads in 11 provinces of Vietnam in 2011. Multivariate logistic regression was used to examine the associations between breastfeeding practices and child illnesses.

Results

On average, the prevalence of diarrhea and ARI among infants 0–5 months was 5.3% and 24.5%, respectively. Though half of all infants were breastfed within one hour of birth, 73.3% were given prelacteal foods in the first three days after birth. Only 20.2% of children 0–5 months old were exclusively breastfed, while 32.4% were predominantly breastfed and 47.4% partially breastfed. After adjusting for confounders, early initiation of breastfeeding was associated with lower prevalence of diarrhea [adjusted odds ratio (AOR)?=?0.74 (95% CI 0.58, 0.93)], while prelacteal feeding was associated with higher prevalence [AOR?=?1.53 (95% CI 1.15, 2.03)]. Compared to infants who were exclusively breastfed, infants who were predominantly [AOR?=?1.52 (95% CI 1.05, 2.21)] or partially breastfed [AOR?=?1.55 (95% CI 1.07, 2.24)] were more likely to have diarrhea. Prelacteal feeding [AOR?=?1.16 (95% CI 1.01, 1.33)] and partial breastfeeding [AOR relative to exclusive breastfeeding?=?1.24 (95% CI 1.03, 1.48)] were associated with higher prevalence of ARI. While the protective effects of exclusive breastfeeding against diarrhea declined with child age, this effect for ARI appears to have remained constant.

Conclusions

Early initiation and exclusive breastfeeding protects against diarrhea and ARI. Results confirm that interventions to improve early and exclusive breastfeeding would contribute to improving child health and nutrition in Vietnam.

Hydrophobins represent a class of unique fungal proteins that have low molecular mass, are cysteine rich, and can self-assemble into two-dimensional arrays at water/air interfaces. These highly surface-active proteins are able to decrease the surface tension of water, thus allowing fungal structures to penetrate hydrophobic–hydrophilic barriers. Due to their unusual biophysical properties, hydrophobins have been suggested for use in a wide range of biotechnological applications. Here we describe a simple method for producing a functionally active class I hydrophobin derived from the entomopathogenic fungus, Beauveria bassiana, in an E. coli host. N-terminal modifications were required for proper expression and purification, and the hydrophobin was expressed as a fusion partner to a cleavable N-terminus chitin-binding domain-intein construct. The protein was purified and reconstituted from E. coli inclusion bodies. Self-assembly of the recombinant hydrophobin was followed kinetically using a thioflavin T fluorescence binding assay, and contact angle measurements of purified recombinant hydrophobin protein (mHyd2) films on a variety of substrata demonstrated its surface modification ability, which remained stable for at least 4 months. Filament or fibril-like structures were imaged using atomic force and transmission electron microscopy. These data confirmed the functional properties of the purified protein and indicate amino acid flexibility at the N-terminus, which can be exploited for various applications of these proteins.  相似文献

A feeding study was conducted to determine the effect of dietary nucleotides (NT) on growth performance, immune responses and resistance of rainbow trout fingerlings to Streptococcus iniae. A basal diet supplemented with 0 (control), 0.5, 1, 1.5 and 2 g NT kg(-1) to formulate five experimental diets. Each diet was randomly allocated to triplicate groups of fish with initial average weight of approximately 23 g. After 8 weeks of feeding trial, fish fed diets with 1.5-2 g NT kg(-1) had the highest final weight and SGR, followed by groups fed the diets with 0.5-1 g NT kg(-1), and lowest in fish fed the NT-free control diet. The feed conversion ratio (FCR) was better when the fish were fed diet with 2 g NT kg(-1). The ACH(50) level and lysozyme activity in serum were found to be significantly (P < 0.05) greater in fish fed nucleotides at 1.5-2 g kg(-1). The fish fed with 2 g NT kg(-1) had higher IgM level. The challenge experiment showed that relative percent survival (RPS) increased significantly (P < 0.05) with increasing levels of nucleotides from 0.5 to 2 g kg(-1) and was lowest in the control group. We therefore recommend dietary nucleotide administration at 1.5-2 g kg(-1) to promote growth and enhance immunity and resistance against S. iniae.  相似文献

The entomogenous filamentous fungus, Beauveria bassiana expresses two hydrophobin genes, hyd1 and hyd2, hypothesized to be involved in cell surface hydrophobicity, adhesion, virulence, and to constitute the protective spore coat structure known as the rodlet layer. Targeted gene inactivation of hyd1 resulted in seemingly 'bald' conidia that contained significantly altered surface fascicles or bundles. These cells displayed decreased spore hydrophobicity, loss of water mediated dispersal, changes in surface carbohydrate epitopes and β-1,3-glucan distribution, lowered virulence in insect bioassays, but no effect on adhesion. In contrast, Δhyd2 mutants retained distorted surface bundles, but truncated/incomplete rodlets could be seen within the bundles. Δhyd2 conidia displayed both decreased cell surface hydrophobicity and adhesion, but the mutant was unaffected in virulence. The double Δhyd1Δhyd2 mutant was distinct from the single mutants, lacking both bundles and rodlets, and displaying additively decreased cell surface hydrophobicity, reduced cell attachment and lowered virulence than the Δhyd1 mutant. Epitope tagged constructs of the proteins were used to examine the expression and distribution of the proteins and to demonstrate the continued presence of Hyd2 in the Δhyd1 strain and vice versa. The implications of our results with respect to fascicle and rodlet assembly on the spore surface are discussed.  相似文献
