The binding and uptake of cholesterol enriched lipoproteins by isolated hepatocytes was decreased at 16 hours after partial hepatectomy, with a tendency to return to control values as the regeneration proceeds. The number of lipoprotein binding sites of total cellular membranes remained similar to control at 16 and 24 hours. The plasma lipoprotein pattern, determined by electrophoretic analysis, showed a lower per cent of very low density lipoproteins (VLDL) and a higher per cent of low density lipoproteins (LDL) at 16 and 24 hours post-partial hepatectomy. At these times, plasma lecithin: cholesterol acyltransferase (LCAT) activity was decreased. It is intriguing to suggest that the regenerating liver could regulated the blood lipoprotein pattern and the uptake of lipoproteins by modulating the surface expression of the receptors.  相似文献

Map-based (positional) cloning has traditionally been the preferred strategy for identifying the causal genes underlying the phenotypes of mutants isolated in forward genetic screens. Massively parallel sequencing technologies are enabling the rapid cloning of genes identified in such screens. We have used a combination of linkage mapping and whole-genome re-sequencing to identify the causal mutations in four loss-of-function angulata (anu) mutants. These mutants were isolated in a screen for mutants with defects in leaf shape and leaf pigmentation. Our results show that the anu1-1, anu4-1, anu9-1 and anu12-1 mutants carry new alleles of the previously characterized SECA2, TRANSLOCON AT THE OUTER MEMBRANE OF CHLOROPLASTS 33 (TOC33), NON-INTRINSIC ABC PROTEIN 14 (NAP14) and CLP PROTEASE PROTEOLYTIC SUBUNIT 1 (CLPR1) genes. Re-sequencing the genomes of fine mapped mutants is a feasible approach that has allowed us to identify a moderate number of candidate mutations, including the one that causes the mutant phenotype, in a nonstandard genetic background. Our results indicate that anu mutations specifically affect plastid-localized proteins involved in diverse processes, such as the movement of peptides through chloroplast membranes (ANU1 and ANU4), metal homeostasis (ANU9) and protein degradation (ANU12).  相似文献

A bioassay-guided fractionation of leaf extracts from Clytostoma callistegioides (Cham.) Bureau ex Griseb. (Bignoniaceae) led to isolation of a natural mixture of four fatty acids with anti-insect activity against aphids. The compounds were identified by GC–MS as palmitic, stearic, linoleic and linolenic acids and quantified as their methyl esters. The anti-aphid activity of the natural mixture was traced to linolenic and linoleic acids, as shown by the settling inhibition activity of synthetic samples. Interestingly, the saturated acids (palmitic and stearic) tested alone stimulated settling on one of the tested aphids (Myzus persicae), but not on the other tested species (Rhopalosiphum padi). Although ubiquitous, none of these free acids have been previously reported in this Bignoniaceae species. The leaf surface chemistry, which is likely involved in modulating aphid settling behavior, was further investigated for the occurrence of lipophilic substances by histochemical staining. Short, stalked glandular trichomes, previously undescribed for this species, stained with osmium tetroxide and Sudan III, suggesting that the secretion of the defensive acids is related to these surface trichomes.  相似文献

Numerous investigations have found a relationship between higher risk of cancer and increased intake of fats, while results of clinical studies of fat reduction and breast cancer recurrence have been mixed. A diet completely free of fats cannot be easily administered to humans, but experimental studies in mice can be done to determine whether this extreme condition influences tumor development. Here, we examined the effects of a FA‐free diet on mammary tumor development and growth rate in female FVB‐neu proto‐oncogene transgenic mice that develop spontaneous multifocal mammary tumors after a long latency period. Mice were fed a fatty acid‐free diet beginning at 112, 35, and 30 days of age. In all these experiments, tumor appearance was delayed, tumor incidence was reduced and the mean number of palpable mammary tumors per mouse was lower, as compared to standard diet‐fed mice. By contrast, tumor growth rate was unaffected in mice fed the fatty acid‐free diet. Plasma of mice fed the fatty acid‐free diet revealed significantly higher contents of oleic, palmitoleic and 20:3ω9 acids and lower contents of linoleic and palmitic acids. In conclusion, these findings indicate that a FA‐free diet reduces tumor incidence and latency but not tumor growth rate, suggesting that a reduction in dietary FAs in humans may have a protective effect on tumorigenesis but not on tumors once they appear. Two computer packages have been developed to teach bacterialgenetics on an introductory genetics course for undergraduatestudents in biology. The first package, ‘CONJUGACION’,is designed to teach bacterial conjugation and its genetic outcomes.It includes four main parts. Firstly, a tutorial part presentsa theoretical framework using screens of text and animated graphics.Secondly, an interactive concept application section requiresstudents to carry out experiments for the determination of thecorrect sex and genotype of 10 bacterial strains. The thirdpart uses the previously obtained data for simulating interruptedmating experiments and mapping the bacterial genome. Finally,an evaluation section allows the students to test their understandingthrough a series of multiple choice questions. The second package,‘LURIDEL’, is intended for teaching the preadaptativecharacter of mutation in bacterial populations on the basisof the fluctuation test of Luria and Delbruck. It simulates,graphically, the appearance of mutations in microbial culturesand gives results of simulated fluctuation experiments. The aim of this work was to improve the knowledge on young suckled Salers bull production and to study the effect of forage type and concentrate level on performance, carcass and muscle characteristics as well as on meat quality. Twenty-four Salers male calves of 150 days of age were assigned to six groups: C0 (fed exclusively with hay and dam's milk and slaughtered at approximately 6 months of age), and HH (hay - high concentrate), HL (hay - low concentrate), GH (cut grass - high concentrate), GL (cut grass - low concentrate) and CP (control pasture: pasture - high concentrate) groups differing in feeds received until slaughter and slaughtered unweaned at approximately 10 months of age. Carcass weights averaged 210 kg at 10 months of age at slaughter. Average daily weight gain (ADWG) in HH and GH groups tended to be higher (P = 0.09) than in HL and GL groups (1354 v. 1248 g/day). ADWG in CP group (1542 g/day) was higher (P < 0.05) than in the other groups. Carcass weight in CP group (230 kg) tended to be higher (P < 0.1) than in HL (198 kg) and GL (200 kg) groups. Carcass muscle weight was higher (P < 0.05) in GH (155 kg) and CP groups (165 kg) than in HL (141 kg) and GL (142 kg) groups. Carcass and offal fatty tissue weights and carcass fatness did not differ between groups. Neither forage type nor concentrate level had significant effect on the area of muscle fibres or on muscle metabolic enzyme activities (namely, lactate dehydrogenase - LDH, phosphofructokinase - PFK, isocitrate dehydrogenase - ICDH, citrate synthase - CS and cytochrome-c oxidase - COX). semitendinosus muscle of CP group presented higher CS enzyme activities (8.10 μmol/min per g) than HH (5.30 μmol/min per g) and GL (4.52 μmol/min per g) groups. Neither total nor insoluble collagen content significantly differed between groups. Lipid content in rectus abdominis muscle was relatively low (average 67.5 mg/g dry matter) and was not affected by diet (P > 0.05). The ratio between n-6 and n-3 polyunsaturated fatty acid content was lower (P = 0.01) in the low-concentrate-fed than in the high-concentrate-fed group (3.95 v. 5.37, respectively). Sensory analysis noted that longissimus thoracis muscle from CP animals was more tender and juicy than that from HH and GH animals (P < 0.05).  相似文献
