—There are three histological layers within the rat olfactory tubercle: plexiform, pyramidal and polymorphic. We have assayed glutamic acid decarboxylase (GAD) and GABA in homogenates of frozen sections cut parallel to these layers. Consecutive sections (16 μm) were homogenized in groups and assayed for GAD or GABA. Every seventh section was stained with Toluidine Blue to monitor the depth and orientation of the plane of section. Steep variations in GAD activity (up to 6-fold) were observed as a function of depth in the tubercle. These were paralleled by corresponding but less marked variations in GABA levels. The lowest values were found in the plexiform layer. This suggests that GAD may play a very limited role there. The highest activities were found in sections from the deepest lamina of the polymorphic layer. This is the only lamina of the tubercle that does not receive a dopaminergic input.  相似文献

Increasing drought frequency is a major driver of changes in forest structure and has been implicated in the decline of the endangered tree species, Eucalyptus gunnii ssp. divaricata (McAulay & Brett) in the Central Plateau region of Tasmania, Australia. In this study, we examined patterns of regeneration, aspects of the water relations of E. gunnii ssp. divaricata and its replacement Eucalyptus pauciflora and, whether shifts in stand dominance have occurred where the subspecies co‐occurs with E. pauciflora could be related to recent changes in climate. Successful E. gunnii ssp. divaricata seedling regeneration was restricted to micro‐sites with relatively deep soils within slight depressions. In contrast, poor E. gunnii ssp. divaricata regeneration and declining adult cohorts of this species all occurred on steeper, concave micro‐sites with shallow soils. This apparent shift in suitable regeneration micro‐site, from sites with shallow to deeper soils, may be linked to an observed 25% reduction in summer rainfall over the last 50 years. On slopes surrounding waterlogged depressions where E. gunnii ssp. divaricata co‐occurs with E. pauciflora, E. pauciflora was in higher abundance than E. gunnii ssp. divaricata in small adult and sapling size‐classes, compared with the adult cohorts (>30 cm d.b.h.), a trend consistent with a shift in stand dominance. Despite existing paradigms related to differential drought tolerance between these two species as a driver of this shift in stand dominance, there were no differences in predawn (Ψpd) water potentials between species. Furthermore, pressure–volume analysis showed that E. gunnii ssp. divaricata had lower values for osmotic potential at turgor loss point (?2.33 ± 0.06 MPa) than E. pauciflora (?2.13 ± 0.03 MPa), suggesting that E. gunnii ssp. divaricata may be more drought tolerant than E. pauciflora, in contrast to the prevailing paradigm that it is more susceptible to drought than E. pauciflora.  相似文献

Investigating the many internal feedbacks within the climate system is a vital component of the effort to quantify the full effects of future anthropogenic climate change. The stomatal apertures of plants tend to close and decrease in number under elevated CO2 concentrations, increasing water‐use efficiency (WUE) and reducing canopy evapotranspiration. Experimental and modelling studies reveal huge variations in these changes such that the warming associated with reduced evapotranspiration (known as physiological forcing) is neither well understood or constrained. Palaeo‐observations of changes in stomatal response and plant WUE under rising CO2 might be used to better understand the processes underlying the physiological forcing feedback and to link measured changes in plant WUE to a specific physiological change in stomata. Here we use time series of tree ring (Pinus sylvestris L.) δ13C and subfossil leaf (Betula nana L.) measurements of stomatal density and geometry to derive records of changes in intrinsic water‐use efficiency (iWUE) and maximum stomatal conductance in the Boreal zone of northern Finland and Sweden. We investigate the rate of change in both proxies, over the recent past. The independent lines of evidence from these two different Boreal species indicate increased iWUE and reduced maximum stomatal conductance of similar magnitude from preindustrial times (ca. ad 1850) to around ad 1970. After this maximum stomatal conductance continues to decrease to ad 2000 in B. nana but iWUE in P. sylvestris reaches a plateau. We suggest that northern boreal P. sylvestris might have reached a threshold in its ability to increase WUE as CO2 rises.  相似文献

With current annual production at over 600 million tonnes, wheat is the third largest crop in the world behind corn and rice, and an essential source of carbohydrates for millions of people. While wheat is grown over a wide range of environments, it is common in the major wheat‐producing countries for grain filling to occur when soil moisture is declining and temperature is increasing. Average global temperatures have increased over the last decades and are predicted to continue rising, along with a greater frequency of extremely hot days. Such events have already been reported for major wheat growing regions in the world. However, the direct impact of past temperature variability and changes in averages and extremes on wheat production has not been quantified. Attributing changes in observed yields over recent decades to a single factor such as temperature is not possible due to the confounding effects of other factors. By using simulation modelling, we were able to separate the impact of temperature from other factors and show that the effect of temperature on wheat production has been underestimated. Surprisingly, observed variations in average growing‐season temperatures of ±2 °C in the main wheat growing regions of Australia can cause reductions in grain production of up to 50%. Most of this can be attributed to increased leaf senescence as a result of temperatures >34 °C. Temperature conditions during grain filling in the major wheat growing regions of the world are similar to the Australian conditions during grain filling. With average temperatures and the frequency of heat events projected to increase world‐wide with global warming, yield reductions due to higher temperatures during the important grain‐filling stage alone could substantially undermine future global food security. Adaptation strategies need to be considered now to prevent substantial yield losses in wheat from increasing future heat stress.  相似文献

