A 25-kb DNA SalI fragment cloned from the chromosomal DNA of Pseudomonas putida OUS82, which utilizes phenanthrene (Phn+) and naphthalene (Nah+), carried all of the genes necessary for upper naphthalene catabolism. Cosmid recombinant pIP7 complemented both the Nah- and Phn- defects of OUS8211 (Trp-Nah-Phn-Sal+[salicylate utilizing]Hna+[1-hydroxy-2-naphthoate utilizing]) and only the Phn- defect of OUS8212 (Trp-Nah-Phn-Sal-Hna+). The results indicate that strain OUS82 uses different pathways after o-hydroxycarboxylic aromatics in the catabolism of naphthalene and phenanthrene.  相似文献

Cytolytic process which was affected by cholera toxin (CT) resulting in the loss of natural killer (NK) cell activity was analyzed. Conjugate formation assay, membrane phospholipid methylation assay and serine esterase (granzyme A) release assay were used to determine the stage of the CT-induced inhibition of NK cell-mediated cytotoxicity. A human NK cell line YT cell-mediated cytotoxicity was completely abolished by CT pretreatment or addition of CT to the assay system. The conjugate formation assay revealed that the binding between YT cells and target cells was not affected by CT. The defined triggering stage which is coupled with membrane phospholipid methylation was not affected by CT treatment, either. On the other hand, the lethal hit stage which is represented by serine esterase (SE) release was completely inhibited by CT treatment of YT cells. Therefore, CT inhibits the stage after binding and triggering—i.e., lethal hit stage of NK cell-mediated cytotoxicity. The results also suggest that there exists a CT-sensitive negative cytotoxic signal transduction pathway as well as usual positive signal transduction pathway and these pathways might cross talk each other in the NK cell cytotoxic process.  相似文献

A Beijerinckia species, capable of oxidizing phenanthrene, biphenyl and other polycyclic aromatic hydrocarbons, was shown to contain two plasmids that were designated pKGl and pKG2. The molecular masses of plasmids pKG1 and pKG2, as determined by electron microscopy, were approximately 147 X 10(6) and 20.8 X 10(6) daltons, respectively. Growth of the organism on benzoate led to the isolation of strains that had lost the ability to grow with phenanthrene and biphenyl. All of the Phn-, Bph- strains had also lost the smaller plasmid, pKG2. The results presented suggest that plasmid pKG2 is responsible for the synthesis of enzymes involved in the degradation of phenanthrene and biphenyl.  相似文献

The basal salt formulation of a medium is a vital but often overlooked component in many in vitro applications, as it regulates the growth and morphology of plant tissues by providing essential nutrients. The MS and B5 formulations are the two most widely used basal media, yet they are suboptimal for many species. The objective of this study was to evaluate the BDS (modified B5) basal salt formulation for in vitro growth and development using rice, maize, soybean, cotton, onion, tobacco, muscadine, raspberry, and gerbera daisy as test species. The responses measured for each species included callus growth (biomass production), plant regeneration, micropropagation rate, hairy root growth, and production of secondary metabolites. BDS was compared to MS, B5, and BABI, a high-calcium version of BDS (440?mg/l?CaCl2). For the majority of the species and responses measured, the results obtained with BDS and/or BABI were equal to or better than those obtained with MS or B5. Because of the wide range of plant species and in vitro systems included in this study, we conclude that BDS??or simple variations of BDS, such as BABI??are better balanced for a variety of uses in plant biotechnology, research, and production systems than either MS or B5.  相似文献

In this work, we have designed cyclic peptides (cIBL, cIBR, cIBC, CH4 and CH7) derived from the parent IB peptide (ICAM-1(1-21)) that are inhibitors of ICAM-1/LFA-1-mediated T-cell adhesion to Caco-2 cell monolayers. Cyclic peptide cIBR has the best activity of any of the peptides evaluated. The active ICAM-1 peptides have a common Pro-Arg-Gly sequence that may be important for binding to LFA-1.  相似文献

Structure and immunological characteristics of the pectic arabinogalactan Vk2a (previously reported as Vk100A2a) from the roots of Vernonia kotschyana Sch. Bip. ex Walp. were investigated after enzymatic digestion of the galacturonan moiety and the side chains of the rhamnogalacturonan structure of Vk2a. endo-alpha-D-(1-->4)-Polygalacturonase digestion released the high molecular weight 'hairy region' (Vk2a-HR) and oligogalacturonides. Vk2a-HR consisted of GalA (4-linked) and Rha (2- or 2,4-linked) in a 1:1 ratio, with 60% of Rha branched at C-4. The Rha located in the rhamnogalacturonan core was branched randomly by Gal units. Vk2a-HR was rich in neutral sugars such as Araf 5- (12.2%) and 3,5-substituted (12.8%) and terminally- (14.1%) linked and Gal 4- (13.0%), 3- (0.9%), 6- (2.2%) and 3,6- (1.1%) substituted. Arabinans with chain lengths up to 11 units were identified. Araf residues were attached to C-3 of alpha-L-(1-->5)-Araf chains and to C-4 of Gal residues. Single Gal units and chains of beta-D-(1-->6)-linked galacto di- to penta-saccharides were attached to a beta-D-(1-->3)-galactan core. All the enzyme resistant fractions expressed potent complement fixation and induction of B-cell mitogenic activity, and the present study indicates that there may be several and possibly structurally different active sites involved in the bioactivity of Vk2a. The bioactive sites may be located both in the more peripheral parts of the molecule but also in the inner core of the 'hairy region' or in larger enzyme-resistant chains.  相似文献

Developmental changes in capacitative Ca(2+) entry and Ca(2+) release from intracellular stores were measured using fura-2 fluorescence method during the pregnancy period (day 3-;18) in mouse mammary epithelial cells. Ca(2+) release was identified with the transient intracellular Ca(2+) ([Ca(2+)](i)) increase induced by thapsigargin addition in a Ca(2+)-free solution. Capacitative Ca(2+) entry was measured by the transient [Ca(2+)](i) increase induced by re-addition of extracellular Ca(2+) after depletion of Ca(2+) stores by thapsigargin. The capacitative Ca(2+) entry was greatest at the early stage of pregnancy (i.e. day 3 of pregnancy) and decreased as pregnancy progressed, while Ca(2+) release remained unchanged throughout the developmental stages. These findings indicate that in contrast to Ca(2+) release, a close correlation exists between capacitative Ca(2+) entry and pregnancy-induced development in mammary epithelial cells.  相似文献

The environmental assessment of nanomanufacturing during the initial process design phase should lead to the development of competitive, safe, and environmentally responsible engineering and commercialization. Given the potential benefits and concerns regarding the use of single‐walled carbon nanotubes (SWNTs), three SWNT production processes have been investigated to assess their associated environmental impacts. These processes include arc ablation (arc), chemical vapor deposition (CVD), and high‐pressure carbon monoxide (HiPco). Without consideration of the currently unknown impacts of SWNT dispersion or other health impacts, life cycle assessment (LCA) methodology is used to analyze the environmental impact and provide a baseline for the environmental footprint of each manufacturing process. Although the technical attributes of the product resulting from each process may not be fully comparable, this study presents comparisons that show that the life cycle impacts are dominated by energy, specifically the electricity used in production. Under base case yield conditions, HiPco shows the lowest environmental impact, while the arc process has the lowest impact under best case yield conditions.  相似文献
