Despite evidence that some individuals achieve both superiorreproductive performance and high survivorship, the factorsunderlying variation in individual quality are not well understood.The compensation and increased-intake hypotheses predict thatbasal metabolic rate (BMR) influences reproductive performance;if so, variation in BMR may be related to differences in individualquality. We evaluated whether BMR measured during the incubationperiod provides a proximate explanation for variation in individualquality by measuring the BMRs and reproductive performance ofLeach's storm-petrels (Oceanodroma leucorhoa) breeding on KentIsland, New Brunswick, Canada, during 2000 and 2001. We statisticallycontrolled for internal (body mass, breeding age, sex) and external(year, date, time of day) effects on BMR. We found that maleswith relatively low BMRs hatched their eggs earlier in the seasonand that their chicks' wing growth rates were faster comparedto males with relatively high BMRs. Conversely, BMR was notrelated to egg volume, hatching date, or chick growth rate forfemales or to lifetime (23 years) hatching success for eithersex. Thus, for males but not for females, our results supportthe compensation hypothesis. This hypothesis predicts that animalswith low BMRs will achieve better reproductive performance thananimals with high BMRs because they have lower self-maintenancecosts and therefore can apportion more energy to reproduction.These results provide evidence that intraspecific variationin reproductive performance is related to BMR and suggest thatBMR may influence individual quality in males.  相似文献

Chondrocytes embedded in agarose and subjected to dynamic deformational loading produce a functional matrix with time in culture, but there is usually a delay in the development of significant differences compared to free swelling. In this study, we hypothesized that the initial presence of a cell-associated matrix would expedite construct development in response to dynamic deformational loading. Seeded samples with enzymatically isolated chondrocytes and chondrons (the chondrocyte and its pericellular matrix) and examined the effects of seeding density and dynamic loading on the development of tissue properties. At 60 million/ml, dynamic loading significantly augmented the development of material properties in chondrocyte- and chondron-seeded constructs. Biochemical content and histological analysis indicated that the deposition of GAG, link protein and collagens are affected by the pre-existing cell-associated matrix of the chondron-seeded samples. The pericellular matrix associated with the chondrons did not expedite the development of material properties in response to deformational loading, disproving our hypothesis. The relative concentration and distribution of matrix proteins may play a major role in the disparate responses observed for the chondrocyte- and chondron-seeded cultures. In further support of these findings, culturing chondrocytes in agarose for two weeks prior to the application of deformational loading also did not exhibit expedited construct development.  相似文献

Age-specific reproductive success has been demonstrated in many species. Three hypotheses have been raised to explain this general phenomenon: the experience hypothesis based on age-specific reproductive experience, the effort hypothesis based on age-specific reproductive effort, and the selection hypothesis based on progressive disappearance of phenotypes due to variation in individual productivity and survival. We used data from a long-term study of Leach's storm-petrels (Oceanodroma leucorhoa) to present a single test of mutually exclusive predictions about the relationship between early breeding success and longevity. There should be no correlation between early breeding success and longevity under the experience hypothesis, a negative correlation under the effort hypothesis, and a positive correlation under the selection hypothesis. We found a significant (P < 0.0001) positive relationship between success in the first two breeding attempts and longevity in this population of long-lived seabirds, strongly suggesting that low-productivity parents were also less likely to survive early breeding. These data provide some of the strongest support to date for the selection hypothesis.  相似文献

