A series of Group 12 metal complexes with bis(N,N-disubstituted amino)carbeniumdithiocarboxylates (RL) have prepared by the reaction of metal perchlorate with RL in EtOH or CH2Cl2, and characterized by the X-ray structural analysis, IR spectrum, and electrochemical measurements. The composition of metal and RL and the coordination mode, such as monodentate, chelate, and bridging or capping, depend on both the central metal ion and the substituent on the diaminocarbenium moiety. The useful criteria for the coordination mode are found to be the wavenumber of the -CS2 asymmetric stretching vibration in the solid state, and the ligand-based reduction potential in solution.  相似文献

doi: 10.1111/j.1741‐2358.2012.00664.x Relationship between behavioral and psychological symptoms of dementia and oral health status in the elderly with vascular dementia Objectives: The aim of this study was to clarify the relationship between behavioral and psychological symptoms of dementia (BPSD) and oral status in the elderly with vascular dementia. Background: There have been some reports of a relationship between disease symptoms and oral status in the elderly with Alzheimer’s disease, but few reports have been conducted in the elderly with vascular dementia. Until now, the relationship between BPSD and oral status has been unknown. Materials and methods: An investigation was conducted concerning BPSD and oral status among 57 subjects with vascular dementia (mean age, 85.7 ± 5.5 years). The wearing of dentures and oral activities of daily living (oral ADL) were examined. Results: Subjects with activity disturbances and those with aggressiveness had significantly lower rates of denture wearing than those without these two symptoms (p < 0.05). Significantly lower oral ADL scores were obtained from subjects with delusional ideas, hallucinations, activity disturbances and diurnal rhythm disturbances (p < 0.05), as well as those with affective disturbances (p < 0.01). Conclusion: This study indicated a relationship between BPSD and the wearing of dentures in the elderly with vascular dementia. The study also demonstrated relationships between BPSD and oral ADL.  相似文献

BackgroundNeuromyelitis optica (NMO) is an inflammatory disease caused by the aquaporin (AQP)-4-antibody. Pathological studies on NMO have revealed extensive astrocytic damage, as evidenced by the loss of AQP4 and glial fibrillary acidic protein (GFAP), specifically in perivascular regions with immunoglobulin and complement depositions, although other pathological patterns, such as a loss of AQP4 without astrocyte destruction and clasmatodendrosis, have also been observed. Previous studies have shown that complement-dependent antibody-mediated astrocyte lysis is likely a major pathomechanism in NMO. However, there are also data to suggest antibody-mediated astrocyte dysfunction in the absence of complement. Thus, the importance of complement inhibitory proteins in complement-dependent AQP4-antibody-mediated astrocyte lysis in NMO is unclear. In most of the previous studies, the complement and target cells (astrocytes or AQP4-transfected cells) were derived from different species; however, the complement inhibitory proteins that are expressed on the cell surface cannot protect themselves against complement-dependent cytolysis unless the complements and complement inhibitory proteins are from the same species. To resolve these issues, we studied human astrocytes in primary culture treated with AQP4-antibody in the presence or absence of human complement and examined the effect of complement inhibitory proteins using small interfering RNA (siRNA).MethodsPurified IgG (10 mg/mL) was obtained from 5 patients with AQP4-antibody-positive NMO, 3 patients with multiple sclerosis (MS), and 3 healthy controls. Confluent human astrocytes transfected with Venus-M1-AQP4-cDNA were incubated with IgG (5% volume). After washing, we cultured the cells with human complements with or without heat inactivation. We observed time-lapse morphological and immunohistochemical changes using a fluorescence microscope. We also evaluated cytotoxicity using a propidium iodide (PI) kit and the lactate dehydrogenase (LDH) assay.ResultAQP4-antibody alone caused clustering and degradation followed by endocytosis of membraneous AQP4, thereby resulting in decreased cellular adherence and the shrinkage of astrocytic processes. However, these changes were partially reversed by the removal of IgG in culture. In contrast, following the application of AQP4-antibody and non-heated human complements, the cell bodies and nuclei started to swell. At 3 h, most of the astrocytes had lost mobility and adherence and were eventually destroyed or had swollen and were then destroyed. In addition, the remaining adherent cells were mostly PI-positive, indicating necrosis. Astrocyte lysis caused by rabbit complement occurred much faster than did cell lysis with human complement. However, the cell lysis was significantly enhanced by the transfection of astrocytes with siRNA against human CD55 and CD59, which are major complement inhibitory proteins on the astrocyte membrane. AQP4-antibody-negative IgG in MS or control did not induce such changes.ConclusionTaken together, these findings suggest that both complement-dependent and complement-independent AQP4-antibody-mediated astrocytopathies may operate in NMO, potentially contributing to diverse pathological patterns. Our results also suggest that the effect of complement inhibitory proteins should be considered when evaluating AQP4-antibody-mediated cytotoxicity in AQP4-expressing cells.  相似文献

