The lack of a mouse model has hampered an understanding of the pathogenesis and immunity of Marburg hemorrhagic fever (MHF), the disease caused by marburgvirus (MARV), and has created a bottleneck in the development of antiviral therapeutics. Primary isolates of the filoviruses, i.e., ebolavirus (EBOV) and MARV, are not lethal to immunocompetent adult mice. Previously, pathological, virologic, and immunologic evaluation of a mouse-adapted EBOV, developed by sequential passages in suckling mice, identified many similarities between this model and EBOV infections in nonhuman primates. We recently demonstrated that serially passaging virus recovered from the liver homogenates of MARV-infected immunodeficient (SCID) mice was highly successful in reducing the time to death in these mice from 50 to 70 days to 7 to 10 days after challenge with the isolate MARV-Ci67, -Musoke, or -Ravn. In this study, we extended our findings to show that further sequential passages of MARV-Ravn in immunocompetent mice caused the MARV to kill BALB/c mice. Serial sampling studies to characterize the pathology of mouse-adapted MARV-Ravn revealed that this model is similar to the guinea pig and nonhuman primate MHF models. Infection of BALB/c mice with mouse-adapted MARV-Ravn caused uncontrolled viremia and high viral titers in the liver, spleen, lymph node, and other organs; profound lymphopenia; destruction of lymphocytes within the spleen and lymph nodes; and marked liver damage and thrombocytopenia. Sequencing the mouse-adapted MARV-Ravn strain revealed differences in 16 predicted amino acids from the progenitor virus, although the exact changes required for adaptation are unclear at this time. This mouse-adapted MARV strain can now be used to develop and evaluate novel vaccines and therapeutics and may also help to provide a better understanding of the virulence factors associated with MARV.The filoviruses, Marburgvirus and Ebolavirus (MARV and EBOV), cause severe hemorrhagic fevers in humans and nonhuman primates (27). The incubation time is estimated to be 3 to 21 days, with human case fatality rates reaching 90% in some outbreaks. Filoviral hemorrhagic fevers are characterized by a nonspecific viral prodrome in the early stage of infection, including fever, headaches, and myalgia (27). This is followed by a hemorrhagic phase that can include development of a maculopapular rash, petechiae, and bleeding from the gums, intestines, and other mucosal surfaces. Death usually occurs within a week of initial symptoms and is thought to be due to uncontrolled viral replication, hypotension-induced shock caused by increased vascular permeability, and multiorgan failure, likely caused by disseminated intravascular coagulation and extensive necroses in the liver, spleen, intestine, and many other major organ systems (27).Human-derived MARVs (isolates Angola, Musoke, Ravn, and Ci67) do not kill immunocompetent adult mice (23). Furthermore, there are no published reports of any lethal mouse-adapted MARV. The current mouse-adapted EBOV, strain Zaire (ZEBOV), was developed by performing nine sequential passages of ZEBOV 1976 virus in suckling mice, followed by two sequential plaque picks. The resulting virus was uniformly lethal to mice inoculated intraperitoneally (i.p.). Pathological evaluation of infected mice identified many similarities and only a few differences between this model (7, 22) and infections in nonhuman primates (21).In a previous study, we took a slightly different approach to mouse adaptation of MARV and found that serially passaging virus recovered from the liver homogenates of MARV-Ravn-infected adult mice with severe combined immunodeficiency (SCID mice) resulted in the generation of SCID-adapted MARV-Ravn (scid-MARV) that rapidly killed SCID mice but did not kill adult immunocompetent mice (51). In this study, we used scid-MARV as starting material for the first round of infection of adult immunocompetent BALB/c mice and serially passaged virus recovered from the liver homogenates of the BALB/c mice. MARV-Ravn was chosen over SCID-adapted MARV-Ci67 or -Musoke because it adapted more rapidly to SCID mice than the other isolates did. This produced a mouse-adapted MARV-Ravn strain (ma-MARV) that could kill adult BALB/c mice. Serial sampling studies to characterize the pathogenesis of ma-MARV revealed that this model was very similar to the guinea pig and nonhuman primate Marburg hemorrhagic fever (MHF) models, including rapid viremia, induction of D-dimers (fibrin degradation products), thrombocytopenia, profound loss of circulating and tissue lymphocytes, and marked liver damage. Additionally, we compared the immunological responses of mice after infection with either nonadapted wild-type MARV-Ravn (wt-MARV) or ma-MARV. This mouse model of MARV infection not only should advance our understanding of MARV pathogenesis and immunity but also may play a critical role in discovery of therapeutics for MARV infection.  相似文献