In addition to information for current functions, the sequence of a gene includes potential information for the evolution of new functions. The wild-type ebgA (evolved beta-galactosidase) gene of Escherichia coli encodes a virtually inactive beta-galactosidase, but that gene has the potential to evolve sufficient activity to replace the lacZ gene for growth on the beta-galactoside sugars lactose and lactulose. Experimental evidence, which has suggested that the evolutionary potential of Ebg enzyme is limited o two specific amino acid replacements, is limited to examining the consequences of single base- substitutions. Thirteen beta-galactosidases homologous with the Ebg beta-galactosidase are widely dispersed, being found in gram-negative and gram-positive eubacteria and in a eukaryote. A comparison of Ebg beta-galactosidase with those 13 beta-galactosidases shows that Ebg is part of an ancient clade that diverged from the paralogous lacZ beta- galactosidase over 2 billion years ago. Ebg differs from other members of its clade at only 2 of the 15 active-site residues, and the two mutations required for full Ebg beta-galactosidase activity bring Ebg into conformity with the other members of its clade. We conclude that either these are the only acceptable amino acids at those positions, or all of the single-base-substitution replacements that must arise as intermediates on the way to other acceptable amino acids are so deleterious that they constitute a deep selective valley that has not been traversed in over 2 billion years. The evolutionary potential of Ebg is thus limited to those two replacements.   相似文献

The electrical penetration graphing or electropenetrography (EPG) technique is essential for understanding interactions of hemipteran insects with their host plants. Typically, 10–12.5 μm diameter gold wire is used as the tethering material in EPG studies. This wire was originally chosen based on suitability for aphids, but application of the EPG technique to other insects necessitates testing of alternative tethering materials that permit natural foraging and probing behavior. Whiteflies are one group for which EPG studies are increasing, with most researchers using 10 or 12.5 μm diameter gold wire even though these insects are smaller than aphids and very different in mobility. However, 2.5 μm diameter Wollaston process platinum wire has been used for a subset of EPG studies and seems to permit more natural movement and feeding behaviors. Here, we compared EPG variables derived from recordings of the sweet potato whitefly (Bemisia tabaci) tethered with 12.5 μm diameter gold wire or 2.5 μm diameter platinum wire. On a suitable host, gold-tethered whiteflies had reduced phloem phases, which are indicative of host plant acceptance, compared to platinum-tethered whiteflies. When we included a treatment known to reduce plant quality (methyl jasmonate application), platinum-tethered whiteflies exhibited expected reductions in EPG variables related to host acceptance, while gold-tethered whiteflies either had no response, or the opposite response. Our results indicate that tethering material strongly influences the outcome of EPG experiments, with important consequences for evaluations of host plant resistance, putative instances of plant virus manipulation, and feeding variables associated with virus transmission.  相似文献

The salient feature of anthropogenic climate change over the last century has been the rise in global mean temperature. However, global mean temperature is not used as an explanatory variable in studies of population‐level response to climate change, perhaps because the signal‐to‐noise ratio of this gross measure makes its effect difficult to detect in any but the longest of datasets. Using a population of Leach's storm‐petrels breeding in the Bay of Fundy, we tested whether local, regional, or global temperature measures are the best index of reproductive success in the face of climate change in species that travel widely between and within seasons. With a 56‐year dataset, we found that annual global mean temperature (AGMT) was the single most important predictor of hatching success, more so than regional sea surface temperatures (breeding season or winter) and local air temperatures at the nesting colony. Storm‐petrel reproductive success showed a quadratic response to rising temperatures, in that hatching success increased up to some critical temperature, and then declined when AGMT exceeded that temperature. The year at which AGMT began to consistently exceed that critical temperature was 1988. Importantly, in this population of known‐age individuals, the impact of changing climate was greatest on inexperienced breeders: reproductive success of inexperienced birds increased more rapidly as temperatures rose and declined more rapidly after the tipping point than did reproductive success of experienced individuals. The generality of our finding that AGMT is the best predictor of reproductive success in this system may hinge on two things. First, an integrative global measure may be best for species in which individuals move across an enormous spatial range, especially within seasons. Second, the length of our dataset and our capacity to account for individual‐ and age‐based variation in reproductive success increase our ability to detect a noisy signal.  相似文献