Circulating lymphocytes infiltrate into local foci at the inflammatory phase of acute wound healing for activation of the immune system and express an immune checkpoint protein programmed cell death 1 (PD-1) at the resolution phase for inactivation of the immune system. Conversely, the PD-1 expression was still found even on circulating lymphocytes of the elder patients with chronic tonsillitis at the palliative stage. Recently, an adhesion G protein coupled receptor 56 (GPR56) was reported to at least work as a proliferation factor for infiltrated lymphocytes into local foci at the resolution phase of acute wound healing. To preliminary examine a similar role of PD-1 and GPR56 at local foci at chronic inflammation, palate tonsils were prepared from small amounts of patients with chronic tonsillitis and tonsillar hypertrophy. A positive relationship of RNA expression might be observed between PD-1 and GPR56 in the elder patients with chronic tonsillitis. In regard to immunohistopathological findings, there were huge and small amounts of PD-1 and GPR56 expression at the marginal zone of lymphoid follicles of palate tonsils with chronic tonsillitis. Moreover, the positive relationship of RNA expression between PD-1 and GPR56 confirmed in large numbers of the elder patients with chronic tonsillitis. Probably, GPR56 participates in a supplement of PD-1+ lymphocytes to circulating bloods of the elder patients with chronic tonsillitis through a lymphocyte cell maintenance system at the marginal zone of the lymphoid follicles of palate tonsils.  相似文献

Antifouling agents with low toxicity are in high demand for sustaining marine industries and the environment. This study aimed to synthesize 15 isothiocyanates derived from β-citronellol and evaluate their antifouling activities and toxicities against cypris larvae of the barnacle Amphibalanus amphitrite. The synthesized isothiocyanates exhibited effective antifouling activities (EC50=0.10–3.33 μg mL−1) with high therapeutic ratios (LC50/EC50 >30). Four isothiocyanates with an amide or isocyano group showed great potential as effective antifouling agents (EC50=0.10–0.32 μg mL−1, LC50/EC50=104–833). The enantiomers of the isothiocyanates only slightly differed in their antifouling activities. These results may serve as a basis for further research and development of β-citronellol-derived isothiocyanates as effective low-toxic antifouling agents. To the best of our knowledge, this study is the first to report the antifouling activities of isothiocyanates derived from accessible natural products.  相似文献

Evergreen broad-leaved forests (EBLFs, lucidophyllous forests) are vegetation types characteristic of East Asia. The extent of EBLFs has decreased significantly due to long-term anthropogenic disturbance, and remnant EBLFs in urban area are rare and important landscape types contributing to biodiversity and sustainable development. This study focuses on remnant EBLFs on Mt. Gele (GL), Mt. Tieshanping (TSP), and Mt. Jinyun (JY), located from the inner city to outskirts of Chongqing metropolis, Southwest China. Species of Theaceae, Lauraceae, Symplocaceae, and other families, which are essential floristic components of primary EBLFs, were still the main components at the three sites. GL and JY showed higher biodiversity, with richer heliophytes and shade-tolerant species, respectively. Castanopsis carlesii var. spinulosa was the sole dominant species at all three sites in woody layer, with codomination by Pinus massoniana and Cinnamomum camphora at GL and by Machilus pingii and Castanopsis fargesii at JY; these evergreen broad-leaved trees all showed inverse-J or sporadic-shaped size distribution with large numbers of small stems, but the conifer tree Pinus massoniana showed unimodal distribution with small stems at GL. The height growth of tree species, especially Castanopsis carlesii var. spinulosa, was increasingly restricted from JY to TSP to GL. Sprouting is an important life history strategy at community and population level, and differences were exhibited from GL to TSP to JY. A rural–urban gradient from JY to TSP to GL was indicated in this study. Species composition, biodiversity, and stand structure of these remnant EBLFs showed obvious differences along this gradient, and conservation responses to address the effects of urbanization need to be carefully considered.  相似文献

