Using an in vitro system, we studied the effect of postischemic reoxygenation on cerebral lipid peroxidation in relation to the dietary intake of vitamin E (VE) in rats. Homogenates prepared from VE-deficient, -normal, and -supplemented brains, which were previously rendered ischemic for 30 min by decapitation, were incubated under air or nitrogen gas for 60 min. The extent of peroxidation in brain tissue was estimated by a thiobarbituric acid (TBA) test and by diene conjugation in total lipid extracts. The brain levels of alpha-tocopherol and of total and free fatty acids (FAs) were also determined. Aerobic incubation increased TBA reactants in all dietary groups; the effect was largest in the VE-deficient group, intermediate in the VE-normal group, and smallest in the VE-supplemented group. In contrast, nitrogen incubation did not alter the basal levels of TBA reactants except for a small rise associated with VE deficiency. Conjugated dienes changed in parallel with TBA reactants. alpha-Tocopherol decreased after aerobic incubation and also, to a lesser degree, after nitrogen incubation in each dietary group. Only in the reoxygenated samples of the VE-deficient group was there a significant fall in total polyunsaturated FAs. The levels of free FAs continuously increased throughout ischemia and subsequent incubation. However, the level of free polyunsaturated FAs was similar after aerobic and nitrogen incubation in each dietary group, and was not affected by VE. Thus, cerebral reoxygenation after ischemia propagates peroxidative reactions within esterified polyunsaturated FAs. The modification by VE of reoxygenation-induced lipid peroxidation suggests free radical mediation.  相似文献

The levels of iron, zinc, and copper in the tissues of the pregnant rat, on d 12, 19, and 21 after impregnation have been determined and compared with controls. Iron levels decreased considerably in late pregnancy as a result of increased fetal requirements, thus diminishing iron stores in rat tissues, but maintaining the circulating plasma levels. Copper levels increased slightly at midpregnancy, but returned to control levels at the end of gestation. Zinc stores also increased slightly during early pregnancy, yet were decreased at the end of pregnancy, but to a lesser extent than those of iron. The data are explained on the basis of equilibrium between assimilation and fetal needs for copper, a slightly higher demand for zinc with altered equilibrium, and a much altered equilibrium for iron that provokes a dwindling of iron maternal reserves that is not compensated by dietary iron.  相似文献

Histoplasma capsulatum yeast cells have been studied by immunoelectron microscopy using rabbit polyclonal antisera and a biotin-avidin-peroxidase detection system. An antigenic surface layer has been visualized in the cell wall of immunostained organisms. This layer was not seen in samples prepared by standard electron microscopic methods or in negative controls used with the immunocytochemical technique. Without immunostaining the cell wall ofHistoplasma appeared almost transparent. In contrast, after immunoperoxidase staining the cell wall was conspicuous, bounded by the darkly stained outer layer. The interaction of the 2'-phosphate-containing nucleotides (NADP+, NADPH, 2'-phosphoadenosine 5'-diphosphoribose, and adenosine 2',5'-bisphosphate) with NADP+ -specific isocitrate dehydrogenase was studied by using 31P NMR spectroscopy. The separate resonances corresponding to free and bound nucleotides, characteristic for slow exchange of nuclei on the NMR time scale, were observed in the spectra of the enzyme (obtained in the presence of excess ligand) with NADP+ and NADPH in the absence and presence of Mg2+ and with 2'-phosphoadenosine 5'-diphosphoribose in the absence of metal or in the presence of the substrate magnesium isocitrate. The position of the 31P resonance of the bound 2'-phosphate group in these spectra is invariant (delta = 6) in the pH range 5-8, indicating that the pK of this group is much lower in the complexes with the enzyme than that (pK = 6.13) in the free nucleotides. The additional downfield shift of this resonance by 1.8 ppm beyond that (delta = 4.22) of the dianionic form of the 2'-phosphate in free nucleotides suggests interaction with a positively charged group(s) and/or distortion of P-O-P angles as the result of binding to the enzyme. A single resonance of 2'-phosphate was observed in the spectrum of the enzyme complex with 2'-phosphoadenosine 5'-diphosphoribose in the presence of Mg2+, with the chemical shift dependent on the nucleotide to enzyme ratio, characteristic for the fast exchange situation. Near-ultraviolet (UV) light (325 to 400 nm), in the presence of air and the absence of exogenous photosensitizing compounds, is lethal and mutagenic for Haemophilus influenzae. The lethal effect is the same for both wild type and streptomycin-resistant mutants, indicating that the mutants are not selected by the irradiation. The inactivation and mutagenicity show a large shoulder, suggesting the existence of repair systems. Filters were used to eliminate the possibility of short-UV irradiation. The effective radiation is between 325 to 400 nm. The lethal and mutagenic effects are higher during mid and late log phase than during early log or stationary phase.  相似文献