The purpose of this study was to investigate the association between physical fitness and mild cognitive impairment (MCI) in elderly Koreans. This was a cross-sectional study that involved 134 men and 299 women aged 65 to 88 years. Six senior fitness tests were used as independent variables: 30 s chair stand for lower body strength, arm curl for upper body strength, chair-sit-and-reach for lower body flexibility, back scratch for upper body flexibility, 8-ft up-and-go for agility/dynamic balance, and 2-min walk for aerobic endurance. Global cognitive function was assessed using the Korean version of the Mini-Mental State Examination (MMSE). Potential covariates such as age, education levels, blood lipids, and insulin resistance (IR) markers were also assessed. Compared to individuals without MMSE-based MCI, individuals with MMSE-based MCI had poor physical fitness based on the senior fitness test (SFT). There were significant positive trends observed for education level (p=0.001) and MMSE score (p<0.001) across incremental levels of physical fitness in this study population. Individuals with moderate (OR=0.341, p=0.006) and high (OR=0.271, p=0.007) physical fitness based on a composite score of the SFT measures were less likely to have MMSE-based MCI than individuals with low physical fitness (referent, OR=1). The strength of the association between moderate (OR=0.377, p=0.038) or high (OR=0.282, p=0.050) physical fitness and MMSE-based MCI was somewhat attenuated but remained statistically significant even after adjustment for the measured compounding factors. We found that poor physical fitness was independently associated with MMSE-based MCI in elderly Koreans.  相似文献

Due to its avascular nature, articular cartilage exhibits a very limited capacity to regenerate and to repair. Although much of the tissue-engineered cartilage in existence has been successful in mimicking the morphological and biochemical appearance of hyaline cartilage, it is generally mechanically inferior to the natural tissue. In this study, we tested the hypothesis that the application of dynamic deformational loading at physiological strain levels enhances chondrocyte matrix elaboration in cell-seeded agarose scaffolds to produce a more functional engineered tissue construct than in free swelling controls. A custom-designed bioreactor was used to load cell-seeded agarose disks dynamically in unconfined compression with a peak-to-peak compressive strain amplitude of 10 percent, at a frequency of 1 Hz, 3 x (1 hour on, 1 hour off)/day, 5 days/week for 4 weeks. Results demonstrated that dynamically loaded disks yielded a sixfold increase in the equilibrium aggregate modulus over free swelling controls after 28 days of loading (100 +/- 16 kPa versus 15 +/- 8 kPa, p < 0.0001). This represented a 21-fold increase over the equilibrium modulus of day 0 (4.8 +/- 2.3 kPa). Sulfated glycosaminoglycan content and hydroxyproline content was also found to be greater in dynamically loaded disks compared to free swelling controls at day 21 (p < 0.0001 and p = 0.002, respectively).  相似文献

Using a custom galvanotaxis chamber and time-lapse digital video microscopy, we report the novel observation that cultured chondrocytes exhibit cathodal migration when subjected to applied direct current (DC) electric fields as low as 0.8 V/cm. The response was dose-dependent for field strengths greater than 4 V/cm. Cell migration appeared to be an active process with extension of cytoplasmic processes in the direction of movement. In some cells, field application for greater than an hour induced elongation of initially round cells accompanied by perpendicular alignment of the long axis with respect to the applied field. Antagonists of the inositol phospholipid pathway, U-73122 and neomycin, were able to inhibit cathodal migration. Cell migration toward the cathode did not require the presence of serum during field application. However, the directed velocity was nearly threefold greater in studies performed with serum. Studies performed at physiologic temperatures (approximately 37 degrees C) revealed a twofold enhancement in migration speed compared to similar studies at room temperature (approximately 25 degrees C). Findings from the present study may help to elucidate basic mechanisms that mediate chondrocyte migration and substrate attachment. Since chondrocyte migration has been implicated in cartilage healing, the ability to direct chondrocyte movement has the potential to impact strategies for addressing cartilage healing/repair and for development of cartilage substitutes.  相似文献
