Background  

Many anticancer agents have poor water solubility and therefore the development of novel delivery systems for such molecules has received significant attention. Nanocarriers show great potential in delivering therapeutic agents into the targeted organs or cells and have recently emerged as a promising approach to cancer treatments. The objective of this experiment was to determine the effect of dietary grain on calcium homeostasis. Six rumen-fistulated dairy cows with 3 or more previous lactations and no history of parturient paresis were randomly assigned to a sequence of diets in a crossover study with 4 periods of 10 days each. Dietary treatments were: A control ration consisting of wrap grass silage alone (1), the control ration supplemented with ammonium chloride and ammonium sulphate salt solution (2), control ration following a period with supplementation (3) and control ration supplemented with increasing amounts of barley from 4 to 10 kg/cow per day, expected to produce subclinical rumen acidosis (4). Daily intake of the diets was adjusted to 14 kg DM/cow per day. On day 11, the calcium-regulating mechanisms in cows were challenged until recumbency by a standardized intravenous EDTA infusion and cows were left to recover spontaneously. Anion supplementation and the feeding of highly fermentable carbohydrate lowered urine pH below 7.0 due to subclinical acidosis. During spontaneous recovery from EDTA induced hypocalcaemia, the cows more quickly regained a whole blood free calcium concentration of 1.00 mmol/L if they had most recently been supplemented with either anionic salts or with increasing amounts of barley, as compared to the basic ration. It is concluded that so-called slug-feeding or 'steaming up' with highly fermentable carbohydrates before parturition in milk fever susceptible cows enhanced calcium homeostasis similar to the effect seen in cows on anionic diets.  相似文献

Sulfated polysaccharides (carrageenans and agars) are among the most important products of red algae that are used as food additives as well as in molecular biology research. The quality and value of the product is greatly dependent on the levels and sites of sulfation of the polysaccharides. Little information is currently available on the molecular details of sulfur metabolism in red algae. Considering the economic importance of sulfated polysaccharide, elucidating the molecular details of sulfur metabolism in these organisms could help in future endeavors to improve algal commercial value, e.g., through genetic engineering. A cDNA library from the red alga Porphyra purpurea (Roth) C. Agardh was used to isolate four cDNAs with homology to genes encoding known sulfur assimilation enzymes: sulfate adenyltransferase (ATP sulfurylase), adenosine 5′-phosphosulfate kinase (APSK), sulfite reductase, and cysteine synthase. These cDNAs were characterized with respect to their molecular properties and a cDNA with homology to APSK was used to functionally complement an Escherichia coli auxotroph APSK mutant. The other cDNAs are being similarly characterized with respect to their ability to produce functional enzymes. Elucidation of the regulation of expression of these genes will aid in future research to determine the biochemical and genetic details of the sulfate assimilation pathway as well as its genetic manipulation in red algae.  相似文献

On 15 November 2017 the mouth of the West Kleinemonde Estuary breached following heavy catchment rains and increased river flow. The water level in the estuary following mouth opening decreased by 1.65 m within 24 h, resulting in an almost complete draining of the littoral zone where large beds of the aquatic macrophyte Ruppia cirrhosa and mats of the associated filamentous algae were present. As the water depth within the plant beds decreased, the macrophytes, together with the algal filaments, created an increasingly dense mat, trapping fish that were resident, foraging or passing through the littoral zone. By 16 November 2017 large numbers of fishes belonging to at least 20 species were trapped in pools and depressions within the littoral, as well as within the R. cirrhosa beds and filamentous algal mats in the lower reaches of this system. Other affected taxa included crustaceans, especially isopods, and large numbers of small bivalves attached to macrophyte vegetation. Beneficiaries of the fish kill, in terms of unexpected food availability, included a variety of piscivorous bird species and the Cape clawless otter Aonyx capensis. This is the first documented account of a diverse species fish kill associated with estuary mouth breaching.  相似文献

The effect of starvation on the feeding rate of larval Chaoborus(Diptera. Chaoboridae) was investigated using Daphnia roseaas prey. The starvation period varied from 12 h to 22 days.The starved Chaoborus were individually incubated with 10 Daphniaunder controlled light and temperature conditions. Observationswere made on prey mortality every 2 h for the first 12 h andonce after 24 h. Feeding rates gradually increased to a maximumbetween 7–11 days of starvation. After this period, feedingrates declined to previous low levels. Generally, feeding rateswere significantly higher during the first 2–4 h of feeding.Thereafter, feeding rates were lower and exhibited no consistentpattems with length of feeding time.  相似文献

Background

Although Odisha is the largest contributor to the malaria burden in India, no systematic study has examined its malaria trends. Hence, the spatio-temporal trends in malaria in Odisha were assessed against the backdrop of the various anti-malaria strategies implemented in the state.