1. We investigate long‐term (>200 years) changes to the composition and spatial structure of macrophyte communities in a shallow, eutrophic lake (Barton Broad, eastern England) and consider the implications for lake restoration. 2. Historical macrophyte data were assembled from a variety of sources: existing plant databases, museum herbaria, journal articles, old photographs and eyewitness accounts. Additionally, two types of sediment core sample were analysed for plant macro‐remains and pollen; bulk basal samples from multiple core sites analysed to provide information on ‘pre‐disturbance’ macrophyte communities and two whole cores analysed to determine historical change. 3. Prior to the late 1800s, macrophyte communities were diverse and included a multilayered mosaic of short‐stature submerged taxa and taller submerged and floating‐leaved species. With the progression of eutrophication after around 1900, the former community was displaced by the latter. Diversity was maintained, however, since an encroaching Schoenoplectus–nymphaeid swamp generated extensive patches of low‐energy habitat affording refugia for several macrophytes otherwise unable to withstand the hydraulic forces associated with open water conditions. When this swamp vegetation disappeared in the 1950s, many of the ‘dependent’ aquatic macrophytes also declined leaving behind a sparse, species‐poor community (as today) resilient to both eutrophication and turbulent open waters. 4. The combination of historical and palaeolimnological data sources offers considerable benefits for reconstructing past changes to the aquatic vegetation of lakes and for setting restoration goals. In this respect, our study suggests that successful restoration might often be better judged by reinstatement of the characteristic structure of plant communities than the fine detail of species lists; when nutrients are low and the structure is right, the right species will follow.  相似文献

Facultative associations are commonly encountered between ants and lycaenids, although the nature and patterns of associations are typically unclear. This study investigated a facultative symbiosis involving the lycaenid Theclinesthes albocincta (Lycaenidae), its host plant Adriana quadripartita and Australian native ants. Ants in the genera Ochetellus and Iridomyrmex were most frequently found in association with T. albocincta larvae, although Iridomyrmex ants were found in much lower abundance than were ants in Ochetellus. The abundances of Ochetellus and Iridomyrmex were highly correlated with larval abundance, but not egg abundance. Observations and experiments recorded oviposi- tion on male inflorescences on more than 95% of occasions, but oviposition was not greater on inflorescences with ants present. Behavioral assays showed that Iridomyrmex ants were aggressive towards female butterflies on significantly more occasions than were Ochetellus ants. These findings indicate potential evolutionary relationships between T. albocincta and two genera of ants that were abundant within the habitat.  相似文献

ABSTRACT We used nucleotide sequence data from a mitochondrial DNA fragment to characterize variation within the endangered Lower Keys marsh rabbit (Sylvilagus palustris hefneri). We observed 5 unique mitochondrial haplotypes across different sampling sites in the Lower Florida Keys, USA. Based on the frequency of these haplotypes at different geographic locations and relationships among haplotypes, we observed 2 distinct clades or groups of sampling sites (western and eastern clades). These 2 groups showed low levels of gene flow. Regardless of their origin, marsh rabbits from the Lower Florida Keys can be separated into 2 genetically distinct management units, which should be considered prior to implementation of translocations as a means of offsetting recent population declines.  相似文献

In vitro melanocyte-stimulating hormone (MSH) stimulates melanogenesis in some, but not all, melanocytes and melanoma cells. In an attempt to explain this variation in response to αMSH, we examined cyclic adenosine monophosphate (cAMP) accumulation, tyrosinase activity, and melanin production in primary (1°) murine B16 melanoma cells and in two B16 cell lines (B16 F1 and B16 F10) that are known to respond to αMSH. In vivo all three B16 melanoma cell types produced pigmented tumours. In vitro αMSH increased tyrosinase activity and melanin content in the F1 and F10 cells but not in the B16 1° cells. αMSH, however, increased cAMP production in all three cell types, confirming that the inability of B16 1° cells to produce melanin in response to αMSH is not due to a lack of αMSH receptors or cAMP response to αMSH. Further, we present evidence for a separate pathway of melanogenesis that is independent of cAMP as calmodulin antagonists, which do not elevate cAMP, increased tyrosinase activity, and melanin production in both 1° and F1 cells.  相似文献