Pregnane glycosides previously isolated from genus Caralluma (C. Penicillata, C. tuberculata and C. russelliana) were tested for their antitrypanosomal activity. Penicilloside E showed the highest antitrypanosomal activity (IC50 1.01 μg/ml) followed by caratuberside C (IC50 1.85 μg/ml), which exhibited the highest selectivity index (SI 12.04). It was noticed that acylation is required for the antitrypanosomal activity while glycosylation at C-20 has no significant effect on the activity.  相似文献

Early diagnosis of acute Kawasaki disease (KD), lying in the spectrum between infectious and autoimmune diseases, can be difficult. To clarify the role of peripheral CD8T cells in KD, we examined their activation, proliferation, maturation, and effector function by four-color flow cytometry. Compared to healthy/febrile controls, acute KD patients showed striking increase in early activation marker CD69+CD8T cells and maturation subsets, but HLA-DR+CD8T cells representing late activation did not increase. Although Ki67+CD8T cells reflecting ongoing cell division increased in acute KD and febrile controls, absolute numbers of CD8T cells and maturation subsets decreased in acute KD versus healthy controls. Effector cells were lower in acute than in convalescent KD. Perforin+CD8T cells, denoting cytolytic activity, were lower in KD patients versus febrile controls. CD69+CD8T cells increase in acute KD but effector differentiation is absent. CD69+CD8T cells could be a marker to determine disease progression, treatment response, and convalescence in acute KD.  相似文献

Structures of three pectic arabinogalactans, one from Vernonia kotschyana (Vk2a) and two from Cochlospermum tinctorium (Ct50A1 and Ct50A2), and their complement fixation and induction of B cell proliferation in vitro were compared. The polysaccharide Vk2a expressed potent biological activity in both assays compared with Ct50A1 and Ct50A2. Vk2a possessed a very high molecular weight (1150 +/- 20 kDa) compared with Ct50A1 and Ct50A2 which both showed a polydisperse nature with the highest molecular weight polymers in each fraction estimated at approximately 105 kDa (Ct1a) and 640 +/- 100 kDa (Ct2a), respectively. The HMW polymers showed complement fixation in the same range as the native fractions. The arabinogalactan II content was low in Vk2a (2%) compared with that in Ct50A1 (23%) and Ct50A2 (12%). The high molecular weight polymers were subjected to digestion with a beta-d-(1, 3)-galactanase-rich fraction from Driselase, oligomers were isolated by HPAEC, and their finer structures were determined by MALDI- and ES-qoToF-MS, linkage, and monosaccharide composition analyses. Vk2a consists of both a galacturonan core and a rhamnogalacturonan core rich in neutral side chains. The backbones of both Ct-polysaccharides consist mainly of RG-I regions with numerous neutral side chains dominated by galactosyl residues, whereas the homogalacturonan regions seem to be small. Differences in the chain lengths of the 6-linked galacto-oligosaccharides attached to the 3-linked galactan core could not be related to the differences in the potencies of the biological activities observed.  相似文献

Flavocytochrome b (Cyt b) is a heterodimeric, integral membrane protein that serves as the central component of an electron transferase system employed by phagocytes for elimination of bacterial and fungal pathogens. This report describes a rapid and efficient single-step purification of Cyt b from human neutrophil plasma membranes by solubilization in the nonionic detergent dodecylmaltoside (DDM) and immunoaffinity chromatography. A similar procedure for isolation of Cyt b directly from intact neutrophils by a combination of heparin and immunoaffinity chromatography is also presented. The stability of Cyt b was enhanced in DDM relative to previously employed solubilizing agents as determined by both monitoring the heme spectrum in crude membrane extracts and assaying resistance to proteolytic degradation following purification. Gel filtration chromatography and dynamic light scattering indicated that DDM maintains a predominantly monodisperse population of Cyt b following immunoaffinity purification. The high degree of purity obtained with this isolation procedure allowed for direct determination of a 2:1 heme to protein stoichiometry, confirming previous structural models. Analysis of the isolated heterodimer by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry allowed for accurate mass determination of p22(phox) as indicated by the gene sequence. Affinity-purified Cyt b was functionally reconstituted into artificial bilayers and demonstrated that catalytic activity of the protein was efficiently retained throughout the purification procedure.  相似文献