Methods

Using the district-wise malaria incidence and blood examination data (2003–2013) from the National Vector Borne Disease Control Program, blood examination-adjusted time-trends in malaria incidence were estimated and predicted for 2003–2013 and 2014–2016, respectively. An interrupted time series analysis using segmented regression was conducted to compare the disease trends between the pre (2003–2007) and post-intensification (2009–2013) periods. Key-informant interviews of state stakeholders were used to collect the information on the various anti-malaria strategies adopted in the state.

Results

The state annual malaria incidence declined from 10.82/1000 to 5.28/1000 during 2003–2013 (adjusted annual decline: -0.54/1000, 95% CI: -0.78 to -0.30). However, the annual blood examination rate remained almost unchanged from 11.25% to 11.77%. The keyinformants revealed that intensification of anti-malaria activities in 2008 led to a more rapid decline in malaria incidence during 2009–2013 as compared to that in 2003–2007 [adjusted decline: -0.83 (-1.30 to -0.37) and -0.27 (-0.41 to -0.13), respectively]. There was a significant difference in the two temporal slopes, i.e., -0.054 (-0.10 to -0.002, p = 0.04) per 1000 population per month, between these two periods, indicating almost a 200% greater decline in the post-intensification period. Although, the seven southern high-burden districts registered the highest decline, they continued to remain in that zone, thereby, making the achievement of malaria elimination (incidence <1/1000) unlikely by 2017.

Conclusion

The anti-malaria strategies in Odisha, especially their intensification since 2008, have helped improve its malaria situation in recent years. These successful measures need to be sustained and perhaps intensified further for eliminating malaria from Odisha.  相似文献

The function of membrane cholesterol (chol) in the regulation of membrane-bound hydrolases and transport proteins has been investigated in chol-enriched membranes of guinea pig intestinal brush borders. Chol-enrichment is accomplished by non-invasive means i.e., dietary manipulation by high-chol diet feeding. Activities of sucrase, lactase and maltase enzyme systems, Na+-dependent and -independent glucose transport and calcium uptake are found to be greatly inhibited by chol both at 22 degrees C and 37 degrees C. Glucose and calcium uptake in native membranes are found to be temperature sensitive processes and produce nonlinear Arrhenius plots with a transition temperature around 22 degrees C. The discontinuity in the Arrhenius expression is lost in chol enriched membranes which is interpreted as the increase in microviscosity imparted by chol in the bulk lipid phase environment where these proteins operate.  相似文献

A full factorial design crossing overstory (O) and understory (U) thinning and prescribed burning (B) was started at Teakettle Experimental Forest, California, in 2001 with the aim of achieving shifts in species composition to favor fire-resistant pines over fir. The goal of the present study was to evaluate the use of metabolic changes as early indicators for predicting the effects of these treatments on species-specific growth and long-term productivity of Pinus jeffreyi, Pinus lambertiana, and Abies concolor. Foliar polyamines, amino acids, soluble proteins and chlorophyll were quantified and related with foliar and soil chemistry. Most changes in soil chemistry occurred only with combined treatments (BU or BO). Changes in foliar metabolism were not driven by soil chemistry. In general, the pines were more sensitive to water stress as compared to fir as indicated by proline content. Understory thinning had no effect on any species. Both O and BO had species-specific effects on foliar amino acids, chlorophyll, soluble proteins, and inorganic ions. In addition, both O and BO were the most extreme thinning treatments for pines as they greatly increase the time needed to replenish carbon stocks. With the exception of some amino acids, B by itself had no major effects on foliar inorganic ions, chlorophyll content, and polyamines for any of the three species while BU caused a decrease in chlorophyll in white fir, suggesting a decline in photosynthetic capacity. These data point to BU as the best management option to specifically decrease the growth of fire-sensitive white fir without a major impact on pines.  相似文献

Distribution of biogenic amines—the diamine putrescine (Put), triamine spermidine (Spd), and tetraamine spermine (Spm)—differs between species with Put and Spd being particularly abundant and Spm the least abundant in plant cells. These amines are important for cell viability and their intracellular levels are tightly regulated, which have made it difficult to characterize individual effects of Put, Spd and Spm on plant growth and developmental processes. The recent transgenic intervention and mutational genetics have made it possible to stably alter levels of naturally occurring polyamines and study their biological effects. We bring together an analysis of certain metabolic changes, particularly in amino acids, to infer the responsive regulation brought about by increased diamine or polyamine levels in actively growing poplar cell cultures (transformed with mouse ornithine decarboxylase gene to accumulate high Put levels) and ripening tomato pericarp (transformed with yeast S-adenosylmethionine decarboxylase gene to accumulate high Spd and Spm levels at the cost of Put). Our analysis indicates that increased Put has little effect on increasing the levels of Spd and Spm, while Spd and Spm levels are inter-dependent. Further, Put levels were positively associated with Ala (α and β), Ile and GABA and negatively correlated with Gln and Glu in both actively growing poplar cell cultures and non-dividing tomato pericarp tissue. Most amino acids showed positive correlations with Spd and Spm levels in actively growing cells. Collectively these results suggest that Put is a negative regulator while Spd–Spm are positive regulators of cellular amino acid metabolism.  相似文献
