Conservation actions that have maintained populations in the past may not necessarily do so in the future. Population viability analysis provides one tool for exploring the impact of management actions on large temporal scales. However, there are relatively few long‐term data sets that provide the demographic and environmental data demanded by such models. Using a 37‐yr data set, we used RAMAS Metapop to model the persistence of natterjack toads Epidalea (Bufo) calamita on a heathland in southern Britain. A retrospective analysis showed that the best fit between the predicted population trajectories and the real population was when the management carried out was modelled as an increase in K of 150 toads yr?1. However, even if ongoing management continues to improve K by a further 40–60 toads yr?1 over the next 50 yr, the population still has an extinction risk of at least 60% if other factors remain unchanged. Sensitivity analyses and simulated management scenarios indicated that the population was most sensitive to changes in the survival of juvenile (i.e. 1–2 yr old) toads. In addition, if the frequency and severity of pond desiccation increases, the risk of extinction was predicted to increase as a result of reduced recruitment. Low levels of extinction risk occurred irrespective of K when juvenile survival was enhanced in combination with low frequency and severity of pond desiccation. The models suggest that populations that are responding to management against a background of natural fluctuations may remain vulnerable to extinction for several decades. These extinction risks may increase if habitat management fails to offset reductions in recruitment and juvenile survival caused by environmental change.  相似文献

Interactions with vimentin intermediate filaments (VimIFs) affect the motility, distribution, and anchorage of mitochondria. In cells lacking VimIFs or in which VimIF organization is disrupted, the motility of mitochondria is increased relative to control cells that express normal VimIF networks. Expression of wild-type VimIF in vimentin-null cells causes mitochondrial motility to return to normal (slower) rates. In contrast, expressing vimentin with mutations in the mid-region of the N-terminal non-α-helical domain (deletions of residues 41-96 or 45-70, or substitution of Pro-57 with Arg) did not inhibit mitochondrial motility even though these mutants retain their ability to assemble into VimIFs in vivo. It was also found that a vimentin peptide consisting of residues 41-94 localizes to mitochondria. Taken together, these data suggest that VimIFs bind directly or indirectly to mitochondria and anchor them within the cytoplasm.  相似文献

Mitochondria are the main source of energy in eukaryotic cells. They also play an important role in the number of other processes, such as regulation of calcium concentration and sequestration of apoptotic factors. Almost all functions of mitochondria depend on their ability to generate and maintain membrane potential by means of aerobic respiration. The level of mitochondrial potential is under the control of different inner and outer factors. However, mechanisms of this regulation are still poorly understood. In the present study we answer the question of how membrane potential of mitochondria depends on their motility. Using the potential-dependent dye MitoTracker Red, fluorescent microscopy of live cells, and the analysis of mitochondrial motility, two sub-populations of mitochondria were determined: (1) moving mitochondria transported along microtubules and (2) stationary mitochondria. We have shown that stationary mitochondria have higher membrane potential than moving mitochondria. It was also found that the level of potential of mitochondria is regulated by their interaction with vimentin intermediate filaments.  相似文献

A stable cell line CV-1 was obtained for vital observation of the transport of mitochondria in animals cells, which express a fragment of the resident protein of mitochondria marked by yellow fluorescent protein. The parameters and conditions of movement of the mitochondria in living cells were established using fluorescence videomicroscopy. Under the normal conditions, only a small part of mitochondria (ca. 7%) was transported over significant distances, while others were in the state of relative rest. The effective transport of mitochondria strictly depended on the dynamic properties of microtubules. Incubation of cell in a serum-free medium suppressed active transport of mitochondria, thus suggesting its dependence on certain, not yet determined environmental factors.  相似文献

Pulmonary acariasis is a sporadic, incidental finding in colony‐raised rhesus macaques (Macaca mulatta). Prophylactic treatment in indoor‐raised and indoor‐housed macaques is not routine due to low prevalence, lack of clinical significance, and potential risk of toxicosis. This case is an unusually severe infestation of Pneumonyssus simicola in an indoor‐housed rhesus macaque, which ultimately resulted in this animal's death.  相似文献

The paper presents an assessment of birch pollen seasons in Finland and Russia. The re-analysis covered the period from 1994 to 2005 and was focused on suspected long-range transport events that were recorded both in Moscow and at several Finnish sites. In order to trace the origin of airborne pollen before the onset of local pollination, we used both aerobiological and phenological observations combined with forward and adjoint (inverse) dispersion model simulations. It is shown that, although the Moscow region is surrounded by extensive birch forests, it still receives substantial amounts of foreign pollen before local pollination. In the Moscow region, the sources of long-range-transported pollen are in the south and south-west, sometimes even in the east. In Finland, there are frequently cases, before the local flowering season, in which Finnish territory receives Russian pollen; however in the opposite direction, from Finland to the Moscow region, no transport episodes were unequivocally registered. Analysis of the end of the seasons was more problematic, due to contributions to pollen observations from local sources; this results in difficulties in the reliable identification of the long-range transport episodes. Apart from its short-term effects on the pollen seasons, long-range transport can have substantial impacts on the exchange of genetic material within Europe. A quick atmospheric pathway for gene transport can be important for adaptation of plants to a changing climate.  相似文献

MOTIVATION: We introduce a dual multiple change-point (MCP) model for recombination detection among aligned nucleotide sequences. The dual MCP model is an extension of the model introduced previously by Suchard and co-workers. In the original single MCP model, one change-point process is used to model spatial phylogenetic variation. Here, we show that using two change-point processes, one for spatial variation of tree topologies and the other for spatial variation of substitution process parameters, increases recombination detection accuracy. Statistical analysis is done in a Bayesian framework using reversible jump Markov chain Monte Carlo sampling to approximate the joint posterior distribution of all model parameters. RESULTS: We use primate mitochondrial DNA data with simulated recombination break-points at specific locations to compare the two models. We also analyze two real HIV sequences to identify recombination break-points using the dual MCP model.  相似文献

Skeletal muscle functions regulated by NO are now firmly established. However, the literature on the compartmentalization of NO signaling in myocytes is highly controversial. To address this issue, we examined localization of enzymes engaged in L-arginine-NO-cGMP signaling in the rat quadriceps muscle. Employing immunocytochemical labeling complemented with tyramide signal amplification and electron microscopy, we found NO synthase expressed not only in the sarcolemma, but also along contractile fibers, in the sarcoplasmic reticulum and mitochondria. The expression pattern of NO synthase in myocytes showed striking parallels with the enzymes engaged in L-arginine-NO-cGMP signaling (arginase, phosphodiesterase, and soluble guanylyl cyclase). Our findings are indicative of an autocrine fashion of NO signaling in skeletal muscles at both cellular and subcellular levels, and challenge the notion that the NO generation is restricted to the sarcolemma.  相似文献