A tissue uptake experiment was conducted to determine the bioavailability of rumen bacterial Selenium (Se) in mice. The donor animal was wether fed a diet containing 0.2 mg Se/kg dietary dry matter (DM). Ruminal fluid was collected 2 h postprandially. Bacterial-rich precipitate was obtained by differential centrifugation of the ruminal fluids. This was later freeze-dried and mixed in the diet to be used in feeding the mice experiment. Thirty growing female mice with a body wt (mean±SD) of 21.4±0.74 g were housed in plastic cages (5 mice/cage) and allotted equally to three dietary treatments. Diet 1 and Diet 2 were formulated based on AIN-76, except that no Se supplementation in the form of selenite was made in the former. In Diet 3, rumen bacterial matter was 20% of the diet, which gave an equivalent of 0.1 mg Se/kg dietary DM. The other two diets, Diet 1 and Diet 2, had an Se content of 0.025 and 0.1 mg/kg dietary DM, respectively. A 7-d feeding commenced after 7 d of acclimatization of the semipurified diet. Results showed that those mice fed an Se- (selenite) supplemented diet (Diet 2) had higher (P<0.05) tissue Se concentrations than those mice fed the other two diets. No statistical differences were observed on various tissue Se concentrations between Diet 1 and Diet 3, although the latter diet had higher values. Kidney and liver had the highest Se concentrations compared to the other tissues. This study concludes that bacterial Se collected from the rumen of wether is not fully available for absorption in the intestine of the mice.  相似文献

Acylglycerol kinase (AGK) is a mitochondrial lipid kinase that contributes to protein biogenesis as a subunit of the TIM22 complex at the inner mitochondrial membrane. Mutations in AGK cause Sengers syndrome, an autosomal recessive condition characterized by congenital cataracts, hypertrophic cardiomyopathy, skeletal myopathy, and lactic acidosis. We mapped the proteomic changes in Sengers patient fibroblasts and AGKKO cell lines to understand the effects of AGK dysfunction on mitochondria. This uncovered down-regulation of a number of proteins at the inner mitochondrial membrane, including many SLC25 carrier family proteins, which are predicted substrates of the complex. We also observed down-regulation of SFXN proteins, which contain five transmembrane domains, and show that they represent a novel class of TIM22 complex substrate. Perturbed biogenesis of SFXN proteins in cells lacking AGK reduces the proliferative capabilities of these cells in the absence of exogenous serine, suggesting that dysregulation of one-carbon metabolism is a molecular feature in the biology of Sengers syndrome.  相似文献

Soluble glass bolus (SGB) with selenium (Se) was administered intraruminally to Se-deficient Philippine does to determine its effect on milk Se and to correlate the Se contents of does' milk and blood of does and kids. Five months after the Se administration, the does in the treated group (n=14) had higher (p<0.01) Se content in their blood (62.2 vs 25.7 μg/L) and milk (5.1 vs 2.5 μg/L) than does in control group (n=13). Consequently, the blood Se of the kids (n=14) from the treated does was higher (p<0.05) than those kids (n=13) in the control group (28.0 vs 5.1 μg/L). Blood and milk Se of does and blood Se of their kids correlated (p<0.01) with each other. The increased Se level of does' milk because of Se supplementation was not regarded as a health hazard to humans.  相似文献
