Progress in metabolomic analysis now allows the evaluation of food quality. This study aims to identify the metabolites in meat from livestock using a metabolomic approach. Using gas chromatography–mass spectrometry (GC/MS), many metabolites were reproducibly detected in meats, and distinct differences between livestock species (cattle, pigs, and chickens) were indicated. A comparison of metabolites between tissues types (muscle, intramuscular fat, and intermuscular fat) in marbled beef of Japanese Black cattle revealed that most metabolites are abundant in the muscle tissue. Several metabolites (medium-chain fatty acids, etc.) involved in triacylglycerol synthesis were uniquely detected in fat tissue. Additionally, the results of multivariate analysis suggest that GC/MS analysis of metabolites can distinguish between cattle breeds. These results provide useful information for the analysis of meat quality using GC/MS-based metabolomic analysis.

Ancestral amino acid residues were inferred for 3-isopropylmalate dehydrogenase (IPMDH), and were introduced into the enzyme of an extreme thermophile, Sulfolobus sp. strain 7. The thermostability of the mutant enzymes was compared with that of the wild type enzyme. At least five of the seven mutants tested showed higher thermal stability than the wild type IPMDH. The results are compatible with the hyperthermophilic universal ancestor hypothesis. The results also provide a new method for designing thermostable enzymes. The method only relies on the first dimensional structures of homologous enzymes that can be obtained from genetic databases.  相似文献

The relaxation effect of cilostazol, a phosphodiesterase III inhibitor, on the thoracic aorta was investigated. Cilostazol induced the relaxation of the thoracic aorta precontracted by phenylephrine in a concentration-dependent manner. The concentration-dependent relaxation was shifted to the right in the endothelium denuded aorta compared with that of intact endothelium, suggesting that this relaxation was partly dependent on endothelium. Cilostazol-induced relaxation of thoracic aorta tone was reversed by treatment with N(G)-nitro L-arginine (L-NNA), a competitive inhibitor of nitric oxide (NO) synthase. Cilostazol also significantly increased the NO level in the porcine thoracic aorta. In rats treated with cilostazol, the urinary excretion of nitrites, a stable metabolite of NO, and basal production of NO of the aortic ring were significantly greater than in those without treatment. These findings indicate that cilostazol-induced vasodilation of the rat thoracic aorta was dependent on the endothelium, which released NO from aortic endothelial cells.  相似文献

Microtubule protein and 30-S dynein (an axonemal ATPase protein) were prepared from cilia of Tetrahymena pyriformis and their mutual interaction was investigated. Microtubular protein stimulated the ATPase activity of 30-S dynein. Spinach chloroplasts in the light and in the dark were treated with several reagents for protein modification to see the effect of light on their resistivity toward modification. The reagents were p-diazobenzenesulfonic acid, diazonium-1-H-tetrazole, sodium-2,4,6-trinitrobenzenesulfonate, sodium-β-naphtoquinone-4,6-disulfonate and diazonium-1,2,4-triazole. No difference in the absorption spectrum was found between chloroplasts treated with these reagents in the light and those treated in the dark. However, these light- and dark-treated samples when solubilized with a nonionic detergent showed a difference in turbidity. Diazonium-1,2,4-triazole was the most suitable of the above reagents, and the solubilized sample of chloroplasts treated with diazonium-1,2,4-triazole in the light showed a turbidity which was about 2-fold higher than that of the same sample treated in the dark. This increase in turbidity was interpreted as being due to a change in the resistivity toward chemical modification of chloroplasts caused by illumination. In the presence of 3-(p-chlorophenyl)-1,1-dimethylurea, pentachlorophenol and 2-methylthio-4,6-bis-isopropylamino-s-triazine, which are inhibitors of the Hill reaction, the light-induced increase of turbidity was suppressed by 72, 78 and 62%, respectively. The addition of ATP caused a much greater increase of turbidity both in the light and in the dark. It was thus found that light and ATP induce a configurational change of chloroplasts or a conformational change of chloroplast proteins inside.  相似文献

The cell cycle inhibitor p21 plays an important role in monocytic cell differentiation, during which it translocates from the nucleus to cytoplasm. This process involves the negative regulation of the p21 nuclear localization signal (NLS). Here, we sought to determine the relationship between the cytoplasmic translocation of p21 and another molecule, Brap2, a cytoplasmic protein which binds the NLS of BRCA1 and was recently reported to inactivate KSR in the Ras-activating signal pathway under the name of IMP. We report that p21 and Brap2 directly interact, both in vitro and in vivo, in a manner requiring the NLS of p21 and the C-terminal portion of Brap2. When it is cotransfected with Brap2, p21 is expressed in the cytoplasm. Monocytic differentiation of the promyelomonocytic cell lines U937 and HL60 is associated with the upregulation of Brap2 expression concomitantly with the upregulation and cytoplasmic relocalization of p21. Our results underscore the role played by Brap2 in the process of cytoplasmic translocation of p21 during monocyte differentiation.  相似文献
