Metabolically stable polyamine derivatives may serve as useful surrogates for the natural polyamines in studies aimed to elucidate the functions of individual polyamines. Here we studied the metabolic stability of alpha-methylspermidine, alpha-methylspermine, and bis-alpha-methylspermine, which all have been reported to fulfill many of the putative physiological functions of the natural polyamines. In vivo studies were performed with the transgenic rats overexpressing spermidine/spermine N(1)-acetyltransferase. alpha-Methylspermidine effectively accumulated in the liver and did not appear to undergo any further metabolism. On the other hand, alpha-methylspermine was readily converted to alpha-methylspermidine and spermidine; similarly, bis-alpha-methylspermine was converted to alpha-methylspermidine to some extent, both conversions being inhibited by the polyamine oxidase inhibitor N(1), N(2)-bis(2,3-butadienyl)-1,4-butanediamine. Furthermore, we used recombinant polyamine oxidase, spermidine/spermine N(1)-acetyltransferase, and the recently discovered spermine oxidase in the kinetic studies. In vitro studies confirmed that methylation did not protect spermine analogs from degradation, whereas the spermidine analog was stable. Both alpha-methylspermidine and bis-alpha-methylspermine overcame the proliferative block of early liver regeneration in transgenic rats and reversed the cytostasis induced by an inhibition of ornithine decarboxylase in cultured fetal fibroblasts.  相似文献

This review considers the chemical and biotechnological synthesis of acids that are obtained by direct oxidation of mono- or oligosaccharide, referred to as sugar acids. It focuses on sugar acids which can be readily derived from plant biomass sources and their current and future applications. The three main classes of sugar acids are aldonic, aldaric and uronic acids. Interest in organic acids derived from sugars has recently increased, as part of the interest to develop biorefineries which produce not only biofuels, but also chemicals to replace those currently derived from petroleum. More than half of the most desirable biologically produced platform chemicals are organic acids. Currently, the only sugar acid with high commercial production is d-gluconic acid. However, other sugar acids such as d-glucaric and meso-galactaric acids are being produced at a lower scale. The sugar acids have application as sequestering agents and binders, corrosion inhibitors, biodegradable chelators for pharmaceuticals and pH regulators. There is also considerable interest in the use of these molecules in the production of synthetic polymers, including polyamides, polyesters and hydrogels. Further development of these sugar acids will lead to higher volume production of the appropriate sugar acids and will help support the next generation of biorefineries.  相似文献

Morphometrics and growth of the critically endangered Saimaa ringed seal (Pusa hispida saimensis), which inhabits a freshwater lake in Finland were studied using data from 344 carcasses. This study presents the first detailed information on ringed seal pup growth and body condition from birth to the age of one year. Newborn pups were on average 68 cm long and weighed 5 kg. Pups attained the majority of their first year growth during the nursing period. Body condition and growth declined after weaning, but pups recovered from postweaning nutritional deprivation at the age of 8 mo. The seals achieved their maximum body length, girth, and mandible size around the age of 4 yr, and asymptotic body mass two years later. Baculum growth indicated that males reached sexual maturity at age 5–6 yr. The Saimaa ringed seals' asymptotic body length and mass were 132 cm and 59 kg, respectively, which is similar to medium sized marine ringed seals. Saimaa ringed seals' survival to adulthood is extremely low due to high bycatch mortality; furthermore climate change may hamper pup growth and elevate mortality. Therefore we recommend continuation of fishing closures to improve pup survival.  相似文献

An extreme marine heat wave which affected 2000 km of the midwest coast of Australia occurred in the 2010/11 austral summer, with sea‐surface temperature (SST) anomalies of 2–5°C above normal climatology. The heat wave was influenced by a strong Leeuwin Current during an extreme La Niña event at a global warming hot spot in the Indian Ocean. This event had a significant effect on the marine ecosystem with changes to seagrass/algae and coral habitats, as well as fish kills and southern extension of the range of some tropical species. The effect has been exacerbated by above‐average SST in the following two summers, 2011/12 and 2012/13. This study examined the major impact the event had on invertebrate fisheries and the management adaption applied. A 99% mortality of Roei abalone (Haliotis roei) and major reductions in recruitment of scallops (Amusium balloti), king (Penaeus latisulcatus) and tiger (P. esculentus) prawns, and blue swimmer crabs were detected with management adapting with effort reductions or spatial/temporal closures to protect the spawning stock and restocking being evaluated. This study illustrates that fisheries management under extreme temperature events requires an early identification of temperature hot spots, early detection of abundance changes (preferably using pre‐recruit surveys), and flexible harvest strategies which allow a quick response to minimize the effect of heavy fishing on poor recruitment to enable protection of the spawning stock. This has required researchers, managers, and industry to adapt to fish stocks affected by an extreme environmental event that may become more frequent due to climate change.  相似文献