Mutants of the petite-negative yeast, Kluyveromyceslactis, resistant to the inhibitors of oxidative phosphorylation, decamethylenediguanidine and octylguanidine were isolated from medium containing ethidium bromide. All mutants were resistant to ethidium bromide; some mutants were resistant to both alkylguanidines, some to one, others to neither. Both nuclear and cytoplasmic inheritance of resistance to decamethylenediguanidine and ethidium bromide was demonstrated by tetrad analysis.  相似文献

A virus having 300 nm long rod-shaped particles was isolated fromPlantago media L. in Yugoslavia. The virus was transmitted to 15 species of host plants the symptoms of which are described in detail. The symptoms corresponded to those that appeared after infection by the original Holmes' ribgrass virus (HRV). The investigated virus was compared both with the common strain of tobacco mosaic and the original Holmes' ribgrass viruses by means of serological tests. The agar double-diffusion tests showed that it is closely related to HRV and remotely related to the common strain of TMV. On the basis of these results we concluded that this virus represented a strain of the HRV. The investigations of the cell inclusions showed that our virus produced rounded plates instead of hexagonal prisms. Electron micrographs of ultrathin sectioned material demonstrated that these plates were formed by virus particles lying perpendicularly to the layers of the plates. The presence of plates also points to the fact that the investigated virus belongs to the HRV.  相似文献

Fine details of the internal and external morphology of Cryptococcus neoformans as seen in ultrathin sections are described and illustrated with electron micrographs. The capsule characteristic of this species contained microfibrils (30 to 40 A in diameter) that appeared to radiate from the cell wall and to coil and intertwine in various directions. These thin, uniformly structured, electron-dense filaments are believed to represent complex polysaccharide molecules. The internal morphology of C. neoformans was in many ways similar to that of yeasts studied by other authors. The cell was uninucleate with a single nucleolus. The nuclear envelope, a pair of unit membranes interrupted by pores, was typical of that found in eucaryotic organisms. Smooth endoplasmic reticulum, mitochondria, vacuoles, storage granules, and ribosomes were consistent features of the cytoplasm. In addition, C. neoformans presented membranous organelles derived from the plasma membrane and comparable to bacterial mesosomes and mitochondria of an annulate type.  相似文献

In the present work we examined the effect of the neutralization of endogenous substance P by the administration of an anti-substance P serum (ASPS) on GABA concentration in the anterior pituitary in hyperprolactinemic conditions induced by 5-hydroxytryptophan or by grafting anterior pituitaries. ASPS reduced the increase in the anterior pituitary GABA concentration induced by hyperprolactinemia. In vitro experiments showed that substance P inhibited K+-evoked GABA efflux from hypothalamic fragments and decreased GABA concentration in the anterior pituitary but ASPS increased it. Our results demonstrate that substance P modifies hypothalamic GABA release and anterior pituitary GABA concentration and suggest that an interaction exists between substance P and GABA.  相似文献

Three antibiotic peptides with amoebolytic activity have been purified from culture supernatants of Bacillus licheniformis M-4 (amoebicins m4-A, m4-B, and m4-C). They were hydrophilic peptides consisting of six different amino acids (Asp, Glu, Ser, Thr, Pro, Tyr). Their molecular weights ranged from 3,000 to 3,200. Purified amoebicins were active against human pathogenic and non-pathogenic strains of Naegleria.
