SAR studies of non-zinc-chelating MMP-13 inhibitors: Improving selectivity and metabolic stability

SAR studies to improve the selectivity and metabolic stability of a class of recently discovered MMP-13 inhibitors are reported. Improved selectivity was achieved by modifying interactions with the S1′ pocket. Metabolic stability was improved through reduction of inhibitor lipophilicity. This translated into lower in vivo clearance for the preferred compound.     

Asymmetric cell division in Mycobacterium tuberculosis and its unique features

Recently, several reports showed that about 80 % of mid-log phase Mycobacterium smegmatis, Mycobacterium marinum, and Mycobacterium bovis BCG cells divide symmetrically with 5–10 % deviation in the septum position from the median. However, the mode of cell division of the pathogenic mycobacterial species, Mycobacterium tuberculosis, remained unclear. Therefore, in the present study, using electron microscopy, fluorescence microscopy of septum- and nucleoid-stained live and fixed cells, and live cell time-lapse imaging, we show the occurrence of asymmetric cell division with unusually deviated septum/constriction in 20 % of the 15 % septating M. tuberculosis cells in the mid-log phase population. The remaining 80 % of the 15 % septating cells divided symmetrically but with 2–5 % deviation in the septum/constriction position, as reported for M. smegmatis, M. marinum, and M. bovis BCG cells. Both the long and the short portions of the asymmetrically dividing M. tuberculosis cells with unusually deviated septum contained nucleoids, thereby generating viable short and long cells from each asymmetric division. M. tuberculosis short cells were acid fast positive and, like the long cells, further readily underwent growth and division to generate micro-colony, thereby showing that they were neither mini cells, spores nor dormant forms of mycobacteria. The freshly diagnosed pulmonary tuberculosis patients’ sputum samples, which are known for the prevalence of oxidative stress conditions, also contained short cells at the same proportion as that in the mid-log phase population. The probable physiological significance of the generation of the short cells through unusually deviated asymmetric cell division is discussed.     

Three new species ofTrichogramma [Hymenoptera: Trichogrammatidae] from India

Three new species ofTrichogramma occurring in India are described. Two of these were reared from eggs of the sugarcane stem-borerChilotraea infuscatellus, while the third was found attacking eggs ofAchaea janata, Tiracola plagiata andHeliothis armigera. The diagnostic value of the male genitalia in this genus is emphasized and a discussion is presented justifying the consideration of these as new species.

Résumé Trois nouvelles espèces deTrichogramma présentes en Inde sont décrites. Deux d'entre elles ont été obtenues d'œufs du foreur des tiges de la canne à sucre,Chilotraea infuscatellus, la troisième s'attaque aux œufs deAchaea janata, Tiracola plagiata etHeliothis armigera. La valeur pour la diagnose des genitalia males dans ce genre est soulignée et les arguments en faveur d'espèces nouvelles sont discutés.

     

Analysis and separation of natural and synthetic mixtures of uroporphyrins by high-pressure liquid chromatography

A new method for the quantitative analysis of mixtures of the methyl esters of uroporphyrins I and III was developed; this can be applied both to the analysis of naturally occurring uroporphyrins and also to their semi-preparative isolation.     

Solvent effect on the relative quantum yield and fluorescence quenching of a newly synthesized coumarin derivative

We estimated the relative florescence quantum yield (Φ) of 8‐methoxy‐3‐[1‐(4,5‐dicarbomethoxy‐1,2,3‐triazoloacetyl)]coumarin [8MDTC] using a single‐point method with quinine sulfate in 0.1 M of sulfuric acid used as a standard reference. The fluorescence lifetimes, radiative and non‐radiative decay rate constants are calculated. Relative quantum yields were found to be less in the non‐polar solvents, indicating that the solute exhibits less fluorescence in a non‐polar environment. The fluorescence quenching of [8MDTC] by aniline was studied at room temperature by examining the steady state in five different solvents in order to explore various possible quenching mechanisms. The experimental results show a positive deviation in Stern–Volmer plots in all solvents. Ground state complex and sphere of action static quenching models were used to interpret the results. Many quenching rate parameters were calculated using these models. The values of these parameters suggest that the sphere of action static quenching model agrees well with the experimental results. Further, a finite sink approximation model was used to check whether these bimolecular reactions were diffusion limited or not. The values of the distance parameter R′ and the diffusion coefficient D were determined and are compared with the values of the encounter distance R and diffusion coefficient D calculated using the Stokes–Einstein equation. Antifeeding and insect-growth-regulating activity of certain metal complexes towardsSpodoptera litura; F

Metal complexes of divalent cobalt, nickel and copper and trivalent iron were synthesized usingN-salicylidene-3-aminocoumarin as chelating agent. The ligand behaves as a monobasic ONO donor towards Co(II), Ni(II) and Cu(II) and as an ON as well as an ONO donor towards Fe(III). All the complexes haven been proposed to have octahedral geometry on the basis of analytical, thermal conductivity, spectral and magnetic data. The complexes have been screened againstSpodoptera litura;F (Lepidoptera: noctuiidae) for antifeeding and insect-growth-regulating activity. The results show appreciable insect-growth-regulating activity associated with metal complexation.
