Binding of foot-and-mouth disease virus (FMDV) to cells requires an arginine-glycine-aspartic acid (RGD) sequence in the capsid protein VP1. We have genetically engineered an FMDV in which these three amino acids have been deleted, producing a virus particle which is unable to bind to cells. Cattle vaccinated with these receptor binding site-deleted virions were protected from disease when challenged with a virulent virus, demonstrating that these RGD-deleted viruses could serve as the basis for foot-and-mouth disease vaccines safer than those currently in use. This strategy may prove useful in the development of vaccines for other viral diseases. 相似文献

The histology of early feeding lesions of the cattle tick B. microplus has been studied using 32P labelled larvae to standardize the duration of attachment. Critical studies were made on 3-h lesions in six separate experiments on different groups of British breed animals. Each group consisted of three animals--one previously unexposed to ticks, one of high resistance and one of low resistance. The degree of mast cell disruption, eosinophil concentration and degranulation, and the extent of epidermal vesiculation were all significantly greater at the site of attachment on highly resistant hosts. In previously unexposed animals there was no mobilization of eosinophils nor mast cell breakdown and no epidermal vesiculation. Possible immune mechanisms producing mast cell disruption and the infiltration and concentration of eosinophils are suggested, and the effect of eosinophil degranulation on larval attachment and feeding is discussed. 相似文献

We have tested the ability of a gentle gradient of neurite-promoting activity to orient the extension of embryonic growth cones. Gradients of neurite-promoting activity were made with biologically active, tritium-labeled laminin. The distributions of laminin bound to glass substrata were visualized by autoradiography and quantified with an image processing system. Embryonic chick sympathetic ganglia were explanted onto laminin gradients and cultured. No tendency for neurites to be oriented up-gradient was detected by examining the morphology of explants. Time-lapse studies of individual growth cones detected no up- or down-gradient bias in growth cone motility. These results suggest that growth cone orientation is relatively insensitive to a graded distribution of a naturally occurring neurite-promoting molecule. 相似文献

Molecular evolution of the period gene in Drosophila athabasca 总被引:1,自引:0,他引:1
We measured nucleotide variability within and between the three semispecies
of the Drosophila athabasca complex, at the period (per) gene by using a
polymerase chain reaction-based four-cutter restriction- enzyme analysis.
The levels of polymorphism varied considerably between the three
semispecies. Our results for per, combined with previous data for X-linked
allozymes, suggest that the X chromosome in the western- northern
semispecies is less variable than expected under an equilibrium-neutral
model. Both the pattern of divergence between the semispecies and a
cladistic clustering of per haplotypes support the previously hypothesized
grouping of eastern A and eastern B as the two most recently diverged
semispecies. A 21-bp in-frame segment in the region of per which shares
sequence similarity with the neuronal development gene single minded is
deleted in all eastern A and eastern B flies examined but is present in all
of the western-northern flies and all other published per sequences.
Despite these hints that there may be significant differences at the per
gene between the semispecies, especially the western-northern group versus
the two eastern groups, there is no compelling evidence that per is
involved in the mating song differences between the semispecies.