We used the simian immunodeficiency virus (SIV) molecular clone SIVmac239 to generate a deletion construct, termed SD2, in which we eliminated 22 nucleotides at positions +398 to +418 within the putative dimerization initiation site (DIS) stem. This SD2 deletion severely impaired viral replication, due to adverse effects on the packaging of viral genomic RNA, the processing of Gag proteins, and viral protein patterns. However, long-term culture of SD2 in either C8166 or CEMx174 cells resulted in restoration of replication capacity, due to two different sets of three compensatory point mutations, located within both the DIS and Gag regions. In the case of C8166 cells, both a K197R and a E49K mutation were identified within the capsid (CA) protein and the p6 protein of Gag, respectively, while the other point mutation (A423G) was found within the putative DIS loop. In the case of CEMx174 cells, two compensatory mutations were present within the viral nucleocapsid (NC) protein, E18G and Q31K, in addition to the same A423G substitution as observed with C8166 cells. A set of all three mutations was required in each case for restoration of replication capacity, and either set of mutations could be substituted for the other in both the C8166 and CEMx174 cell lines.  相似文献

The breeding habitat and lair structures of the ringed seal (Phoca hispida ladogensis) were studied by snowmobile expeditions in northern Lake Ladoga, Russia, during 1996 to 1999. Mostly flat ice and no large pack ice existed in northern Lake Ladoga during this study. All birth lairs and 88% of haul-out lairs were located in snowdrifts in the shorelines of islands or islets and 12% of the haul-out lairs occurred in pressured ice ridges. Some large haul-out lairs had been used communally by the seals. A total of 15% of the lairs observed had been attacked or marked by red fox (Vulpes vulpes), wolf (Canis lupus) or unidentified canines. The colour of the lanugo fur of the Ladoga ringed seal pup appeared to be dark grey. We conclude that the northern part of Lake Ladoga is an active breeding area for the ringed seal, which attests conservation needs for this area. Accepted: 18 September 2000  相似文献

During cold acclimation, winter rye (Secale cereale L.) plants secrete antifreeze proteins that are similar to pathogenesis-related (PR) proteins. In this experiment, the secretion of PR proteins was induced at warm temperatures by infection with pink snow mold (Microdochium nivale), a pathogen of overwintering cereals. A comparison of cold-induced and pathogen-induced proteins showed that PR proteins accumulated in the leaf apoplast to a greater level in response to cold. The PR proteins induced by cold and by snow mold were similar when separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and examined by immunoblotting. Both groups of PR proteins contained glucanase-like, chitinase-like, and thaumatin-like proteins, and both groups exhibited similar levels of glucanase and chitinase activities. However, only the PR proteins induced by cold exhibited antifreeze activity. Our findings suggest that the cold-induced PR proteins may be isoforms that function as antifreeze proteins to modify the growth of ice during freezing while also providing resistance to the growth of low-temperature pathogens in advance of infection. Both functions of the cold-induced PR proteins may improve the survival of overwintering cereals.  相似文献

Activation of polyamine catabolism through the overexpression of spermidine/spermine N1-acetyltransferase (SSAT) in transgenic rodents does not only lead to distorted tissue polyamine homeostasis, manifested as striking accumulation of putrescine, appearance N1-acetylspermidine and reduction of tissue spermidine and/or spermine pools, but likewise creates striking phenotypic changes. The latter include loss of hair, lipoatrophy and female infertility. Forced expression of SSAT modulates skin, prostate and intestinal carcinogenesis, induces acute pancreatitis and blocks early liver regeneration. Although many of these features are directly attributable to altered tissue polyamine pools, some of them are more likely related to the greatly accelerated flux of the polyamines caused by activated catabolism and compensatorily enhanced biosynthesis.  相似文献
