Herpesvirus nucleocapsids escape from the nucleus in a process orchestrated by a highly conserved, viral nuclear egress complex. In human cytomegalovirus, the complex consists of two proteins, UL50 and UL53. We solved structures of versions of UL53 and the complex by X‐ray crystallography. The UL53 structures, determined at 1.93 and 3.0 Å resolution, contained unexpected features including a Bergerat fold resembling that found in certain nucleotide‐binding proteins, and a Cys3His zinc finger. Substitutions of zinc‐coordinating residues decreased UL50–UL53 co‐localization in transfected cells, and, when incorporated into the HCMV genome, ablated viral replication. The structure of the complex, determined at 2.47 Å resolution, revealed a mechanism of heterodimerization in which UL50 clamps onto helices of UL53 like a vise. Substitutions of particular residues on the interaction interface disrupted UL50–UL53 co‐localization in transfected cells and abolished virus production. The structures and the identification of contacts can be harnessed toward the rational design of novel and highly specific antiviral drugs and will aid in the detailed understanding of nuclear egress.  相似文献

Dientamoeba fragilis is a protozoan parasite emerging as a cause of diarrhoea and “irritable-bowel-like” gastrointestinal disease in humans with a propensity for establishing long-term, chronic infections in humans. Although Dientamoeba was discovered over a century ago its life cycle and mode of transmission is not known. No cyst stage has been described and no animal models are presently available for the study of this parasite. Here we describe the establishment of an animal model using laboratory rodents, the fulfilling of Koch’s postulates, and the discovery of a new cyst stage in the life cycle of D. fragilis. Our demonstration of long-term parasite carriage by rodents and prolonged shedding of cysts, together with elevated levels of calprotectin in the stool, confirms the capacity of this organism to cause disease and indicates dientamoebiasis should be considered in the differential diagnosis of gastrointestinal diseases such as Inflammatory Bowel Syndrome (IBS). Finally, we suggest that the cyst stage described here is the vehicle that mediates faecal–oral transmission of D. fragilis between hosts.  相似文献

A new flavanone, shisoflavanone A (1), and several flavonoids were purified from the ethyl acetate-soluble fraction of green perilla leaves (Perilla frutescens Britton var. crispa form viridis), and their structures were identified. Shisoflavanone A was elucidated as 8-hydroxy-6,7-dimethoxyflavanone based on its spectral data. Other constituents of the ethyl acetate-soluble fraction, i.e. 5,8-dihydroxy-7-methoxyflavanone (2), negletein (5,6-dihydroxy-7-methoxyflavone) (3), luteolin (4), apigenin (5), esculetin (6), and protocatechuic acid (7), were identified. This is the first time that constituents 2, 3, and 6 have been found in green perilla. Shisoflavanone A and the other constituents (except 7) significantly inhibited nitric oxide production in interleukin 1β-stimulated rat hepatocytes, which have been used to monitor the anti-inflammatory effects of herbal constituents. The present findings suggest that these constituents, including shisoflavanone A, may be involved in the anti-inflammatory effects of green perilla leaves.  相似文献

Manganese (Mn) and iron (Fe) concentrations were measured in coral skeletons (Porites spp.) collected from the Todoroki River on Ishigaki Island, Japan, to reconstruct the history of land use in the river catchment area. We prepared (1) five bulk samples to investigate the present spatial distribution and (2) micro-samples from two long cores to study the temporal variability of sediment loading from the Todoroki River. The existing state of the elements Mn and Fe in bulk coral skeleton samples was examined by a chemical cleaning experiment. The results of the experiment suggested that Fe was not incorporated into the crystal lattice of the coral skeleton but that Mn was incorporated, as previously reported. The bulk sample data, with and without chemical cleaning, indicated that the spatial distribution of both elements in corals collected along a sampling line from the river mouth toward the reef crest was complex and most likely reflected salinity changes and the amount of suspended particulate matter. The temporal variation of Mn and Fe, in particular the variation of baseline/background levels, mainly reflected the history of land development on Ishigaki Island. In addition, Mn showed clear seasonal variability that appeared to be controlled by a combination of temperature, primary productivity, and precipitation. The results of the present study suggest that Mn may be a useful proxy for river discharge or biological activity depending on local marine conditions, if the specific behavior of Mn at the coral growth site is known.  相似文献