SKIP (skeletal muscle and kidney enriched inositol phosphatase) is a recently identified phosphatidylinositol 3,4,5-trisphosphate- and phosphatidylinositol 4,5-bisphosphate-specific 5-phosphatase. In this study, we investigated the intracellular localization of SKIP. Indirect immunofluorescence and subcellular fractionation showed that, in serum-starved cells, both endogenous and recombinant SKIP colocalized with markers of the endoplasmic reticulum (ER). Following epidermal growth factor (EGF) stimulation, SKIP transiently translocated to plasma membrane ruffles and colocalized with submembranous actin. Data base searching demonstrated a novel 128-amino acid domain in the C terminus of SKIP, designated SKICH for SKIP carboxyl homology, which is also found in the 107-kDa 5-phosphatase PIPP and in members of the TRAF6-binding protein family. Recombinant SKIP lacking the SKICH domain localized to the ER, but did not translocate to membrane ruffles following EGF stimulation. The SKIP SKICH domain showed perinuclear localization and mediated EGF-stimulated plasma membrane ruffle localization. The SKICH domain of the 5-phosphatase PIPP also mediated plasma membrane ruffle localization. Mutational analysis identified the core sequence within the SKICH domain that mediated constitutive membrane association and C-terminal sequences unique to SKIP that contributed to ER localization. Collectively, these studies demonstrate a novel membrane-targeting domain that serves to recruit SKIP and PIPP to membrane ruffles.  相似文献

Induction of tissue factor expression in endothelial cells via ligation of CD40 probably figures prominently in the pathogenesis of prevalent inflammatory diseases, including atherosclerosis. However, the molecular mechanisms of tissue factor gene expression triggered by CD40 ligand (CD40L) in this cell type remain unknown. We demonstrate here that the tissue factor promoter region -278 bp to +121 bp contains the CD40L-responsive elements, consisting of activator protein 1 (AP-1)+/-, nuclear factor (NF) kappaB-, and Egr-1-binding sites. Mutations of either the AP-1- or NF-kappaB-binding sites markedly reduced the CD40L-dependent promoter activation. The AP-1 and NF-kappaB sites displayed constitutive and CD40L-enhanceable DNA binding activity, respectively. Of note, mutation of the Egr-1-binding sites, previously not associated with CD40 signaling, impaired activation of the tissue factor promoter. Accordingly, CD40L strongly induced Egr-1 protein expression and DNA binding activity to all three bindings sites. In contrast to CD40L, other established inducers of tissue factor in endothelial cells, interleukin-1beta or tumor necrosis factor alpha, did not increase the expression of Egr-1. In conclusion, induction of tissue factor gene expression in human endothelial cells by CD40L involves AP-1 and NF-kappaB as well as Egr-1, a pathway previously not implicated in CD40 signaling and distinct from that employed by certain other proinflammatory cytokines.  相似文献

Goatfish use a pair of large chin barbels to probe the sea bottom to detect buried prey. The barbels are studded with taste buds but little else is known about the neural organization of this system. We found that the taste buds of the barbel are innervated in a strict orthogonal fashion. The barbel is innervated by a main nerve trunk running in the core of the barbel. A longitudinal nerve bundle originates from the main trunk and, after running a short distance distally, divides into two circumferential nerve bundles (CNB) extending respectively, medially and laterally around the barbel. Approximately 15 CNBs innervate each 1 mm length of barbel. At each transverse level, the CNB innervates two clusters of taste buds, each containing 14 end-organs. The primary taste centre in the brain is similarly extraordinary. The sensory inputs from the barbel terminate in a derived dorsal facial lobe, which has a highly convoluted surface forming a multitude of tubercles. Electrophysiological mapping experiments show that the entire barbel is somatotopically represented in a recurved elongate tubular fashion within the dorsal facial lobe.  相似文献

A water-soluble glucan, AR-Glucan, from the roots of Angelica acutiloba was obtained homogeneous as determined by ultracentrifugal analysis, electrophoresis, and gel filtration. AR-Glucan was composed Of d-glucose, and its MW was estimated to be 13 500. Methylation analysis indicated that AR-Glucan contained 4-O- and 4,6-di-O-substituted glucosyl residues. 1H and 13C NMR data accorded with the results of methylation analysis, and the glycosidic linkages in AR-Glucan were shown to have the α-configuration. The results of β-amylase, α-amylase, and pullulanase treatments of AR-Glucan showed that it contained (1 → 4) linked α-d-glucosyl side chains of long chain length such as amylopectin. Thus, AR-Glucan is a (1 → 4) linked α-d-glucan to which are attached glucosyl side chains at O-6 of the glucosyl residues of the main chain.  相似文献