Interest in belowground plant growth is increasing, especially in relation to arguments that shallow‐rooted cultivars are efficient at exploiting soil phosphorus while deep‐rooted ones will access water at depth. However, methods for assessing roots in large numbers of plants are diverse and direct comparisons of methods are rare. Three methods for measuring root growth traits were evaluated for utility in discriminating rice cultivars: soil‐filled rhizotrons, hydroponics and soil‐filled pots whose bottom was sealed with a non‐woven fabric (a potential method for assessing root penetration ability). A set of 38 rice genotypes including the OryzaSNP set of 20 cultivars, additional parents of mapping populations and products of marker‐assisted selection for root QTLs were assessed. A novel method of image analysis for assessing rooting angles from rhizotron photographs was employed. The non‐woven fabric was the easiest yet least discriminatory method, while the rhizotron was highly discriminatory and allowed the most traits to be measured but required more than three times the labour of the other methods. The hydroponics was both easy and discriminatory, allowed temporal measurements, but is most likely to suffer from artefacts. Image analysis of rhizotrons compared favourably to manual methods for discriminating between cultivars. Previous observations that cultivars from the indica subpopulation have shallower rooting angles than aus or japonica cultivars were confirmed in the rhizotrons, and indica and temperate japonicas had lower maximum root lengths in rhizotrons and hydroponics. It is concluded that rhizotrons are the preferred method for root screening, particularly since root angles can be assessed.  相似文献

The intracellular enzyme urea amidolyase (Dur1,2p) enables C. albicans to utilize urea as a sole nitrogen source. Because deletion of the DUR1,2 gene reduces survival of C. albicans co-cultured with a murine macrophage cell line, we investigated the role of Dur1,2p in pathogenesis using a mouse model of disseminated candidiasis. A dur1,2Δ/dur1,2Δ strain was significantly less virulent than the wild-type strain, showing significantly higher survival rate, better renal function, and decreased and less sustained fungal colonization in kidney and brain. Complementation of the mutant restored virulence. DUR1,2 deletion resulted in a milder host inflammatory reaction. Immunohistochemistry, flow cytometry, and magnetic resonance imaging showed decreased phagocytic infiltration into infected kidneys. Systemic cytokine levels of wild-type mice infected with the dur1,2 mutant showed a more balanced systemic pro-inflammatory cytokine response. Host gene expression and protein analysis in infected kidneys revealed parallel changes in the local immune response. Significant differences were observed in the kidney IL-1 inflammatory pathway, IL-15 signaling, MAP kinase signaling, and the alternative complement pathway. We conclude that Dur1,2p is important for kidney colonization during disseminated candidiasis and contributes to an unbalanced host inflammatory response and subsequent renal failure. Therefore, this Candida-specific enzyme may represent a useful drug target to protect the host from kidney damage associated with disseminated candidiasis.  相似文献

The configuration at the C-3 quaternary carbon atoms in two pairs (1 and 2, 3 and 4) of 3-C-hydroxymethyl, branched-chain, 1,2:4,6-diacetalated aldohexo-pyranoses have been determined from their 13 C-n.m.r. spectra. The stereochemical assignments were achieved by comparison of the spectra with those of the Z (13) and E isomers (14) of 4-tert-butyl-l-hydroxymethylcyclohexanol and with those of the corresponding diacetalated gluco- and allo-pyranoses (5, 6, 9, and 10). The spectra of 13 and 14 showed that an axial hydroxyl group shielded the α, β, and μ ring carbon atoms more than an axial hydroxymethyl group and that the carbon atom in the latter group was shielded relative to that in an equatorial hydroxymethyl group The spectra of 5, 6, 9, and 10 indicated the effect of an axial HO-3 on the shifts of the carbon atoms in the 1,2-O-alkylidene groups. The stereochemistry of an isomeric pair of 1,2:4,6-di-O-alkylidene-3-C-methyl-aldohexopyranoses (11 and 12) has also been determined.  相似文献
